Background: Glycine dehydrogenase(GLDC) plays an important role in the initiation and proliferation of several human cancers. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnostic value for hepatitis B virus-associated hepatocellular carcinoma(HBV-HCC). Methods: We enrolled 197 patients, 111 with HBV-HCC, 51 with chronic hepatitis B(CHB), and 35 healthy controls(HCs). The methylation status of GLDC promoter in peripheral mononuclear cells(PBMCs) was identified by methylation specific polymerase chain reaction(MSP). The mRNA expression was examined using real-time quantitative polymerase chain reaction(q PCR). Results: The methylation frequency of the GLDC promoter was significantly lower in HBV-HCC patients(27.0%) compared to that in CHB patients(68.6%) and HCs(74.3%)( P < 0.001). The methylated group had lower alanine aminotransferase level( P = 0.035) and lower rates of tumor node metastasis(TNM) Ⅲ/Ⅳ( P = 0.043) and T3/T4( P = 0.026). TNM stage was identified to be an independent factor for GLDC promoter methylation. GLDC mRNA levels in CHB patients and HCs were significantly lower than those in HBV-HCC patients( P = 0.022 and P < 0.001, respectively). GLDC mRNA levels were significantly higher in HBV-HCC patients with unmethylated GLDC promoters than those with methylated GLDC promoters( P = 0.003). The diagnostic accuracy of alpha-fetoprotein(AFP) combined with GLDC promoter methylation for HBV-HCC was improved compared with that of AFP alone(AUC: 0.782 vs. 0.630, P < 0.001). In addition, GLDC promoter methylation was an independent predictor for overall survival of HBV-HCC patients( P = 0.038). Conclusions: The methylation frequency of GLDC promoter was lower in PBMCs from HBV-HCC patients than that from patients with CHB and HCs. The combination of AFP and GLDC promoter hypomethylation significantly improved the diagnostic accuracy of HBV-HCC.

The selective electrochemical conversion of glycerol into value-added products is a green and sustainable strategy for the biomass *** this work,Au nanowires(Au-NW)modified with polyethyleneimine(PEI)molecule(Au-NW@PEI)is obtained by an up-bottom post-modification *** characterization,molecular dynamics simulation and density functional theory demonstrate that the loose-packed PEI monolayer firmly and uniformly distribute on the Au-NW surface due to the strong Au-N *** experiments and product analysis display that PEI modification significantly enhance the electro-activity of Au-NW for the glycerol electro-oxidation reaction(GEOR)due to the electronic ***,the steric hindrance and electrostatic effect of PEI layer make the optimizing adsorption of intermediates ***,the selectivity of C3 product glyceric acid over Au-NW@PEI is increased by nearly 20%.The work thus indicates that the rational design of metal-organic interface can effectively elevate the electro-activity and selectivity of Au nanostructures,which may have wide application in biomass development.

To improve room-temperature hydrogen storage, palladium (Pd) nanoparticles were innovatively decorated by carbon bridge onto the amino-group functioned Zr-terephthalate metal-organic framework (MOF) UiO-66 to reduce the diffusion energy barrier and then improve the hydrogen spillover effect. Powder X-ray diffraction shows broad Pd peak and retained UiO-66-NH_(2) integrity after Pd decoration. The hydrogen uptake capacity show that UiO-66-NH_(2)-Pd exhibits best hydrogen storage performance than UiO-66-NH_(2) and pristine UiO-66. The hydrogen up taken in Pd decorated UiO-66 (UiO-66-NH_(2)-1Pd) was close to 4 wt% under 20 MPa at room temperature. Density functional theory (DFT) calculations show that hydrogen adsorption energy of UiO-66-NH_(2)-Pd was −0.5897 eV, which was much lower than that of UiO-66-NH_(2) (−0.3716 eV) and UiO-66 (−0.2975 eV). Ultimately, Pd decorated NH_(2) group functioned UiO-66 enable improve storage capacities through hydrogen spillover under ambient conditions which could satisfy the demand for sustainable energy, especially for the long-term storage energy media.

OBJECTIVE: To investigate the underlying protein molecular mechanisms of "Qi stagnation and blood stasis syndrome"(QS) and "Qi deficiency and blood stasis syndrome"(QD), as two subtypes of coronary artery disease(CAD) in Traditional Chinese Medicine(TCM),following percutaneous coronary intervention(PCI).METHODS: In this study, a total of 227 CAD patients with QS and 211 CAD patients with QD were enrolled;all participants underwent PCI. Label-free quantification proteomics were employed to analyze the changes in serum in two subtypes of CAD patients before and 6 months after PCI, aiming to elucidate the intervention mechanism of PCI in treating CAD characterized by two different TCM ***: Biochemical analysis revealed significant changes in tumor necrosis factor-α, high density lipoprotein cholesterol, blood stasis clinical symptoms observation, and Gensini levels in both patient groups post-PCI;Proteomic analysis identified 79 and 95 differentially expressed proteins in the QS and QD patient groups, respectively, compared to their control *** C8 alpha chain, complement factor H,apolipoprotein H, apolipoprotein B, plasminogen,carbonic anhydrase 2, and complement factor Ⅰ were altered in both comparison groups. Furthermore,enrichment analysis demonstrated that cell adhesion and connectivity-related processes underwent changes in QS patients post-PCI, whereas lipid metabolism-related pathways, including the peroxisome proliferator-activated receptor signaling pathway and extracellular matrix receptor interaction, underwent changes in the QD *** protein-protein interaction network analysis further enriched 52 node proteins, including apolipoprotein B,lipoprotein(a), complement C5, apolipoprotein A4,complement C8 alpha chain, complement C8 beta chain,complement C8 gamma chain, apolipoprotein H,apolipoprotein A-Ⅱ, albumin, complement C4-B,apolipoprotein C3, among others. Dielectric elastomers(DEs)have emerged as one of the most promising artificial muscle technologies,due to their exceptional properties such as large actuation strain,fast response,high energy density,and flexible processibility for various *** the past two decades,researchers have been working on developing DE materials with improved properties and exploring innovative applications of dielectric elastomer actuators(DEAs).This review article focuses on two main topics:recent material innovation of DEs and development of multilayer stacking processes for DEAs,which are important to promoting commercialization of *** begins by explaining the working principle of a ***,recently developed strategies for preparing new DE materials are introduced,including reducing mechanical stiffness,increasing dielectric permittivity,suppressing viscoelasticity loss,and mitigating electromechanical instability without *** the next section,different multilayer stacking methods for fabricating multilayer DEAs are discussed,including conventional dry stacking,wet stacking,a novel dry stacking method,and micro-fabrication-enabled stacking *** review provides a comprehensive and up-to-date overview of recent developments in high-performance DE materials and multilayer stacking *** highlights the progress made in the field and also discusses potential future directions for further advancements.

To investigate the influence of electric parameters and slag system on the solidification quality of electroslag ingot during electroslag remelting,different power supply modes,current strengths and remelting slag systems were used to conduct electroslag remelting experiments on 304L austenitic stainless steel,and the macrostructure of electroslag ingots was *** results indicate that the depth of the metal pool decreases with the reduction of remelting frequency in the low frequency power supply *** effects of different power supply modes,such as low-frequency,direct current straight polarity(DCSP),and direct current reverse polarity(DCRP),on reducing the depth of the metal pool increase in that *** reducing the remelting current strength in the same power supply mode,the depth of metal pool is *** compared to the binary slag system of 70%CaF2+30%Al2O3,the ternary slag system of 60%CaF2+20%Al2O3+20%CaO is more effective in reducing the depth of the metal pool during *** the 60%CaF2+20%Al2O3+20%CaO ternary slag system results in a shallower and flatter metal pool,with columnar crystal growth occurring closer to the axial *** effect is observed for both low frequency and direct current(DC)power supply modes.

The prolonged thermal exposure with centrifugal load results in microstructural degradation,which ultimately leads to a reduction in the fatigue and creep resistance of the turbine *** present work proposes a multi-scale framework to estimate the life reduction of turbine blades,which combines a microstructural degradation model,a two-phase constitutive model,and a microstructure-dependent fatigue and creep life reduction *** framework with multi-scale models is validated by a Single Crystal(SC)Ni-based superalloy at the microstructural length-scale and is then applied to calculate the microstructural degradation and the fatigue and creep life reduction of turbine blades under two specific service *** simulation results and quantitative analysis show that the microstructural degradation and fatigue and creep life reduction of the turbine blade are heavily influenced by the variations in the proportion of the intermediate state,namely,the maximum rotor speed status,in the two specific service *** intermediate state accelerates the microstructural degradation and leads to a reduction of the life,especially the effective fatigue life reserve due to the higher temperature and rotational speed than that of the 93%maximum rotor speed status marked as the reference *** proposed multi-scale framework provides a capable approach to analyze the reduction of the fatigue and creep life for turbine blade induced by microstructural degradation,which can assist to determine a reasonable Time Between Overhaul(TBO)of the engine.
