Analysis of human B cell response to recombinant Leishmania LPG3

Objective: To evaluate the capability of recombinant Leishmania LPG3 and its fragments in the activation of B cells. Methods: In the present study, human B cells were purified from peripheral blood of 10 adult healthy subjects using magnetic-activated cell sorting technique. Subsequently, purified B cells were treated with recombinant LPG3, and its N-terminal and C-terminal fragments at different concentrations(2, 10 and 20 μg/m L). B cell activation was assessed through expression of CD69 molecule by flowcytometry and secretion of IL-6, TNF-α and IL-10 cytokines via enzyme-linked immunosorbent assay following treatment with recombinant antigens. Results: Our results showed that while the recombinant LPG-3 could significantly increase the production of IL-6 and TNF-α(P<0.05) in B cells, it had no effect on the secretion of IL-10 by B cells. Conclusions: Our study indicated that recombinant LPG-3 and especially its N-terminal fragment could stimulate B cell response as an important immune response component against leishmaniasis. Thus, it seems that it can be considered as an effective adjuvant in vaccine developments against leishmaniasis.

AIM To evaluate the effect of body mass index(BMI) on short-term functional outcome and complications in primary total knee arthroplasty. METHODS All patients undergoing primary total knee arthroplasty at a single institution between 2007 and 2013 were identified from a prospective arthroplasty database. 2180 patients were included in the study. Age, gender, BMI, pre- and post-operative functional scores [Western Ontario and Mc Master University Arthritis Index(WOMAC) and SF-36], complications and revision rate were recorded. Patients were grouped according to the WHO BMI classification. The functional outcome of the normal weight cohort(BMI 0.05). Sub-group analysis revealed significantly worse WOMAC scores in class 2 obese 30.80 compared to overweight 25.80(P 25. Patients should not be denied total knee arthroplasty based solely on weight alone.

Background:Dietary supplementation with a fucoidan-rich Ascophyllum nodosum extract(ANE),possessing an in vitro anti-Salmonella Typhimurium activity could be a promising on-farm strategy to control Salmonella infection in *** objectives of this study were to:1)evaluate the *** activity of ANE(containing 46.6%fucoidan,18.6%laminarin,10.7%mannitol,4.6%alginate)in vitro,and;2)compare the effects of dietary supplementation with ANE and Zinc oxide(ZnO)on growth performance,Salmonella shedding and selected gut parameters in naturally infected *** was established post-weaning(newly weaned pig experiment)and following regrouping of post-weaned pigs and experimental re-infection with ***(challenge experiment).Results:In the in vitro assay,increasing ANE concentrations led to a linear reduction in *** counts(P0.05).ANE supplementation decreased the Enterobacteriaceae counts compared to the *** counts were also reduced in the ZnO-residual group compared to the control(P<0.05).ANE supplementation decreased the expression of interleukin 22 and transforming growth factor beta 1 in the ileum compared to the control(P<0.05).Conclusions:ANE supplementation was associated with some beneficial changes in the composition of the colonic microbiota,Salmonella sh

Background： The effect of time of fecal sampling on the accuracy of acid-detergent insoluble ash （ADIA） and alkaline-peroxide lignin （APL） for the prediction of fecal output （FO） in cattle was evaluated. Eight ruminally cannulated cows （594 _＋ 35.5 kg） were allocated randomly to 4 bermudagrass [Cynodon dactylon （L.） Pers.] hay diets markedly different in crude protein concentration （79-164 g/kg） with 2 replicates per diet for 3 periods. Cows were offered hay individually at 20 g DM/kg of body weight daily in equal feedings at 08：00 and 16：00 h for a 10-d adaptation period followed by 5-d of total fecal collection. Fecal grab samples also were taken each day during the fecal collection period at 06：00, 12：00, 18：00, and 24：00 h either directly from the rectum or from freshly voided feces. Samples were composited within cow and time across the 5 d total fecal collection period. Additionally, forage, ort, and fecal samples were analyzed for concentrations of APL and ADIA. Results： Fecal concentrations of ADIA and APL were not affected by sampling time （P 〉 0.22）, even though diet affected （P 〈 0.01） fecal ADIA and APL concentrations. There were no diet x sampling time interactions （P ≥ 0.60）. Estimates of FO and dry matter digestibility （DMD） from ADIA and APL were not affected （P 〉 0.16） by sampling time or the diet x sampling time interaction （P 〉 0.74）. Estimates of FO and DMD from markers from different sampling times or all different combinations of sampling time were not different （P 〉 0.72） from those of total collection among internal markers. Conclusion： Little variation in concentrations of ADIA and APL in daily fecal excretion across time increases flexibility in fecal grab sampling schedules for predicting FO and DMD.

The pyrolysis of zeolitic imidazolate frameworks(ZIFs) is becoming a popular approach for the synthesis of catalysts comprising porphyrin-like metal single atom catalysts(SACs) on N-doped carbons(M-N-C).Understanding the structural evolution of M-N-C as a function of ZIF pyrolysis temperature is important for realizing high performance ***,we report a detailed investigation of the evolution of Zn single atom catalyst sites during the pyrolysis of ZIF-8 at temperatures ranging from 500 to 900℃.Results from Zn L-edge and Zn K-edge X-ray absorption spectroscopy studies reveal that tetrahedral ZnN4 centers in ZIF-8 transform to porphyrin-like ZnN4 centers supported on N-doped carbon at temperatures as low as 600℃.As the pyrolysis temperature increased in the range 600-900℃,the Zn atoms moved closer to the N4 coordination *** subtle geometry change in the ZnN4 sites alters the electron density on the Zn atoms(formally Zn2+),strongly impacting the catalytic performance for the peroxidase-like decomposition of H2 *** catalyst obtained at 800℃(Zn-N-C-800) offered the best performance for H2 O2 *** work provides valuable new insights about the evolution of porphyrin-like single metal sites on N-doped carbons from ZIF precursors and the factors influencing SAC activity.
