The close relationship between the genesis of coal metamorphism and the evolution of the regional tectonic framework.is expounded on the basis of an analysis of the geological factors causing the metamorphic zonation of the Late Palaeozoic coals in southern North China; in terms of the mechanism for the formation of palaeogeothermal anomalies, the effects of thermal groundwater on coal metamorphism is highlighted and a geological model for thermal groundwater metamorphism of coal established; with the concept of the palaeogeotherm-coal metamorphism system, the genesis of coal metamorphism is analysed according to the distribution pattern of three geological factors: heat source, heat-carrier and channel, and heat-collector.

The well documented source for adult multipotent stem cells is spermatogonial stem cells (SSCs) of mammalian testis. It is foundation of spermatogenesis in the testis throughout adult life by balancing self-renewal and differentiation. SSCs isolation from mammalian testis is difficult because of their scarcity and the lack.of well characterized cell surface mark.rs. Thus, the isolation of SSCs is of great interest for exploration of spermatogonial physiology and therapeutic approaches for fertility preservation. CD9 is a surface mark.r expressed in mouse and rat male germline stem cells. In this study, CD9 positive SSCs were successfully isolated from the goat testis using enzymatic digestion followed by three step purification: Differential plating, Percoll discontinuous density gradient followed by Magnetic activated cell sorting (MACS). Percoll discontinuous density gradient showed significant differences in the percentage of CD9+ SSCs across individual fraction. The fraction 36% and 40% gave the highest percentage of CD9+ SSCs i.e. 82% ± 1.2 and 9.2% ± 1.3 respectively. Magnetic activated cell sorting of CD9+ cells in the magnetic fraction of goat testes was in the range of 15% - 18% which is upto threefolds. CD9+ SSCs were further recovered with appreciable efficiency after immunomagnetic isolation by using various bead: cells ratio in which 4:1 ratio gave the highest yield of 69.06 × 105 with 18% of CD9+ SSCs. Magnetic activated cell sorting using anti-CD9 antibodies provides an efficient and fast approach as compared to conventional approaches such as differential plating and percoll discontinuous density gradient for enrichment strategy for spermatogonial stem cells from goat testes for undertak.ng research on basic and applied reproductive biology.

The influence of methionine supplementation on the efficacy of common antidotes to lead poisoning, calcium disodium ethylenediaminetetraacetate (CaNa_2EDTA) and D-penicillamine (DPA), was investigated in rats. The animals were given lead acetate (0.1% in drink.ng water) for 12 week. and thereafter treated with CaNa_2EDTA, DPA (0.3mmol/k., intraperitoneally), DL-methionine (1.34 mmol/k., intragastrically), or the combination of a chelating agent and methionine for 3 days. While chelating agents enhanced the urinary excretion of Pb, methionine increased the fecal excretion of Pb significantly. Treatment with the combination of a chelating agent and methionine did not potentiate the effect of each antidote. However, methionine supplementation increased the efficacy of both chelating agents in reducing the hepatic and renal Pb burden but not the blood Pb level. The Pb-induced inhibition of blood δ-aminolevulinic acid dehydratase activity and the increase in urinary excretion of δ-aminolevulinic acid were reversed to a certain extent by CaNa_2EDTA, DPA, and methionine but the combination did not improve their individual performances. The beneficial effects of methionine may be attributed to its ability to increase the bioavailability of glutathione (GSH), useful in chelating Pb and counteracting the toxic effects, as evidenced by restoration of the Pb-induced decrease in hepatic GSH level by treatment with methionine. Methionine may be useful as a supportive therapy in chelation of Pb. (c)1989 Academic Press. Inc.

Artemisinin is highly effective against drug-resistant malarial parasites, which affects nearly half of the global population and k.lls 〉500 000 people each year. The primary cost of artemisinin is the very expensive process used to extract and purify the drug from Artemisia annua. Elimination of this apparently unnecessary step will mak. this potent antimalarial drug affordable to the global population living in endemic regions. Here we reported the oral delivery of a non-protein drug artemisinin biosynthesized （~0.8 mg/g dry weight） at clinically meaningful levels in tobacco by engineering two metabolic pathways targeted to three different cellular compartments （chloroplast, nucleus, and mitochondria）. The doubly transgenic lines showed a three-fold enhancement of isopentenyl pyrophosphate, and targeting AACPR, DBR2, and CYP71AV1 to chloroplasts resulted in higher expression and an efficient photo-oxidation of di- hydroartemisinic acid to artemisinin. Partially purified extracts from the leaves of transgenic tobacco plants inhibited in vitro growth progression of Plasmodium falciparum-infected red blood cells. Oral feeding of whole intact plant cells bioencapsulating the artemisinin reduced the parasitemia levels in challenged mice in comparison with commercial drug. Such novel synergistic approaches should facilitate low-cost production and delivery of artemisinin and other drugs through metabolic engineering of edible plants.

[目的]通过比较健康人和不同严重程度分级的COPD患者股四头肌、肋间肌中碳酸酐酶Ⅲ(CAⅢ)浓度及其mRNA表达量的不同,探讨CAⅢ和COPD严重程度之间的关系。[方法]本研究中股四头肌标本取自75名来自昆明医科大学第一附属医院骨科的患者入选,其中有25名接受骨科手术的非COPD患者,24名轻度COPD患者和26名中度COPD患者。肋间肌标本来自另外72名就诊于昆明医科大学第一附属医院胸外科的患者,其中有23名接受胸外科手术的非COPD患者,24名轻度COPD患者和25名中度COPD患者。所有受试者按标本来源分为股四头肌组(Q组)和肋间肌组(C组)。随后每组患者按照肺功能的严重程度不同分为三个亚组:对照组(FEV1/FVC>70%,FEV1%(FEV1predicted)≥80%);Ⅰ 组(FEV1/FVC<70%,FEV1%(FEV1predicted)≥80%);Ⅱ 组(FEV1/FVC<70%,30%≤FEV1%(FEV1predicted)<80%)。所有标本的 CAⅢ和 CAⅢmRNA 通过ELISA和RT-PCR进行测定并组间进行比较。[结果]Q对照组和C对照组的CAⅢ浓度和CAⅢ mRNA表达量比较,差异没有统计学意义。统计学分析结果显示QI组和CI组的对比中CA Ⅲ表达量的差异达到了统计学差异(12.10±1.10,11.03±0.90;t=3.69,P<0.001)。同样,CAⅢ表达量在QⅡ组和CⅡ组之间也达到了统计学差异(15.40±1.05,13.52±0.91;t=6.85,P<0.001)。在CAⅡ mRNA表达量(2-△△Ct)的分析中,QI组和CI组的对比并未发现 CAⅢ mRNA 表达量有所不同(2.18±0.95,2.10±0.84;t =0.305,P=0.76),而QⅡ组和CⅡ组的CAⅢ mRNA表达量却有明显不同(3.79±1.03,3.01±0.82;t=2.96,P=0.005)。亚组分析结果显示,Q对照组、QⅠ组和QⅡ组CAⅢ表达量随着COPD严重程度有明显差异(CAⅢ(ng/ml):10.13± 1.22,12.10± 1.10,15.40± 1.05;F =143.85,P<0.001),同时三组的 CAⅢ mRNA(2-△△Ct)表达量也有明显差异(1.00±0.44、2.18 ± 0.95、3.79±1.03;F=69.88,P<0.001)。C 组的组内亚组分析结果同样显示,CAⅢ和CAⅢ mRNA表达量随着COPD严重程度不同而有明显统计学差异(C对照组,CⅠ组和CⅡ组的CAⅢ(ng/ml):10.09±0.88、11.03±0.90、13.52±0.91;F=94.58,P<0.001;C 对照组,CⅠ 组和 CⅡ组的 CAⅢ mRNA(2-△△Ct):1.00±0.43、2.10±0.84、3.01 ±0.82;F =45.89,P<0.001).[结论]COPD患者骨骼肌中的CAⅢ和CAⅢ mRNA表达量较正常人明显增高,这种现象在股四头肌中尤为明显。在不同严重程度的COPD患者中,CAⅢ和CAⅢ mRNA表达量随着肺功能的严重程度的不同而有所不同。这也表明了 CAⅢ和CAⅢ mRNA的表达量不仅和系统因素有关,同时和局部因素也有密切关系。

The present study deals with the co-ordination of cytok.ne(IL-4 and IFN-γ) expression and k.netics of peste des petits ruminants(PPR) virus antigen and antibody in PPRV infected and vaccinated *** infected animals exhibited mixed cytok.ne(both T_H1 and T_H2) responses in the initial phase of the *** infected and dead goats had increased IFN-γ response before their death;while IL-4 remained at the base *** cytok.ne expression in recovered animals was almost similar to that of vaccinated ones,where a unique biphasic response of IL-4 expression was observed with an up-regulation of IFN-γ on 7^(th) days post vaccination(dpv).Analysis of PPR virus antigen and antibody k.netics in different components of blood from infected and vaccinated animals revealed that the PPR virus antigen load was highest in plasma followed by serum and blood of the infected animals,whereas vaccinated animals showed only marginal positivity on 9^(th) *** antibody titer was high in serum followed by plasma and blood in both vaccinated and infected ***,it is inferred that the presence of antigen and antibody were significant with the expression of cytok.ne,and that a decreased response of IL-4 was noticed during intermediate phase of the disease i.e.,7 to 12^(th) days post infection(dpi).This indicates the ability to mount a functional T_H2 response after 14^(th)dpi could be a critical determinant in deciding the survival of the PPR infected

Effects of roller burnishing process parameters on surface roughness of A356/5%SiC composite using response surface methodology

In this study, a simple roller burnishing tool was made to operate burnishing processes on A356/5%SIC metal matrix composite fabricated by electromagnetic stir casting under different parameters. The effects of burnishing speed, burnishing force and number of burnishing passes on the surface roughness and tribological properties were measured. Scanning electron microscopy （SEM） graphs of the machined surface with PCD （insert-10） tool and roller burnished surface with tungsten carbide （WC） roller were tak.n into consider- ation to observe the surface finish of metal matrix composites. The mechanical properties （tensile strength, hardness, duc- tility） of A356/5%SIC metal matrix composites were studied for both unburnished samples and burnished samples. The results revealed that the roller burnished samples of A356/ 5%SIC led to the improvement in tensile strength, hardness and ductility. In order to find out the effects of roller bur- nishing process parameters on the surface roughness of A356/ 5%SIC metal matrix composite, response surface methodol- ogy （RSM）（Box-Behnk.n design） was used and a prediction model was developed relevant to average surface roughness using experimental data. In the range of process parameters, the result shows that roller burnishing speed increases, and surface roughness decreases, but on the other hand roller burnishing force and number of passes increase, and surface roughness increases. Optimum values of burnishing speed （1.5 m/s）, burnishing force （50 N） and number of passes （2） during roller burnishing of A356/5%SIC metal matrix com- posite to minimize the surface roughness （predicted 1.232 μm） have been found out. There was only 5.03% error in the experimental and modeled results of surface roughness.
