Activating transcription factor 5(ATF5) is a member of the activating transcription factor/cA MP response element binding protein(ATF/CREB) family, and is highly expressed in liver and adipose tissue. Previous reports have shown that ATF5 promoted 3T3-L1 preadipocytes differentiation. In this study, we found that ATF5 was highly expressed in mature adipocytes, suggesting a potential role of ATF5 in mature adipocytes, which has not been reported previously. To understand the function of ATF5 in mature adipocytes, we knocked down the expression of ATF5 in 3T3-L1 mature adipocytes and observed decreased lipid droplets. Consistent with the in vitro experiment, the knockdown of ATF5 in white adipose tissue led to less adipose tissue and smaller adipocytes size. Further research revealed that the inhibition of ATF5 diminished the adipocytes size via the inhibition of fatty acid synthetase, stearyl coenzyme A desaturation enzyme 1, and the induction of carnitine palmitoyl transferase 1, one key enzyme of lipid metabolism. In addition, ATF5 knockdown in inguinal white adipose tissue improved whole body insulin *** work provides a new understanding of ATF5 function in mature adipocytes and a potential therapeutic target of diabetes.

As a protein expression vector, the baculovirus demonstrates many advantages over other vectors. With the development of biotechnology, baculoviral vectors have been genetically modified to facilitate high level expression of heterologous proteins in both insect and mammalian cells. These modifications include utilization of different promoters and signal peptides, deletion or replacement of viral genes for increasing protein secretion, integration of polycistronic expression cassette for producing protein complexes, and baculovirus pseudotyping, promoter accommodation or surface display for enhancing mammalian cell targeting gene delivery. This review summarizes the development and the current state of art of the baculovirus expression system. Further development of baculovirus expression systems will make them even more feasible and accessible for advanced applications.

Adipocytes and immune cells are vital for the development of adipose ***-pokines secreted by adipocytes regulate adipogenesis and body *** is one of the ***,the function and mechanism of chemerin in adipose tissue are not fully *** with wild type(WT)mice,Rarres2^(-/-)mice gained weight and significantly increased fat distribution in subcutaneous adipose tissue(SAT),rather than visceral adipose tissue(VAT)on high fat diet(HFD).PPARg and C/EBPa,the master regulators of adipogenesis,were up-regulated in SAT and down-regulated in VAT in Rarres2^(-/-)mice comparing with WT *** of chemerin deficiency or not,the ratio of adipocyte-progenitors to total cells and the differentiation capacity of adipocyte-progenitors were similar in SAT and VAT,but macrophage infiltration in VAT was more severe than in SAT in Rarres2^(-/-)***,CD45þimmune cells supernatant from Rarres2^(-/-)SAT pro-moted the differentiation of adipocyte-progenitors and 3T3-L1 *** array assay of CD45þimmune cells supernatant revealed that metalloproteinase inhibitor 1(TIMP1),an in-hibitor of adipogenesis,was reduced in Rarres2^(-/-)SAT,but increased in Rarres2^(-/-)*** we specifically knocked down chemerin in SAT,TIMP1 was down-regulated and adipogenesis was promoted with reducing infiltration of *** present study demonstrates that the effects of chemerin on adipose tissue is depot different,and specific knock down chemerin in SAT promote adipogenesis and improve glucose tolerance test(GTT)and insulin tolerance test(ITT).This suggests a potential therapeutic target for chemerin in the treatment of obesity related metabolic disorder.

DNA methylation is a crucial regulatory mechanism in many biological ***,limited studies have dissected the contribution of DNA methylation to sexual differentiation in dioecious *** this study,we investigated the variances in methylation and transcriptional patterns of male and female flowers of garden *** with male flowers,female flowers at the same stages showed higher levels of DNA *** male and female flowers gained DNA methylation globally from the premeiotic to meiotic *** analysis revealed that the increased DNA methylation was largely due to increased CHH *** analysis of differentially expressed genes and differentially methylated regions suggested that DNA methylation might not have contributed to the expression variation of the sex-determining genes SOFF and TDF1 but probably played important roles in sexual differentiation and flower development of garden *** upregulated genes AoMS1,AoLAP3,AoAMS,and AoLAP5 with varied methylated CHH regions might have been involved in sexual differentiation and flower development of garden *** hormone signaling genes and transcription factor genes also participated in sexual differentiation and flower development with potential epigenetic *** addition,the CG and CHG methylation levels in the Y chromosome were notably higher than those in the X chromosome,implying that DNA methylation might have been involved in Y chromosome *** data provide insights into the epigenetic modification of sexual differentiation and flower development and improve our understanding of sex chromosome evolution in garden asparagus.

8-Sphingolipid desaturase is the key enzyme that catalyses desaturation at the C8 position of the long-chain base of sphingolipids in higher plants. There have been no previous studies on the genes encoding AS-sphingolipid desaturases in Brassica rapa. In this study, four genes encoding AS-sphingolipid desaturases from B. rapa were isolated and characterised. Phylogenetic analyses indicated that these genes could be divided into two groups： BrD8A, BrD8C and BrD8D in group I, and BrD8B in group II. The two groups of genes diverged before the separation of Arabidopsis and Brassica. Though the four genes shared a high sequence similarity, and their coding desaturases all located in endoplasmic reticulum, they exhibited distinct expression patterns. Heterologous expression in Saccharomyces cerevisiae revealed that BrD8A/B/C/D were functionally diverse AS-sphingolipid desaturases that catalyse different ratios of the two products 8（Z）- and 8（E）-C18-phytosphingenine. The aluminium tolerance of transgenic yeasts expressing BrD8A/B/C/D was enhanced compared with that of control cells. Expression of BrD8A in Arabidopsis changed the ratio of 8（Z）：8（E）-C 18-phytosphingenine in transgenic plants. The information reported here provides new insights into the biochemical functional diversity and evolutionary relationship of AS-sphingolipid desaturase in plants and lays a foundation for further investigation of the mechanism of 8（Z）- and 8（E）-C18- phytosphingenine biosynthesis.

Asparagus setaceus is a popular ornamental plant cultivated in tropical and subtropical regions ***,we constructed a chromosome-scale reference genome of *** to facilitate the investigation of its genome characteristics and *** a combination of Nanopore long reads,Illumina short reads,10×Genomics linked reads,and Hi-C data,we generated a high-quality genome assembly of *** covering 710.15 Mb,accounting for 98.63%of the estimated genome size.A total of 96.85%of the sequences were anchored to ten superscaffolds corresponding to the ten *** genome of *** was predicted to contain 28,410 genes,25,649(90.28%)of which were functionally annotated.A total of 65.59%of the genome was occupied by repetitive sequences,among which long terminal repeats were predominant(42.51%of the whole genome).Evolutionary analysis revealed an estimated divergence time of *** from its close relative *** of~9.66 million years ago,and *** underwent two rounds of whole-genome *** addition,762 specific gene families,96 positively selected genes,and 76 resistance(R)genes were detected and functionally predicted in *** findings provide new knowledge about the characteristics and evolution of the *** genome,and will facilitate comparative genetic and genomic research on the genus Asparagus.

Objective:To study the effect of dezocine pretreatment on the emergence agitation and related internal environment changes in children with general anesthesia ***: A total of 200 children who underwent general anesthesia surgery in the Central Hospital of Xiaogan between January 2015 and December 2017 were enrolled in the study and divided into two groups by random number table method, dezocine group received routine general anesthesia and intravenous injection of dezocine 30 min before the end of surgery, and control group received routine general anesthesia and intravenous injection of saline 30 min before the end of surgery. The CHEPOS score of emergence agitation was performed during anesthesia recovery;the levels of inflammatory stress mediators in serum as well as the expression of stress signal molecules and inflammatory signal molecules in peripheral blood were measured before anesthesia induction and during anesthesia ***: The CHEPOS score of emergence agitation of dezocine group was lower than that of control group;compared with those of same group before anesthesia induction, serum ACTH, Cor, INS, ICAM1 and TNF-α levels as well as peripheral blood CHOP, GRP78, JNK, c-jun, CD14 and SR expression intensity of both groups of patients were significantly higher whereas IRS-1, IRS-2 and PKB expression intensity were significantly lower during anesthesia recovery, and serum ACTH, Cor, INS, ICAM1 and TNF-α levels as well as peripheral blood CHOP, GRP78, JNK, c-jun, CD14 and SR expression intensity of dezocine group during anesthesia recovery were significantly lower than those of control group whereas IRS-1, IRS-2 and PKB expression intensity were significantly higher than those of control ***: Dezocine pretreatment has improving effect on the emergence agitation and related internal environment changes in children with general anesthesia surgery.

Photovoltaic metal halide perovskite solar cells(PSCs) convert light to electricity more efficiently than crystalline silicon cells, and the cost of materials used to make them is lower than that of silicon *** efficiency is not a core issue affecting the application of perovskite solar cells in special *** present, stability is the major technical encounters that hinders its further commercial development. Microstrain in PSCs is currently a significant factor responsible for the device's instability. Strain-induced ion migration is widely believed to accelerate perovskite degradation even when external stimuli are ***, it is imperative to study strain to enhance the stability of PSCs. This paper reviews recent developments to understand strain's origin and effect mechanisms on performance of PSCs, including ion migration,failure behavior, defect formation, and its effect on photoelectric properties, stability, and ***, several well-known strain management strategies are systematically introduced based on the strain effect mechanism and strain engineering on the film, providing more clues for further preparation with increased stability. The manipulation of external physical strain applied from films to entire devices has been extensively studied. Furthermore, recommendations for future research directions and chemical approaches have been provided. It is emphasized that strain engineering plays a crucial role in improving the efficiency and longevity of PSCs. Tensile strain causes rapid degradation, while moderate compressive strain and external strain control could improve properties and stability. Efforts should focus on controlling compressive strain to mitigate residual tensile strain and introducing it in a controlled manner. Future research endeavors may focus on exploring these pathways to improve the efficiency and lifespan of PSCs.
