Prolonged use of bedaquiline in two patients with pulmonary extensively drug-resistant tuberculosis: Two case reports

BACKGROUND Bedaquiline is among the prioritized drugs recommended by the World Health Organization for the treatment of extensively drug-resistant tuberculosis(XDRTB).Many patients have not achieved better clinical improvement after bedaquiline is stopped at 24 ***,there is no recommendation or guideline on bedaquiline administration beyond 24 wk,which is an important consideration when balancing the benefit of prognosis for XDR-TB against the uncertain safety concerning the newer *** SUMMARY This paper reported 2 patients with XDR-TB(a female of 58 years of age and a female of 18 years of age)who received bedaquiline for 36 wk,as local experience to be *** 2 cases had negative cultures after 24 wk of treatment,but lung imaging was still *** discussion among experts,the consensus was made to bedaquiline prolongation by another 12 *** 36-wk prolonged use of bedaquiline in both cases achieved a favorable response without increasing the risk of cardiac events or new safety *** Longer regimen,including 36-wk bedaquiline treatment,might be an option for patients with *** studies are needed to explore the effectiveness and safety of prolonged use of bedaquiline for 36 wk vs standard 24 wk in the treatment of multidrug-resistant/XDR-TB or to investigate further the biomarkers and criteria indicative for extension of bedaquline to facilitate clinical use of thisnovel drug.

Monogamy and polygamy for the generalized W-class states using unified-(q,s)entropy

We describe the entanglement distribution and restricted shareability of the multipartite generalized W-class states and their reduced density matrix under arbitrary partitions by using monogamy and polygamy relation based on the unified-(q,s)***,we provide an analytical formula of unified-(q,s)entanglement(UE)and an analytical lower bound of unified-(q,s)entanglement of assistance(UEo A)for a reduced density matrix of a generalized W-class ***,we use these two analytical formulas to derive the monogamy and polygamy inequalities for a reduced density matrix of a qudit generalized W-class(GW)*** establish two partition-dependent residual entanglements based on the new monogamy relation,which is helpful to obtain a comprehensive analysis of entanglement dynamics of generalized W-class ***,we investigate tighter monogamy and polygamy relations based on the power ofαth(α≥0)for UE andβth(β≥0)for UEo A,*** results show that the entanglement distribution characteristics of generalized W-class states satisfying stronger constraints can be described more accurately.

Supervised learning often requires a large number of labeled examples,which has become a critical bottleneck in the case that manual annotating the class labels is *** mitigate this issue,a new framework called pairwise comparison(Pcomp)classification is proposed to allow training examples only weakly annotated with pairwise comparison,i.e.,which one of two examples is more likely to be *** previous study solves Pcomp problems by minimizing the classification error,which may lead to less robust model due to its sensitivity to class *** this paper,we propose a robust learning framework for Pcomp data along with a pairwise surrogate loss called *** provides an unbiased estimator to equivalently maximize AUC without accessing the precise class ***,we prove the consistency with respect to AUC and further provide the estimation error bound for the proposed *** studies on multiple datasets validate the effectiveness of the proposed method.

Anti-explosion performance and dynamic response of an innovative multi-layer composite explosion containment vessel

An innovative multi-layer composite explosion containment vessel(CECV)utilizing a sliding steel platealuminum honeycomb-fiber cloth sandwich is put forward to improve the anti-explosion capacity of a conventional single-layer explosion containment vessel(SECV).Firstly,a series of experiments and finite element(FE)simulations of internal explosions are implemented to understand the basic anti-explosion characteristics of a SECV and the rationality of the computational models and methods is verified by the comparison between the experimental results and simulation *** on this,the CECV is designed in detail and a variety of FE simulations are carried out to investigate effects of the sandwich structure,the explosive quantity and the laying mode of the fiber cloth on anti-explosion performance and dynamic response of the CECV under internal *** results indicate that the end cover is the critical position for both the SECV and *** maximum pressure of the explosion shock wave and the maximum strain of the CECV can be extremely declined compared to those of the *** a result,the explosive quantity the CECV can sustain is up to 20 times of that the SECV can ***,as the explosive quantity increases,the internal pressure of the CECV keeps growing and the plastic deformation and failure of the sandwich structure become more and more severe,yielding plastic strain of the CECV in addition to elastic *** results also reveal that the laying angles of the fiber cloth's five layers have an impact on the anti-explosion performance of the *** example,the CECV with fiber cloth layered in 0°/45°/90°/45°/0°mode has the optimal anti-capacity,compared to 0°/0°/0°/0°/0°and 0°/30°/60°/30°/0°***,owing to remarkable anti-explosion capacity,this CECV can be regarded as a promising candidate for explosion resistance.

Optimal Coordination of Transportable Power Sources and Repair Crews for Service Restoration of Distribution Networks Considering Uncertainty of Traffic Congestion

This paper proposes a new method for service restoration of distribution network with the support of transportable power sources(TPSs)and repair crews(RCs).Firstly,a coupling model of distribution networks and vehicle routing of TPSs and RCs is proposed,where the TPSs serve as emergency power supply sources,and the RCs are used to repair the faulted *** the uncertainty of traffic congestion,the probability distribution of the travel time spent on each road is derived based on the Nesterov user equilibrium model,and a two-stage stochastic program is formulated to determine the optimal routings of TPSs and *** efficiently solve the proposed stochastic mixed-integer linear program(MILP),a two-phase scenario reduction method is then developed to scale down the problem size,and an adaptive progressive hedging algorithm is used for an efficient *** effectiveness of the proposed methods and algorithms has been illustrated in a modified IEEE 33-bus system.

Evolution of microstructure and properties of a novel Ni-based superalloy during stress relief annealing

We discussed the decrease in residual stress,precipitation evolution,and mechanical properties of GH4151 alloy in different annealing temperatures,which were studied by the scanning electron microscope(SEM),high-resolution transmission electron microscopy(HRTEM),and electron backscatter diffraction(EBSD).The findings reveal that annealing processing has a significant impact on diminishing residual *** the annealing temperature rose from 950 to 1150℃,the majority of the residual stresses were relieved from 60.1 MPa down to 10.9 ***,the stress relaxation mechanism transitioned from being mainly controlled by dislocation slip to a combination of dislocation slip and grain boundary ***,the annealing treatment promotes the decomposition of the Laves,accompanied by the precipitation ofμ-(Mo_(6)Co_(7))starting at 950℃ and reaching a maximum value at 1050℃.The tensile strength and plasticity of the annealing alloy at 1150℃ reached the maximum(1394 MPa,56.1%)which was 131%,200%fold than those of the as-cast alloy(1060 MPa,26.6%),but the oxidation process in the alloy was accelerated at 1150℃.The enhancement in durability and flexibility is primarily due to the dissolution of the brittle phase,along with the shape and dispersal of theγ′phase.

Microstructural characterization and mechanical properties of(TiC+TiB)/TA15 composites prepared by an in-situ synthesis method

Titanium matrix composites reinforced with ceramic particles are considered a promising engineering material due to their combination of high specific strength,low density,and high *** this study,the TA15-based composites reinforced with a volume fraction of 10% to 25%(TiB+TiC)were prepared using powder metallurgy and casting *** characterization and phase constitution were examined using optical microscopy(OM),scanning electron microscopy(SEM),and X-ray diffraction(XRD).In addition,the microhardness,room temperature(RT)and high temperature(HT)tensile properties of the composites were *** revealed that the reinforcements are distributed uniformly even in the composites with a high volume of TiB and ***,as the volume fraction exceeds 15%,TiB and TiC particles become coarsening and exhibit rod-like and dendritic-like *** increases gradually from 321.2 HV for the base alloy to a maximum of 473.3 HV as the reinforcement increases to 25vol.%.Tensile test results indicate that a reinforcement volume fraction above 20% is beneficial for enhancing tensile strength and yield strength at high temperatures,but it has an adverse effect on room temperature ***,if the reinforcement volume fraction is below 20%,it can improve high-temperature elongation when the temperature exceeds 600℃.

PKHD1L1 blocks the malignant behavior of lung adenocarcinoma cells and restricts tumor growth by regulating CBX7

Objective:To explore the role of polycystic kidney and hepatic disease 1-like 1(PKHD1L1)in lung adenocarcinoma(LUAD).Methods:Bioinformatics tools were utilized to examine the clinical profile of PKHD1L1 and chromobox protein homolog 7(CBX7)in *** Cell Counting Kit-8,colony formation,terminal deoxynucleotidyl transferase dUTP nick end labeling,Transwell,and wound-healing assays were carried out to assess the proliferative,apoptotic,invasive,and migrative capacities of the ***,the interrelation between PKHD1L1 and CBX7 was validated using a co-immunoprecipitation assay.A LUAD mice model was constructed by subcutaneous injection of A549 ***,immunohistochemical staining was performed to evaluate CBX7 and Ki67 ***:PKHD1L1 was downregulated in LUAD and predicted dismal outcomes in patients with ***1L1 upregulation repressed the proliferative,invasive,and migrative capabilities of A549 cells and exacerbated the apoptotic ***,PKHD1L1 may bind to CBX7 and positively modulate CBX7 ***7 deletion partly abrogated the effects of PKHD1L1 upregulation on the cellular biological activities in A549 ***,the PKHD1L1/CBX7 axis regulates the Hippo signaling pathway in A549 ***1L1 restricted tumor growth in LUAD xenograft mice;this was partly abolished by CBX7 ***:PKHD1L1 can hinder LUAD progression by regulating CBX7-mediated Hippo signaling.

Development of a Mengla virus minigenome and comparison of its polymerase complexes with those of other filoviruses

Ebola virus(EBOV)and Marburg virus(MARV),members of the Filoviridae family,are highly pathogenic and can cause hemorrhagic fevers,significantly impacting human *** are considered reservoirs of these viruses because related filoviruses have been discovered in ***,due to the requirement for maximum containment laboratories when studying infectious viruses,the characterization of bat filoviruses often relies on pseudoviruses and minigenome *** this study,we used RACE technology to sequence the 30-leader and 50-trailer of Mengla virus(MLAV)and constructed a *** to MARV,the transcription activities of the MLAV minigenome are independent of *** further assessed the effects of polymorphisms at the 50 end on MLAV minigenome activity and identified certain mutations that decrease minigenome reporter efficiency,probably due to alterations in the RNA secondary *** reporter activity upon recombination of the 30-leaders and 50-trailers of MLAV,MARV,and EBOV with those of the homologous or heterologous minigenomes was compared and it was found that the polymerase complex and leader and trailer sequences exhibit intrinsic ***,we investigated whether the polymerase complex proteins from EBOV and MARV support MLAV minigenome RNA synthesis and found that the homologous system is more efficient than the heterologous *** efficiently inhibited MLAV as well as EBOV *** summary,this study provides new information on bat filoviruses and the minigenome will be a useful tool for high-throughput antiviral drug screening.

Advanced oxidation via the synergy of C-defective/C-O band modified ultrathin porous g-C3N4 and PMS for efficient photothermal degradation of bisphenol pollutants and lignin derivatives

This work uses thermal polymerization of urea nitrate, oxyacetic acid and urea as the raw material to prepare ultra-thin porous carbon nitride with carbon defects and C-O band（OA-UN-CN）. Density functional theory（DFT） calculations showed OA-UN-CN had narrower band gap,faster electron transport and a new internal construction electric field. Additionally, the prepared OA-UN-CN significantly enhanced photocatalytic activation of peroxymonosulfate（PMS） due to enhanced light absorption performance and faster electron overflow. As the result, the OA-UN-CN/PMS could entirely degrade bisphenol A（BPA） within 30 min, where the photodegradation rate was 81.8 and 7.9 times higher than that of g-C3N4and OA-UN-CN, respectively. Beyond, the OA-UN-CN/PMS could likewise degrade other bisphenol pollutants and sodium lignosulfonate efficiently. We suggested possible photocatalytic degradation pathways accordingly and explored the toxicity of its degradation products. This work provides a new idea on the development of advanced photocatalytic oxidation processes for the treatment of bisphenol pollutants and lignin derivatives, via a metal-free photothermal-catalyst.
