Discriminating ability of molecular markers and morphological characterization in the establishment of genetic relationships in cultivated genotypes of almond and related wild species

A total 23 morphological traits, 19 AFLP-primer combinations, 80 RAPD primers and 32 SSR primer pair were used to compare the informativeness and efficiency of random amplified polymorphic DNA (RAPD), amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers in establishing genetic relationships among 29 almond cultivars and three related wild species. SSRs presented a high level of polymorphism and greater information content, as assessed by the expected hetrozygosity, compared to AFLPs and RAPDs. The lowest values of expected hetrozygosity were obtained for AFLPs; however AFLPs showed the highest efficiency, owing to their capacity to reveal large numbers of bands per reaction, which led to high values for various types of indices of diversity. All the three techniques discriminated almond genotypes very effectively, except that SSRs failed to discriminate between ‘Monagha’ and ‘Sefied’ almond genotypes. The correlation coefficients of similarity were statistically significant for all the three marker systems, but were lower for the SSR data than for RAPDs and AFLPs. For all the markers, high similarity in dendrogram topologies was obtained, although some differences were observed. All the dendrograms, including that obtained by the combined use of all the marker data, reflect relationships for most of cultivars according to their geographic diffusion. AMOVA detected more variation among cultivated and related wild species of almond within each geographic group. Bootstrap analysis revealed that the number of markers used was sufficient for reliable estimation of genetic similarity and for meaningful comparisons of marker types.

Synthesis of Nano Shell by Amino Functionalization of Multi-Walled Carbon Nano Tubes

The fabrication of buckypapers from different amino functionalized Multi walled Carbon nanotubes (MWCNTs) is accomplished. After purification and oxidation of MWCNTs with HNO3, the product reacts with Oxalyl chloride until lead to MWCNTs-COCl. This product reacts by several amine compounds such as 1, 4-phenylen di amine and 1, 8-di amino octane and 1, 2-di amino propane separately. Each product in each step characterized by spectroscopic methods such as Fourier transform Infrared Spectra (FT-IR), Raman Spectra, Nuclear Magnetic Resonance (NMR) and scanning electron microscopy (SEM). All of derivatives lead to nano structures such as buckypapers, nano sheet, and nano bobble shell.

At semi-arid areas that dominant portion of required water for agriculture is provided by exploitation of groundwater,these resources encounter with more *** identifying of potential runoff generating sites and estimation of runoff depth can be a significant step for storing runoff for agricultural activities and groundwater *** main purposes of this study are use of GIS(geographic information system)ability for identifying of potential runoff generating sites,and thus identifying high priority areas for groundwater recharge in the Gharehchay River watershed in the north of Hamedan province,*** runoff generating sites were identified by using watershed features same as slope,land use and hydrological soil ***,CN I(Curve Number I)technique,which is one of the eight derivations of the NRCS-CN(natural resources conservation services curve number)method,was utilized to calculate rainfall-runoff depth in the study ***,map layers were ranked in order of highest priority to lowest priority,based on the criteria of each dataset,and high priority areas for groundwater recharge were identified by integrating potential runoff map,runoff depth and depth to groundwater *** analysis revealed that 51%of the study region has a high priority for groundwater recharge.

Objective:To identify the pathogenic gene of Panton-Valentine leukocidin in methicillin-resistant Staphylococcus aureus strain isolated from clinical specimens of burn patients in Shaheed Zare Burn ***:A total of 104 samples of Staphylococcus were collected and 78 aureus samples were isolated from Zare Hospital patients from November 2016 to July *** isolates were identified using a standard biochemical and laboratory methodology in accordance with CLSI principles,and disk agar diffusion antibiogram were performed to identify methicillin resistant *** the presence of the Panton-Valentine leukocidin gene was tested by ***:Of the methicillin-resistant Staphylococcus aureus samples,80%were negative for the Panton-Valentine leukocidin gene,and only 20%of the samples had Panton-Valentine leukocidin *** and female patients showed no significant difference in the positivity rate of Panton-Valentine leukocidin gene(16.12%vs.33.33%)(P=0.25).Besides,there was no significant difference in bacterial resistance or susceptibility to antibiotics between samples with or without the Panton-Valentine leukocidin ***:In recent years,increased resistance has been a serious *** resistance or susceptibility of Staphylococcus aureus strains to different antibiotics is different in different geographical locations.

Recombinant outer membrane protein F-B subunit of LT protein as a prophylactic measure against Pseudomonas aeruginosa burn infection in mice

AIM: To study immunogenicity of outer membrane protein F(Opr F) fused with B subunit of LT(LTB), against Pseudomonas aeruginosa(P. aeruginosa). METHODS: The Opr F, a major surface exposed outer membrane protein that is antigenically conserved in various strains of P. aeruginosa, is a promising immunogen against P. aeruginosa. In the present study recombinant Opr F and Opr F-LTB fusion gene was cloned, expressed and purified. BALB/c mice and rabbits were immunized using recombinant Opr F and Opr F-LTB and challenged at the burn site with P. aeruginosa lethal dose of 104 CFU. The protective efficacy of rabbit anti Opr F Ig G against P. aeruginosa burn infection was investigated by passive immunization. RESULTS: It has been well established that the LTB is a powerful immunomodulator with strong adjuvant activity. LTB as a bacterial adjuvant enhanced immunogenicity of Opr F and anti Opr F Ig G titer in serum was increased. Experimental findings showed significantly higher average survival rate in burned mice immunized with Opr F-LTB than immunized with Opr F or the control group. Rabbits anti Opr F Ig G brought about 75% survival of mice following challenge with P. aeruginosa. Post challenge hepatic and splenic tissues of mice group immunized with Opr F-LTB had significantly lower bacterial load than those immunized with Opr F or the control groups. CONCLUSION: These results demonstrate that LTBfused Opr F might be a potential candidate protein for a prophylactic measure against P. aeruginosa in burn infection.

Adjuvant role of Pseudomonas flagellin for Acinetobacter baumannii biofilm associated protein

AIM To study immunogenicity of Pseudomonas N terminal flagellin as an adjuvant for Acinetobacter baumannii(A. baumanni) biofilm associated protein(Bap).METHODS The N terminal flagellin gene was amplified. The p ET28a(+) and polymerase chain reaction products weredigested with HindⅢ and Eco R Ⅰ. The ligation of N terminal flagellin into p ET28a(?+) was performed using T4 DNA ligase and was then transformed into Escherichia coli BL21(DE3) as a suitable expression host. p ET28a(?+) vector harboring a conserved region of Bap from our previous work was used. The recombinant proteins were expressed, analyzed by SDS-PAGE method and was purified by affinity chromatography with His-Tag residues followed by confirmation with western blotting. Mice were immunized with recombinant N terminal flagellin and Bap subunits. The immunized animals were intranasally(i.n) challenged with A. baumannii and Pseudomonas aeruginosa(P. aeruginosa).RESULTS The flagellin enhanced the immunogenicity of Bap causing an increase in specific Ig G titers in serum(P < 0.001). Internal organs, i.e., liver, lung and spleen of the BapFlagellin immunized group challenged with A. baumannii showed significantly lower bacterial load compared to the control group. The bacterial loads were studied in internal organs. A. baumannii infected immunized animals with Bap-Flagellin exhibited internal organs with minor bacterial load while P. aeruginosa PAO1 infected group showed heavy bacterial load of(4.3 ± 0.12) × 106,(1.1 ± 0.01) × 106 and(2.2 ± 0.22) × 106 per gram of lungs, liver and spleen respectively. Bacterial loads were detected per gram of lungs, liver and spleen of the mice group immunized with Bap were(1.2 ± 0.06) × 107,(11.1 ± 0.041) × 105 and(3.6 ± 0.42) × 106 respectively. In vivo neutralization assay indicated that all experimental mice groups, except for Flagellin administered group was significantly(P < 0.05) protected against A. baumannii. 目的帕金森病(PD)、帕金森综合征(PS)和特发性震颤(ET)是不同类型的运动障碍疾病, 但有部分相同的症状, 因此较难被鉴别诊断。该研究旨在探讨99Tcm-TRODAT-1显像用于区分PD与ET及其他PS疾病的价值。方法 75例患者纳入该研究, 包括29例PD、6例疑似PD、22例ET和18例PS患者。99Tcm-TRODAT-1给药后, 应用双探头SPECT/CT仪行基底神经节(BG)显像。BG摄取值被标准化为全脑和枕叶的活性。所有患者确诊随访2～22个月。结果相比于其他亚型, ET患者和药物诱导的PS患者标准化BG摄取明显增高;但PD和PS患者之间无显著差异。区分相关疾病的患者是否与BG功能障碍有关的灵敏度与特异性分别为80%和83.3%。将当前显性症状侧的壳核和(或)尾状核位点摄取的不对称和不均匀作为附加衡量指标, 可以获得的阳性预测值为82.6%。结论 99Tcm-TRODAT-1显像可用于区分PD或PS与ET。通过组合扫描模式, 包括显性症状侧BG的摄取不对称及尾状核和壳核的不均匀, 可能对PD和其他PS的鉴别诊断有价值。
