Partial endothelial-to-mesenchymal transition(EndMT)is an intermediate phenotype observed in endothelial cells(ECs)undergoing a transition toward a mesenchymal state to support neovascularization during(patho)physiological ***,we investigated the occurrence of partial EndMT in ECs under hypoxic/ischemic conditions and identified general transcription factor IIH subunit 4(GTF2H4)as a positive regulator of this *** addition,we discovered that GTF2H4 collaborates with its target protein excision repair cross-complementation group 3(ERCC3)to co-regulate partial ***,by using phosphorylation proteomics and site-directed mutagenesis,we demonstrated that GTF2H4 was involved in the phosphorylation of receptor coactivator 3(NCOA3)at serine 1330,which promoted the interaction between NCOA3 and p65,resulting in the transcriptional activation of NF-κB and the NF-kB/Snail signaling axis during partial *** vivo experiments confirmed that GTF2H4 significantly promoted partial EndMT and angiogenesis after ischemic ***,our findings reveal that targeting GTF2H4 is promising for tissue repair and offers potential opportunities for treating hypoxic/ischemic diseases.

Pharmacogenomics can improve antipsychotic treatment in schizophrenia

Schizophrenia is a widespread mental disease with a prevalence of about 1%in the world population,and heritability of up to 80%.Drug therapy is an important approach to treating the ***,the curative effect of antipsychotic is far from satisfactory in terms of tolerability and side *** studies have indicated that about 30%of the patients exhibit little or no improvements associated with *** response of individual patients who are given the same dose of the same drug varies *** addition,antipsychotic drugs are often accompanied by adverse drug reactions(ADRs),which can cause considerable financial loss in addition to the obvious societal ***,it is strongly recommended that personalized medicine should be implemented both to improve drug efficacy and to minimize adverse events and *** is therefore a need for pharmacogenomic studies into the factors affecting response of schizophrenia patients to antipsychotic drugs to provide informed guidance for *** differences in drug response is due to a combination of many complex factors including ADEM(absorption,distribution,metabolism,excretion)process,transporting,binding with receptor and intracellular signal *** and pharmacogenomic studies have successfully identified genetic variants that contribute to this interindividual variability in antipsychotics *** addition,epigenetic factors such as methylation of DNA and regulation by miRNA have also been reported to play an important role in the complex interactions between the multiple genes and environmental factors which influence individual drug response phenotypes in *** this review,we will focus on the latest research on polymorphisms of candidate genes that code for drug metabolic enzymes(CYP2D6,CYP1A2,CYP3A4,etc.),drug transporters(mainly ABCB1)and neurotransmitter receptors(dopamine receptors and serotonin receptors,etc.).We also discuss the genome-wi

The unsymmetrical bis-Schiff base manganese（III） and cobalt（II） complexes with either benzo-10-aza-crown ether pendants （MnL1Cl, MnLZCl） or morpholino pendant （MnL3Cl, CoL3） have been employed as models for hy- drolase by studying the kinetics of their hydrolysis reactions with p-nitrophenyl picolinate （PNPP）. A kinetic model of PNPP cleavage catalyzed by these complexes is proposed. The effects of complex structures and reaction temperature on the rate of PNPP hydrolysis have been examined. All four complexes exhibit high catalytic activity and the rate increases with pH under 25℃. The complexes of ligands containing a crown ether group exhibit higher catalytic activities than the non-crown analogues. The catalytic activity of the complexes follows the order Mn（III）〉 Co（II） under the same ligands.

Drug response biomarkers of Pien Tze Huang(片仔癀) treatment for hepatic fibrosis induced by carbon tetrachloride

OBJECTIVE:To explore biomarkers of Pien Tze Huang(片仔癀)that ameliorated the symptoms of hepatic ***:Two groups of carbon tetrachloride-induced hepatic fibrosis(HF)mice model were constructed in our study:one group received PZH treatment and another group received no *** performed this study to investigate the role of PZH in the regulation process of hepatic ***:We identified 31 down-regulated and 39 upregulated mi RNAs using small RNA-seq *** RNA-Seq data analysis,our study revealed 7 significant target genes(Sp4,Slc2 a6,Tln2,Hmga2,Ank3,Pax9,Fgf9).The results of real-time quantitative polymerase chain reaction analysis suggested that the expression level of 6 genes(Sp4,Tln2,Hmga2,Ank3,Pax9,Fgf9)were down-regulated compared to control *** the other hand,the expression level of Slc2 a6 appeared to be *** protein mass spectrometry showed that PZH group had lower protein expression of Tln2 compared to control ***:We identified 7 genes that were significantly related to PZH response in HF mice using multiple conjoint analysis *** genes could participate in underlying regulation mechanism of hepatic fibrosis during PZH treatment.

Schizophrenia is a life-long,complex mental illness that still lacks satisfactory *** recent years,increasing numbers of candidate biomarkers of schizophrenia occurrences and drug responses to schizophrenia therapies have been successfully identified by many omics *** review discusses the latest discoveries regarding effective drug targets and relevant drug classifications in *** also assesses our understanding of biomarkers for drug efficacy and adverse drug reactions in current schizophrenia treatments using omics *** applications in clinical practice have been proposed based on these new findings,and are now considered highly promising strategies to better treat ***,we explore several novel approaches that aim to reveal additional genetic signatures of schizophrenia using multi-omics data,which are hoped to improve the diagnosis and treatment of this illness in the future.

四Schiff碱(二苯并-18-冠-6)及其钾(Ⅰ)/钴(Ⅱ)配合物的合成和性质

由二苯并-18-冠-6出发,合成了4′,4″,5′,5″-四(2-羟基苯亚甲基亚氨基)二苯并-18-冠-6(L^1H_4)及其5位取代衍生物(取代基R=CH_3,OH,OCH_3,Cl,NO_2)L^2H_4～L^6H_4.它们依次与硝酸钾和醋酸钴反应,制得1:1:2的钾(Ⅰ)/双钴(Ⅱ)配合物LCo_2·2H_2O·KNO_3(L=L^5,L^6)或其某些二氧加合物LCo_2(2O_2)·2H_2O·KNO_3(L=L^1～L^4).考察了取代基R和冠醚环配合的钾离子对二氧加合物形成及稳定性的影响.结果表明,含吸电子基(R=Cl,NO_2)的钾(Ⅰ)/双钴(Ⅱ)配合物不能形成二氧加合物;冠环中的钾离子会导致二氧加合物中两个Co—O_2健热稳定性的差异.

近年来,氮杂大环冠醚因可以作为生物膜活性物质的模型化合物、NMR成像造影剂、重金属解毒剂,及其金属配合物可改变入射光波长等性质已引起国内外广泛的注意[1].作者曾用NMR法、电位法对标题化合物的配合性质进行了研究[2,3],并发现这些配体在水溶液中不稳定,易发生水解.当水溶液的pH为7.0时,水解逐渐发生.配体1是最不稳定的,即使在酸性溶液中,数小时后也开始水解.按已发表的合成法[4],大环醚双内酯的产率低,实验重复性差,且原始产物通常为奶油状棕色粘性物,难以纯化.本文报道N,N′-二羧甲基大环醚双内酯的合成改进方法.
