To investigate the biological character of human adipose-derived adult stem cells (hADAS cells) when cultured in vitro and the relationship between hADAS cell’s replication activity and the donor’s age factor, and to assess the stem cells as a new source for tissue engineering. hADAS cells are isolated from human adipose tissue of different age groups (from adolescents to olds: 61 years old groups). The protein markers (CD29, CD34, CD44, CD45, CD49d, HLA-DR, CD106) of hADAS cells were detected by flow cytometry (FCM) to identify the stem cell, and the cell cycle was examined for P20 hADAS cells to evaluate the safety of the subculture in vitro. The generative activity of hADAS cells in different age groups was also examined by MTT method. The formula “ log2T D = t logN t ? logN 0” was used to get the time doubling (TD) of the cells. The results showed that the cells kept heredity stabilization by chromosome analysis for at least 20 passages. The TD of these cells increased progressively by ageing, and the TD of the he >61 years old group (statistical analysis of variance (ANOVA), P=0.002, Phese find- ings suggested that a higher level of hADAS cells replication activity was found in the younger dona- tors, and they represent novel and valuable seed cells for studies of tissue engineering.

BACKGROUND: The life expectancy of a patient with primary hepatic carcinoma (PHC) is hard to predict, and it is related to many prognostic factors. The Chinese classification system including five parameters: tumor, vascular thrombosis, lymph node metastasis, distant metastasis and Child-Pugh stage developed in 1999 was adopted by the 8th National Conference on liver Cancer of the Chinese Anti-Cancer Association in 2001. In this study, the discriminatory ability of the Chinese classification system -was compared with that of the TNM staging in patients for resection of PHC, in addition to the evaluation of prognostic value. METHODS: The data of 246 patients who had undergone resection of PHC from January 1986 to December 2000 (average age, 51 years; male/female ratio, 213/33) were retrospectively studied. Among the 246 patients, 227 were followed up for at least 3 years. RESULTS: The 1-, 3-, 5-, 7-, and 10-year tumor-free survival rates were 55% , 30% , 25% , 20% and 18% , respectively. The Chinese classification system was better than the TNM staging system in predicting survival rate of patients with PHC, as confirmed by survival curves shown by the Kaplain-Meier method. The mean survival time was 155, 70, 39, 16, and 4 months in patients with die Chinese classification stages Ⅰa, Ⅰb, Ⅱa,Ⅱb, and Ⅲ, respectively. The 1-, 3-, 5-, 7-, and 10-year tumor-free survival rates of the Chinese classification system and TNM staging were statistically significant and had a slightly positive relationship. The predictive capacity of the Chinese classification system was confirmed in any two subgroups of patients undergoing operation. COX proportional hazards regression analysis showed that the Chinese classification system was the only independent prognostic factor for survival. CONCLUSIONS: Taking both tumor extension and liver function into account, we consider that the Chinese classification system making up for the deficiency of UICC TNM staging is more precise in predictin

AIM: To investigate the inhibition effects on the productionof collagen type I, Ⅲ secreted by activated rat hepatic stellatecells (rHSCs) by antisense tissue inhibitors of metalloproteinase1 (TIMP-1) recombinant plasmid through elevating interstitialcollagenase ***: rHSCs were extracted from normal rat liverby pronase and collagenase digestion and purified bycentrifugal elutriation, and were cultured on plastic dishesuntil they were activated to a myofibroblastic phenotypeafter 7-10 days. RT-Nest-PCR and gene recombinanttechniques were used to construct the rat antisense TIMP-1 recombinant plasmids which can express in eucaryoticcells. The recombinant plasmid and the pcDNA3 emptyplasmid were transfected in rHSCs by Effectene (QIAGEN)separately. Cells were selected after growing in DMEMcontaining 400 μg/ml G418 for 2-3 weeks. Expression ofexogenous gene was assessed by Northern blot, andexpression oflIMP-1 in rHSCs was determined by Northernblot and Western blot. We tested the interstitial collagenaseactivity with FITC-labled type I collagen as ***, we quantified the type Ⅰ, Ⅲ collagen byWestern ***: The exogenous antisense TIMP-1 recombinantplasmid could be expressed in rHSCs well, which couldblock the expression of TIMP-1 greatly, the ratio of TIMP-1/GAPDH was 0.67, 2.41, and 2.97 separately at mRNAlevel (P＜0.05); the ratio of TIMP-1/β-actin was 0.31, 0.98and 1.32 separately at protein level (P＜0.05); It mightelevate active and latent interstitial collagenase activity,the collagenase activity was 0.3049, 0.1411 and 0.1196respectively. AIM: To study the effect of 5-lipoxygenase inhibitors on the expression of intercellular adhesion molecule-1 (ICAM-1) inmelanoma cells. METHODS: ICAM-1 protein of human melanoma cell a375 was detected by enzyme-linkedimmunosorbent, flow cytometry and Western blot analysis. Level of ICAM-1 mRNA in a375 was evaluated byNorthern blot analysis. Adhesion of a375 to endothelial cell EC304 was analyzed by isotopic tracing. RESULTS:5-Lipoxygenase inhibitors nordihydroguaiaretic acid, AA861 and MK886, could suppress the expression of ICAM-1protein as well as of its mRNA in a375 cells and reduce the adhesion of a375 to EC304. CONCLUSION:5-Lipoxygenase inhibitors can inhibit the expression of ICAM-1 in human melanoma cells and may be valuable fortreatment of melanoma metastasis.

天然麻粒岩高温流变实验研究

本实验在气体介质三轴高温流变仪上,采用怀安瓦窑口麻粒岩,在温度900～1200℃、围压300 MPa、应变速率10^(-4)～10^(-6)/s条件下,开展高温流变实验.实验样品麻粒岩由斜长石(52%)、单斜辉石和斜方辉石(40%)、石英(3%)、磁铁矿和钛铁矿(5%)组成,矿物平均粒度为:斜长石294μm、单斜辉石和斜方辉石282μm、石英97μm、磁铁矿和钛铁矿109μm.利用傅里叶变换红外光谱仪分析获得变形后样品的水含量约为0.17±0.05wt%.实验样品的强度随温度升高而降低,随应变速率降低而降低.基于力学数据,采用稳态流变方程,获得实验样品在900～1000℃时的应力指数为8.1～12.9,在1050～1150℃时的应力指数为4.8～5.8,平均值5.2.应力指数随着温度升高而降低.显微结构和成分分析表明,在900℃时麻粒岩出现矿物压扁与定向拉长特征,样品以位错滑移和微破裂变形为主;在950～1000℃时,麻粒岩样品中颗粒边界变得圆滑,表现出位错攀移特征,辉石和磁铁矿边缘出现微量熔体;在1050～1200℃时麻粒岩出现部分熔融,而且随着温度和实验时间(应变)增加,熔体含量增加,熔体结晶出微粒斜长石、辉石和橄榄石,部分辉石通过固体反应生成橄榄石.颗粒边界熔体和矿物反应促进了扩散作用,导致位错攀移和熔体引起的扩散蠕变共同控制了麻粒岩的高温流变.

A LQP BASED INTERIOR PREDICTION-CORRECTION METHOD FOR NONLINEAR COMPLEMENTARITY PROBLEMS

To solve nonlinear complementarity problems （NCP）, at each iteration, the classical proximal point algorithm solves a well-conditioned sub-NCP while the Logarithmic-Quadratic Proximal （LQP） method solves a system of nonlinear equations （LQP system）. This paper presents a practical LQP method-based prediction-correction method for NCP. The predictor is obtained via solving the LQP system approximately under significantly relaxed restriction, and the new iterate （the corrector） is computed directly by an explicit formula derived from the original LQP method. The implementations are very easy to be carried out. Global convergence of the method is proved under the same mild assumptions as the original LQP method. Finally, numerical results for traffic equilibrium problems are provided to verify that the method is effective for some practical problems.
