Preliminary delivery efficiency prediction of nanotherapeutics into crucial cell populations in bone marrow niche

Several crucial stromal cell populations regulate hematopoiesis and malignant diseases in bone marrow *** regulation of these cell types can remodel niches and develop new *** nanocarriers have been developed to transport drugs into the bone marrow ***,the delivery efficiency of these nanotherapeutics into crucial niche cells is still unknown,and there is no method available for predicting delivery efficiency in these cell ***,we constructed a three-dimensional bone marrow niche composed of three crucial cell populations:endothelial cells(ECs),mesenchymal stromal cells(MSCs),and osteoblasts(OBs).Mimetic niches were used to detect the cellular uptake of three typical drug nanocarriers into ECs/MSCs/OBs in *** than 5%of nanocarriers were taken up by three stromal cell types,and most of themwere located in the extracellular *** efficiency in sinusoidal ECs,arteriole ECs,MSCs,and OBs in vivo was *** correlation analysis showed that the cellular uptake of three nanocarriers in crucial cell types in vitro is positively linear correlated with its delivery efficiency in *** delivery efficiency into MSCs was remarkably higher than that into ECs and OBs,no matterwhat kind of *** overall efficiency into sinusoidal ECswas greatly lower than that into arteriole *** nanocarriers were hard to be delivered into OBs(<1%).Our findings revealed that cell tropisms of nanocarriers with different compositions and ligand attachments in vivo could be predicted via detecting their cellular uptake in bone marrow niches in *** study provided the methodology for niche-directed nanotherapeutics development.

Objectives:Cisplatin(CDDP)is a widely used and effective basic chemotherapeutic drug for the treatment of a variety of tumors,including ovarian ***,adverse side effects and acquired drug resistance are observed in the clinical application of *** a mode of administration that can alleviate side effects and reduce drug resistance has become a promising strategy to solve this ***:In this study,3 D printing technology was used to prepare a CDDP-poly(lactic-co-glycolic acid)(CDDP-PLGA)polymer compound stent,and its physicochemical properties and cytotoxicity were evaluated both in vitro and in ***:The CDDP-PLGA stent had a significant effect on cell proliferation and apoptosis and clearly decreased the size of subcutaneous tumors in nude mice,whereas the systemic side effects were mild compared with those of intraperitoneal CDDP *** with the control group,CDDP-PLGA significantly increased the mRNA and protein levels of p-glycoprotein(P<0.01;P<0.01)and decreased vascular endothelial growth factor mRNA(P<0.05)and protein levels(P<0.01),however,CDDP-PLGA significantly decreased the mR NA and protein levels of p-glycoprotein(P<0.01;P<0.01)and vascular endothelial growth factor(P<0.01;P<0.01),which are associated with chemoresistance,in subcutaneous tumor *** assay results revealed that,in the CDDP-PLGA group,the staining of the proliferation-related genes Ki67 and PCNA were lightly,and the apoptosis-related gene caspase-3 stained ***:PLGA biomaterials loaded with CDDP,as compared with the same amount of free CDDP,showed good efficacy in terms of cytotoxicity,as evidenced by changes in *** local CDDP release can decrease the systemic side effects of this drug and the occurrence of drug resistance and angiogenesis,and improve the therapeutic *** new approach may be an effective strategy for the local treatment of epithelial ovarian cancer.

A method was used to fabricate a fully inkjet-printed gas sensor matrix on photographic *** electrode matrix comprising 36 interdigital electrodes in a high-density layout that is easy to integrate has been fabricated using a combination of insulating ink and commercial silver ***-imprinted polymer(MIP)inks were then made using a simple solution mixing method,and these inks were printed together with carbon black ink on the electrode matrix to complete production of the ***,experimental dynamic sensing of volatile organic compounds verifies that for detection of gases corresponding to the MIP template molecules,the MIP layer offers improvements in both sensitivity and selectivity when compared with non-imprinted polymer *** matrix can produce a response of more than 20%to 3 ppm propenoic acid gas through adjustment of the printing times for the carbon black layer and the MIP layer.

With the development of computational power and numerical algorithms,computational fluid dynamics(CFD) has become an important strategy for the design of aircraft,which significantly reduces the reliance on wind-tunnel and flight *** this paper,we conducted a numerical investigation on the flow past a full commercial aircraft at Mach number 0.2 and 14 degrees angle of attack by means of Reynolds-averaged Navier-Stokes(RANS),detached-eddy simulation(DES) and our newly developed constrained large-eddy simulation(CLES).The objective of this paper is to study the capability of these models in simulating turbulent *** our knowledge,this is the first large-eddy simulation method for full commercial aircraft *** results show that the CLES can predict the mean statistical quantities well,qualitatively consistent with traditional methods,and can capture more small-scale structures near the surface of the aircraft with massive *** study demonstrates that CLES is a promising alternative for simulating real engineering turbulent flows.

It’s important for mobile operators to recommend new services. Traditional method is sending advertising messages to all mobile users. But most of users who are not interested in these services treat the messages as Spam. This paper presents a method to find potential customers who are likely to accept the services. This method searchs the maximum frequent itemsets which indicate potential customers’ features from a large data set of users’ information, then find potential customers from those maximum frequent itemsets by using a bayesian network classifier. Experimental results demonstrate this method can select users with higher accuracy.

Nursing Care of 13 Cases with Acute Retinal Necrosis Associated with HIV/AIDS during Day Surgery

Objective: Summarizing the nursing points from the treatment of the 13 cases of AIDS-related acute retinal necrosis syndrome (ARNS). Methods: Retrospective analysis of clinical nursing process of 13 cases (15 eyes) of AIDS-related ARNS patient treated in our hospital from January to December 2019. Results: The retina of all the patients were flat after operation. Postoperative visual acuity: bilateral visual acuity 0.1 in 11 eyes, 0.01 - 0.1 in 3 eyes, >0.1 in 1 eye. No adverse events and nursing safety events occurred. Patients recovered well after operation during the follow-up after discharge. Conclusion: The focus of nursing care for patients with HIV/AIDS combined with acute retinal necrosis during daytime surgery is multifaceted, apart from the daily, perioperative nursing in day-care unit, continuous nursing, psychological counseling, disinfection and isolation should be paid attention to. At the same time, with the help of promotion and application of information education means, the treatment compliance of patients can be improved, which plays an essential role in the advance of the treatment effect.

Simultaneous measurement of strain and temperature using Fabry–Pérot interferometry and antiresonant mechanism in a hollow-core fiber

A fiber-optic sensor for the simultaneous measurement of strain and temperature is proposed and experimentally demonstrated based on Fabry–Pérot(FP) interference and the antiresonance(AR) mechanism. The sensor was implemented using a single-mode fiber(SMF)–hollow-core fiber–SMF structure. A temperature sensitivity of 21.11 pm/℃ was achieved by tracing the troughs of the envelope caused by the AR mechanism, and a strain sensitivity of 2 pm/με was achieved by detecting the fine fringes caused by the FP cavity. The results indicate that the dual-parameter sensor is stable and reliable.
