Dynamic immune recovery process after liver transplantation revealed by single-cell multi-omics analysis

Elucidating the temporal process of immune remodeling under immunosuppressive treatment after liver transplantation(LT)is critical for precise clinical management strategies.Here,we performed a single-cell multi-omics analysis of peripheral blood mononuclear cells(PBMCs)collected from LT patients(with and without acute cellular rejection[ACR])at 13 time points.Validation was performed in two independent cohorts with additional LT patients and healthy controls.Our study revealed a four-phase recovery process after LT and delineated changes in immune cell composition,expression programs,and interactions along this process.The intensity of the immune response differs between the ACR and non-ACR patients.Notably,the newly identified inflamed NK cells,CD14+RNASE2+monocytes,and FOS-expressing monocytes emerged as predictive indicators of ACR.This study illuminates the longitudinal evolution of the immune cell landscape under tacrolimus-based immunosuppressive treatment during LT recovery,providing a four-phase framework that aids the clinical management of LT patients.

Traffic accidents of autonomous vehicles based on knowledge mapping:A review

As a characteristic representative of the new generation of vehicles,autonomous driving is expected to improve people's driving experience.The study on traffic accident of autonomous vehicles(AVs)helps provide suggestions for autonomous driving safety from multiple disciplines and perspectives,and provide support for formulating traffic accident treatment schemes.Knowledge mapping,as a cutting-edge research method in bibliometrics,scientifically and objectively displays the relevant research status using visual means.This paper uses CiteSpace 6.1.r3 to analyze 5068 related literature on the Web of Science database from 1991 to 2022 and finds out major thematic clusters,important documents and representative journals according to citation frequency.The results show that research on traffic accidents involving AVs focuses on accident preventing technologies,including how to avoid collisions,track lane-position,and enhance vehicle-toeverything(V2X)communication.This paper extracts the mean research topics and key points involved in the field and illustrates journals related to AVs and traffic accidents,which provides guidance for subsequent researchers to carry out in-depth research and contribute their papers.Popular journals are in disciplines of mathematics,systems,computer,economics,and social science.This paper also suggests scholars to consider the aspects of scene reconstruction,cause analysis,and injury of vulnerable road users,so as to investigate traffic accidents and put forward effective treatment schemes to reduce AV accidents,and ultimately improve road safety.

Both preclinical and established rheumatoid arthritis(RA)patients display alterations in the gut microbiome.Prevotella spp.are preferentially enriched in a subset of RA patients.Here,we isolated a Prevotella strain,P.copri RA,from the feces of RA patients and showed that colonization of P.copri RA exacerbated arthritis in a collagen-induced arthritis(CIA)model.With the presence of P.copri RA colonization,a high-fiber diet exacerbated arthritis via microbial alterations and intestinal inflammation.Colonization of P.copri together with a high-fiber diet enabled the digestion of complex fiber,which led to the overproduction of organic acids,including fumarate,succinate and short-chain fatty acids.Succinate promoted proinflammatory responses in macrophages,and supplementation with succinate exacerbated arthritis in the CIA model.Our findings highlight the importance of dysbiosis when evaluating the effects of dietary interventions on RA pathogenesis and provide new insight into dietary interventions or microbiome modifications to improve RA management.

MAT1 correlates with molecular subtypes and predicts poor survival in breast cancer

Objective：Menage a trois 1（MAT1）is a targeting subunit of cyclin-dependent kinase-activating kinase and general transcription factor IIH kinase,which modulates cell cycle,transcription and DNA repair.Its dysregulation is responsible for diseases including cancers.To further explore the role of MAT1 in breast cancer,we investigated the pathways in which MAT1 might be involved,the association between MAT1 and molecular subtypes,and the role of MAT1 in clinical outcomes of breast cancer patients.Methods：We conducted immunohistochemistry staining on tissue microarray and immunofluorescence staining on sections of MAT1 stable breast cancer cells.Also,we performed Kyoto Encyclopedia of Genes and Genomes pathway analysis,correlation analysis and prognosis analysis on public databases.Results：MAT1 was involved in multiple pathways including normal physiology signaling and disease-related signaling.Furthermore,MAT1 positively correlated with the protein status of estrogen receptor and progesterone receptor,and was enriched in luminal-type and human epidermal growth factor receptor 2-enriched breast cancer in comparison with basal-like subtype at both m RNA and protein levels.Correlation analysis revealed significant association between MAT1 m RNA amount and epithelial markers,mesenchymal markers,cancer stem cell markers,apoptosis markers,transcription markers and oncogenes.Consistently,the results of immunofluorescence stain indicated that MAT1 overexpression enhanced the protein abundance of epidermal growth factor receptor,vimentin,sex determining region Y-box 2 and sine oculis homeobox homolog 1.Importantly,Kaplan-Meier Plotter analysis reflected that MAT1 could serve as a prognostic biomarker predicting worse relapse-free survival and metastasis-free survival.Conclusions：MAT1 is correlated with molecular subtypes and is associated with unfavorable prognosis for breast cancer patients.

Fabrication of bilayer Pd-Pt nanocages with sub-nanometer thin shells for enhanced hydrogen evolution reaction

The hydrogen evolution reaction (HER),which generates molecular hydrogen through the electrochemical reduction of water,is an important clean-energy technology.Platinum (Pt) is an ideal material for HER electrocatalysts in terms of low overpotential and fast kinetics.An effective method to improve the atom utilization efficiency of Pt is to fabricate Pt-based core-shell or nanocage structures with ultra-thin walls.This paper describes the construction of bilayer palladium (Pd)-Pt alloy nanocages catalyst with enhanced HER catalytic activity.The nanocages were fabricated by etching away the Pd templates of multishelled nanocubes composed of alternate shells of Pd and Pt with well-defined (100) facets.The bilayer Pd-Pt nanocages with sub-nanometer shells have a high dispersion of the active atoms on the outside and inside surfaces of outer layer and inner layer,respectively.Moreover,the Pd-Pt alloy lowers the overpotential for HER and speeds up the reaction rate of HER due to the synergies between Pd and Pt.The rational design of bilayer nanocages provided a novel route for boosting the atom utilization efficiency of Pt catalysts.

在立德树人根本任务的指导下,新文科建设与课程思政建设成为高校教学改革的重要方向和内容,各类课程必须注重价值塑造、思想引领的育人效果。新闻传播伦理与法规课程注重培养学生的法治意识,提高学生的道德修养,其教学效果直接影响新闻传播人才的价值理念、从业操守,影响新闻舆论环境。积极思考该课程在新文科建设与课程思政建设背景下的教学改革并付诸实践,具有重要意义。文章结合笔者多年的教学实践,从教学目标设定、教学方法调整、学习资源建设、考核体系改革等方面展开论述,探讨教学各环节进行新文科建设与课程思政建设的方法与路径,旨在强化课程育人效果。课程教学改革实践表明,要明确且细化育人教学目标,层层设计,环环相扣,突出其德育功能;要重点改革教学方法,采取多种教学方法并行的手段,激发学生的学习兴趣,提高课堂的活跃度与参与度;要为学生自主学习能力的培养添砖加瓦,积极建设课外学习资源,拓宽学生的视野,提升学生的理论水平;要改革课程考核评价体系,保证各项考核内容灵活多样且有针对性,并合理分配比例,推动德育目标的实现。

目的:制备氨甲环酸醇质体(TA@ES),并初步考察其体外透皮性能。方法:采用乙醇注入-挤出法制备TA@ES。通过单因素考察和Box-Behnken设计-响应面法对处方进行筛选及优化;按最优处方制备TA@ES,对其进行质量评价,并进行体外透皮试验。结果:优化后的TA@ES处方脂药比为3.20∶1,胆固醇用量为0.62%,乙醇用量为19.37%。制得的TA@ES为类球形,平均粒径为(194.3±4.1)nm,Zeta电位为(-32.2±2.6)mV,包封率为(17.20±0.89)%。体外透皮试验结果表明,与氨甲环酸水溶液相比,TA@ES不仅提高了药物经皮渗透量(4.98倍),而且还增加了深层皮肤滞留量(1.74倍)。结论:制备的TA@ES能够提高氨甲环酸的经皮渗透量和深层皮肤滞留量,有望成为黄褐斑治疗药物的新载体。

济南市大一新生结核病防治知信行现状及行为影响因素

目的了解济南市大一新生结核病防治知识知晓、态度和行为现状,分析结核病防治行为影响因素,为有针对性开展健康教育工作提供依据。方法采用多阶段分层整群抽样方法抽取8所驻地为济南市的高校,利用问卷星平台对7564名大一新生进行问卷调查。调查内容包括基本信息、结核病防治核心知识知晓情况、结核病防治态度和行为、结核病防治知识获取渠道和偏好。采用Logistic回归分析影响该群体结核病防治行为的因素。结果济南市大一新生结核防治总知晓率为89.81%,其中“怀疑患病后正确做法”知晓率最高,为98.49%;“减免政策”知晓率最低,为79.71%。不同性别、专业、家人结核病史、本人结核病史的学生结核病知识知晓合格率存在差异(χ^(2)值分别为12.42、7.63、6.80、7.27),差异均有统计学意义(P<0.05)。结核病防治正确态度持有率为68.77%,不同性别、年龄、专业、同学朋友结核病史学生结核病态度持有情况存在差异(χ^(2)值分别为34.81、6.57、324.40、20.99),差异均有统计学意义(P<0.05);结核病防治正性行为发生率为92.50%,不同专业、防治知识知晓情况、防治态度持有情况的学生防治行为存在差异(χ^(2)值分别为77.84、74.67、27.79),差别均有统计学意义(P<0.01)。Logistic回归分析显示,非医学专业(OR=7.03,95%CI:4.11~12.02)是影响结核病防治行为的危险因素,知识知晓情况合格(OR=0.25,95%CI:0.17~0.37)、结核病防治态度正确(OR=0.78,95%CI:0.65~0.93)是影响结核病防治行为的保护因素(P均<0.05)。结论济南市大一新生结核病防治知信行情况较好,所学专业、防治知识知晓和态度情况是防治行为的主要影响因素。

Small but mighty:Empowering sodium/potassium-ion battery performance with S-doped SnO2 quantum dots embedded in N,S codoped carbon fiber network

SnO_(2) has been extensively investigated as an anode material for sodium-ion batteries(SIBs)and potassium-ion batteries(PIBs)due to its high Na/K storage capacity,high abundance,and low toxicity.However,the sluggish reaction kinetics,low electronic conductivity,and large volume changes during charge and discharge hinder the practical applications of SnO_(2)-based electrodes for SIBs and PIBs.Engineering rational structures with fast charge/ion transfer and robust stability is important to overcoming these challenges.Herein,S-doped SnO_(2)(S-SnO_(2))quantum dots(QDs)(≈3 nm)encapsulated in an N,S codoped carbon fiber networks(S-SnO_(2)-CFN)are rationally fabricated using a sequential freeze-drying,calcination,and S-doping strategy.Experimental analysis and density functional theory calculations reveal that the integration of S-SnO_(2) QDs with N,S codoped carbon fiber network remarkably decreases the adsorption energies of Na/K atoms in the interlayer of SnO_(2)-CFN,and the S doping can increase the conductivity of SnO_(2),thereby enhancing the ion transfer kinetics.The synergistic interaction between S-SnO_(2) QDs and N,S codoped carbon fiber network results in a composite with fast Na+/K+storage and extraordinary long-term cyclability.Specifically,the S-SnO_(2)-CFN delivers high rate capacities of 141.0 mAh g^(−1) at 20 A g^(−1) in SIBs and 102.8 mAh g^(−1) at 10 A g^(−1) in PIBs.Impressively,it delivers ultra-stable sodium storage up to 10,000 cycles at 5 A g^(−1) and potassium storage up to 5000 cycles at 2 A g^(−1).This study provides insights into constructing metal oxide-based carbon fiber network structures for high-performance electrochemical energy storage and conversion devices.

In this study,numerical simulation and discharge current tests were conducted on an inductively coupled radio frequency(RF)plasma cathode.Numerical simulations and experimental measurements were performed to study the factors influencing the electron extraction characteristics,including the gas type,gas flow,input power and extracting voltage.The simulation results were approximately consistent with the experimental results.We experimentally found that the RF input power mainly determines the extracted electron current.An electron current greater than 1 A was acquired at 270 W(RF input power),2.766 sccm(xenon gas).Our results prove that an inductively coupled RF plasma cathode can be reasonable and feasible,particularly for low power electric propulsion devices.
