Protein@PP-Zn nanocomplex assembled by coordination of zinc ions used for intracellular protein delivery

In recent years,intracellular delivery of protein drugs has attracted great attention,and polymer-based systems have been extensively exploited to develop efficient and safe ***,efficient intracellular delivery of protein drugs remains a challenge because of the cell membrane barrier and endosome ***,we report a protein@PP-Zn nanocomplex,which consists of an imidazole-containing block polymer poly(ethylene glycol)-block-poly(β-amino ester)(PEG-b-PAE(Im),PP),zinc ions,and protein drugs,for efficient intracellular protein ***-b-PAE(Im)could conjugate proteins via the bridging effect of zinc ions which simultaneously coordinate with imidazole groups on polymer and electron donor groups,such as imidazole and primary amine groups,on protein to improve the loading stability of *** a slightly acidic environment near cancer cells,the protonation of PAE(Im)backbone increases the positive charge density of the nanocomplex and promotes *** under a more acidic environment in endosomes,further protonation of imidazole groups leads to the disintegration of the nanocomplex and the breakdown of endosomes because of the proton sponge ***,protein is released into the *** the assistance of the nanocomplex,proteins with different sizes and isoelectric points are effectively delivered into *** work provides a stable,efficient and universal strategy for intracellular protein delivery.

CONTINUOUS AUXILIARY COVARIATE IN ADDITIVE HAZARDS REGRESSION FOR SURVIVAL DATA

This paper considers the additive hazards iliary covariate information to improve the efficiency regression analysis by utilizing continuous aux- of the statistical inference when the primary covariate is ascertained only for a randomly selected subsample. The authors construct a martingale based estimating equation for the regression parameter and establish the asymptotic consistency and normality of the resultant estimators. Simulation study shows that the proposed method can greatly improve the efficiency compared with the estimator which discards the auxiliary covariate information in a variety of settings. A real example is also provided as an illustration.

Recent Progress in π-Conjugated Polymer-Inorganic Heterostructures for Photocatalysis

Polymer-inorganic(P-Ⅰ) soft-hard heterostructures & heterojunction photocatalysts,featured by large interfacial contact, efficient charge separation, broad light absorption and maximized redox capacity, have received great attention for their applications in energy conversion and environmental remediation. In this minireview, the classification and mechanism of P-Ⅰ heterojunctions, i.e., type-Ⅰ/Ⅱ, p-n, Z-scheme and S-scheme heterojunctions, and their preparation methods are firstly introduced. Next, the photocatalytic applications of P-Ⅰ heterojunctions, including water splitting, environmental remediation and carbon dioxide reduction, are extensively reviewed. Lastly,a brief summary and perspectives on ongoing challenges and opportunities to construct high performance P-Ⅰ soft-hard photocatalysts are intensively highlighted. We envision this review will provide a picture of the state-of-the-art achievements and promote the photocatalytic applications of P-Ⅰ heterostructures in energy conversion and environmental remediation.

Achieving the synergistic of strength and ductility in Mg-15Gd-1Zn-0.4Zr alloy with hierarchical structure

Currently,the hierarchical structure is one of the most effective means to enhance the strength and plasticity of metal materials,since the strain localization can be effectively delayed by the coordination of the unique *** this study,a hierarchical structure of Mg-15Gd-1Zn-0.4Zr(GZ151K)alloys containing grain,twin,and precipitation structural units was prepared by ultrasonic surface rolling process(USRP)and recrystallization annealing(RU).The results showed that the stress gradient generated by USRP formed a twin gradient structure,which will activate the twin-assisted precipitation(TAP)effect and twin-induced recrystallization(TIR)effect during ***,the twin gradient structure transformed into a twin-precipitation gradient structure,and finally into a hierarchical structure with grain-twinprecipitation as the increasement of recrystallization ***,the dual gradient structure with twin and precipitation structural units had the highest strength and microhardness owing to the precipitation ***,the hierarchical structure with grain,twin,and precipitation structural units exhibited the most excellent combination of strength and plasticity under grain refinement and precipitation strengthening.

Mainchain Semifluorinated Polymers with Ultra High Molecular Weight via Reaction-enhanced Reactivity of Intermediate(RERI) Mechanism

Achieving the linear polymers with high molecular weight via step-growth polymerization of A2 and B2 monomers is significantly limited by the requirement of strict stoichiometry of two monomers when the reactivity of A and B groups are not changed during the ***,a unique step-growth polymerization based on copper-catalyzed azide-alkyne cycloaddition(CuAAC)with reactionenhanced reactivity of intermediate(RERI)mechanism was developed for the preparation of mainchain semifluorinated polymers with high molecular *** CuAAC polymerization of bis-alkynyl-terminated fluorinated monomers(A2)and 2,2-bis(azidomethyl)propane-1,3-diyl bis(2-methylpropanoate)(BiAz,B2)with RERI effect at different stoichiometric ratio was systematically *** results indicated that the semifluorinated polymers with ultrahigh molecular weight,Mw,MALLS>106 g/mol,could be efficiently synthesized by using excess molar of BiAz *** resultant high-molecular-weight semifluorinated polymers show good thermostability and high *** addition,the glass transition temperature(Tg)of these mainchain semifluorinated polymers could be tuned conveniently due to the bis-alkynyl-terminated comonomers could be consumed completely when excessive BiAz monomers were used in this this step-growth polymerization.

背景:研究表明,血管因素对糖尿病足的发生具有重要影响。目的:探讨血管生成素样蛋白4在糖尿病足形成中的重要作用。方法:①对糖尿病足患者的基因表达谱数据进行生物信息学分析,找到关键基因。收集糖尿病足患者以及无糖尿病健康人的皮肤标本进行苏木精-伊红染色、免疫组化染色以及qRT-PCR实验,检测血管生成素样蛋白4表达情况。②培养人永生化皮肤成纤维细胞系和原代人脐静脉内皮细胞,将2种细胞分别分为对照组和外源性补充血管生成素样蛋白4组,通过划痕实验以及CCK-8实验分别检测成纤维细胞的迁移能力和增殖能力,通过Ki67实验检测内皮细胞的增殖能力。结果与结论:①生信分析发现,血管生成素样蛋白4基因的下调可能是导致糖尿病足形成的关键基因。②苏木精-伊红染色结果显示,与正常皮肤相比,血管生成素样蛋白在糖尿病足皮肤内弱表达,且其mRNA水平相对表达量降低(P<0.01)。③划痕实验结果显示,与对照组相比,血管生成素样蛋白4组成纤维细胞迁移能力明显增强;CCK-8细胞增殖实验显示,血管生成素样蛋白4组成纤维细胞的吸光度值在24,48 h均高于对照组(P<0.01,P<0.001);提示血管生成素样蛋白4可增强高糖处理的成纤维细胞迁移及增殖能力。④Ki67实验结果显示,与对照组相比,血管生成素样蛋白4组内皮细胞Ki67阳性细胞数目明显多于对照组;CCK-8细胞增殖实验显示,血管生成素样蛋白4组内皮细胞的吸光度值在24,48 h均高于对照组(P<0.05,P<0.001)。(5)以上结果均提示血管生成素样蛋白4可增强高糖处理内皮细胞的增殖能力。

FATOC:Bug Isolation Based Multi-Fault Localization by Using OPTICS Clustering

Bug isolation is a popular approach for multi-fault localization(MFL),where all failed test cases are clustered into several groups,and then the failed test cases in each group combined with all passed test cases are used to localize only a single ***,existing clustering algorithms cannot always obtain completely correct clustering results,which is a potential threat for bug isolation based MFL *** address this issue,we first analyze the influence of the accuracy of the clustering on the performance of MFL,and the results of a controlled study indicate that using the clustering algorithm with the highest accuracy can achieve the best performance of ***,previous studies on clustering algorithms also show that the elements in a higher density cluster have a higher *** on the above motivation,we propose a novel approach FATOC(One-Fault-at-a-Time via OPTICS Clustering).In particular,FATOC first leverages the OPTICS(Ordering Points to Identify the Clustering Structure)clustering algorithm to group failed test cases,and then identifies a cluster with the highest *** clustering is a density-based clustering algorithm,which can reduce the misgrouping and calculate a density value for each *** a density value of each cluster is helpful for finding a cluster with the highest clustering *** then combines the failed test cases in this cluster with all passed test cases to localize a single-fault through the traditional spectrum-based fault localization(SBFL)*** this fault is localized and fixed,FATOC will use the same method to localize the next single-fault,until all the test cases are *** evaluation results show that FATOC can significantly outperform the traditional SBFL technique and a state-of-the-art MFL approach MSeer on 804 multi-faulty versions from nine real-world ***,FATOC’s performance is 10.32%higher than that of traditional SBFL when using Ochiai formul
