AIM: To investigate the expression of markers that are correlated with the prognosis of colorectal cancer (CRC) patients. METHODS: One hundred and fifty-six CRC patientswere followed up for more than 3 years after radical surgery. Immunohistochemical (IHC) analysis was performed to detect the expression of 14 pathway-related markers (p53, APC, p21ras, E-cadherin, endothelin-B receptor, Shp2, ADCY-2, SPARCL1, neuroligin1, hsp27, mmp-9, MAPK, MSH2 and rho) in specimens from these patients. Bioinformatics analysis involving a Support Vector Machine (SVM) was used to determine the best prognostic model from combinations of these markers. RESULTS: Seven markers (SPARCL1, Shp2, MSH2, E-cadherin, p53, ADCY-2 and MAPK) were significantly related to the prognosis and clinical pathological features of the CRC patients (P < 0.05). Prognostic models were established through SVM from combinations of these 7 markers and proved able to differentiate patients with dissimilar survival, especially in stage Ⅱ/Ⅲ patients. According to the best prognostic model, the p53/SPARCL1 model, patients having high p53 and low SPARCL1 expression had about 50% lower 3-year survival than others (P < 0.001). CONCLUSION: SPARCL1, Shp2, MSH2, E-cadherin, p53, ADCY-2 and MAPK are potential prognostic markers in CRC. A p53/SPARCL1 bioinformatics model may be used as a supplement to tumor-nodes-metastasis staging.

BACKGROUND: The role of programmed death 1 (PD-1) and programmed death ligand 1 (PD-L1) in persistent HBV infection is controversial. Increasing PD-1 and PD-L1 expression has been found in hepatitis B patients during immune clearance phase, but not in HBV-tolerant patients. We investigated PD-1 and PD-L1 expression and inflammation in chronic hepatitis B. METHODS: Twenty patients with chronic hepatitis B participated in this study. Fifteen patients were in the immune clearance phase, and 5 were in the immune inactive phase. Circulating HBV-specific T cells were analyzed by flow cytometric detection of major histocompatibility complex (MHC) class I peptide complexes, known as pentamers. Intra-hepatic PD-1 and PD-L1 expressions were analyzed by immunostaining. RESULTS: The frequency of pentamers, including core 18-27 (1.88%±0.36%), env 335-343 (1.85%±0.37%), and pol 575-583 (1.56%±0.29%) was 8.30-, 7.71- and 8.48-fold greater during immune clearance phase than those during the immune inactive phase. In addition, more than 70% of circulating pentamers were PD-1 positive. During immune clearance phase, the numbers of intra-hepatic PD-1 and PD-L1 positive cells were 108±23/HPF and 97±20/HPF respectively, in contrast, there was a paucity of PD-1 and PD-L1 positive cells in the immune inactive phase. The numbers of intra-hepatic PD-1 and PD-L1 positive cells were positively correlated with serum alanine aminotransferase and the number of intra-hepatic CD8 + T cells. Immunofluorescence showed that almost all of the intra- hepatic CD8 + T cells were PD-1 and CCR6 positive. These cells aggregated around macrophage inflammatory protein-3 alpha (MIP3α) positive cells and mixed with PD-L1 positive ***: PD-1 and PD-L1 expressions were significantly correlated with inflammation. BACKGROUND: Pancreatic cancer is a devastating disease with abnormal genetic changes. The pituitary tumor-derived transforming gene (PTTG) is considered to be implicated in the tumorigenesis of cancers when the gene is epigenetically transformed. In this study, we investigated the relationships between aberrant expression and epigenetic changes of the PTTG1 gene in pancreatic cancer. METHODS: We chose 4 cell lines (PANC-1, Colo357, T3M-4 and PancTu I) and pancreatic ductal adenocarcinoma (PDAC) tissues. After using restriction isoschizomer endonucleases (Msp I /Hpa II) to digest the DNA sequence (5'-CCGG-3'), we performed PCR reaction to amplify the product. And RT-PCR was applied to determine the gene expression. RESULTS: The mRNA expression of the PTTG1 gene was higher in pancreatic tumor than in normal tissue. The gene was also expressed in the 4 PDAC cell lines. The methylation states of the upstream regions of the PTTG1 gene were almost identical in normal, tumor pancreatic tissues and the 4 PDAC cell lines. Some (5'-CCGG-3') areas in the upstream region of PTTG1 were methylated, while some others were unmethylated. CONCLUSIONS: The oncogene PTTG1 was overexpressed in pancreatic tumor tissues and verified by RT-PCR detection. The methylation status of DNA in promoter areas was involved in the gene expression with the help of other factors in pancreatic cancer.

The regulatory mechanism of soil cadmium （Cd） on antioxidases in sorghum was studied using 3 sorghum species viz., sweet sorghum [Sorghum bicolor （L.）] Moench. cv. Hunnigreen, sorghum hybrid sudangrass （Sorghum bicolor × S. sudanense, cv. Everlush） and sudangrass [Sorghum sudanense （Piper） Stapf cv. Xinjiang]. The results showed that low concentrations of Cd^2＋（410 mg kg-0 induced the activities of 4 antioxidases in leaves of the 3 species of sorghum, but high Cd^2＋ concentrations （≥ 50 mg kg-0 significantly reduced the activities of all the four antioxidases. The glutathione （GSH） contents in leaves of the 3 species of sorghum correlated positively with the Cd^2＋ concentrations. The maximum antioxidase activities were observed at the elongation stage of sorghum during the whole growth period.

With the development of attosecond science, tunneling time can now be measured experimentally with the attoclock technique. However, there are many different theoretical definitions of tunneling time and no consensus has been ***, we bridge the relationship between different definitions of tunneling time based on a quantum travel time in onedimensional rectangular barrier tunneling problem. We find that the real quantum travel time t_(Re) is equal to the Bohmian time t_(Bohmian), which is related to the resonance lifetime of a bound state. The total quantum travel time τt can perfectly retrieve the transversal time t_x and the Büttiker–Landauer time τ_(BL) in two opposite limits, regardless of the particle energy.

AIM： To investigate the effects of polo-like kinase-1 （PLK1） antisense phosphorothioate oligodeoxynucleotide （ASODN） on apoptosis and cell cycle of human colon cancer cell line SW480. METHODS： After SW480 colon cancer cells were transfected with PLK1 ASODN, Northern and Western blot analyses were used to examine PLK1 gene expression in cancer cells. We studied apoptosis using terminal uridine deoxynucleotidyl nick end labeling. Apoptosis and cell cycle of SW480 cells were examined by fluorescence-activated cell sorter scan. RESULTS： The levels of PLK1 mRNA and protein were greatly inhibited by PLK1 ASODN in SW480 cancer cells transfected with PLK1 ASODN. Apoptosis index （AI） induced PLK1 ASODN in a time- and dose-dependent manner. Results from FLM showed that sub-2N DNA content of transfected cancer cells was significantly increased and arrested at G2/M compared with control groups. CONCLUSION： PLK1 ASODN can induce apoptosis of human colon cancer cell line SW480.

The generalized estimating equations(GEE) approach is perhaps one of the most widely used methods for longitudinal data analysis. While the GEE method guarantees the consistency of its estimators under working correlation structure misspecification, the corresponding efficiency can be severely affected. In this paper, we propose a new two-step estimation method in which the correlation matrix is assumed to be a linear combination of some known working matrices. Asymptotic properties of the new estimators are *** studies are conducted to examine the performance of the proposed estimators. We illustrate the methodology with an epileptic data set.

A facies and palaeogeography-based approach for analysis of petroleum systems in United Arab Emirates

The United Arab Emirates(UAE) is the 8 th largest oil producing country and is rich in oil and gas resources. By the end of 2015, 68 oil and 23 gas fields had been discovered. The initial proved and probable(2 P) oil, gas and condensate reserves amount to 81,135.9 MMb(million barrels), 192.09 Tcf(trillion cubic feet),and 6496.58 MMb respectively, which are mostly reservoired in the Jurassic and Cretaceous carbonates. With the latest field data, this study attempts to document the salient features of petroleum systems in UAE. Based on depositional facies of source rock intervals, pods of source rocks were delineated. On the basis of an oiland gas-source correlation, five known petroleum systems were identified and they are Lower Silurian-Upper Permian Khuff gas, northeast foreland Upper Jurassic-Lower Cretaceous gas, Upper Jurassic-Jurassic petroleum, Upper Jurassic/Lower Cretaceous-Lower Cretaceous composite petroleum, and Middle Cretaceous-Middle to Upper Cretaceous/Cenozoic petroleum systems. Of them, the Upper Jurassic/Lower Cretaceous-Lower Cretaceous composite petroleum system contains 73.2% of the total 2 P reserves and thus it is the focus of this study. The Upper Jurassic and Lower Cretaceous source rocks consist of argillaceous limestone, mudstone and shale, which were deposited as intrashelf basin facies. The distribution of oil and gas in this system is controlled by the source kitchens and the regional evaporite seal.
