慢性鼻窦炎(CRS)的发病机制复杂,呈高度异质性。随着微生物组学与人体疾病研究的不断进展,微生物组参与CRS的发病受到越来越多的关注。现有研究认为,微生物组在CRS发病微环境中失平衡,进而影响鼻腔鼻窦免疫平衡、黏膜屏障及淋巴细胞分化,参与CRS的发生发展。进一步揭示微生物组与CRS发病机制的关系,将有利于为CRS的疾病预防、病情评估和临床治疗等提供新的视角。本文对近年来微生物组学与CRS发生和发展关系的相关研究进行综述。

通过添加不同比例乳清分离蛋白(WPI)制备烘焙预拌粉,研究WPI对预拌粉糊化和流变性质的影响。并进一步将预拌粉制成马芬蛋糕和曲奇饼干,研究其消化、质构及感官性质。结果表明,WPI的添加降低了烘焙预拌粉的峰值黏度、最终黏度、回生值和衰减值。随着WPI的增加,最终黏度从1038.00 cP降低至417.67 cP,回生值从495.00 cP降低至230.67 cP。随着WPI的添加,预拌粉凝胶的储能模量(G)和损耗模量(G)增加,并使预拌粉的黏性特征增加程度更高。添加WPI烘焙成品中快消化淀粉(RDS)含量显著降低,而抗性淀粉(RS)含量显著上升,WPI的加入抑制了淀粉的消化。并且,WPI对马芬蛋糕消化性的抑制作用高于曲奇饼干。随着WPI的添加,马芬蛋糕的硬度增大,而曲奇饼干则变得更脆。

当前教育数字化转型的推进已经进入深水区,《义务教育信息科技课程标准(2022年版)》的出台肩负着促进学生数字素养提升,助力教育数字化转型的历史使命。文章深入分析教育数字化转型“转”什么的问题,挖掘了教育数字化转型的三个典型层面:制度层面的创新、物质层面的升级以及人的素养的提升。进一步地探讨教育数字化转型怎么“转”的问题,构建了教育数字化转型的系统运行模型。在此基础上,分析了信息科技课程在教育数字化转型系统中的制度耦合价值、物质统整价值、素养发展价值。随后,在教育数字化转型系统中,审慎分析并指出当前信息科技课程的实施困境:在制度层面,政策理解与师资队伍发展不足;在物质层面,数字化软硬件设施与资源建设不完善;在人的层面,数字化教育实践意识与能力薄弱。最后,针对当前落实信息科技课程的困境,提出了针对性的实践策略:强化培训深度与广度,建立完善的信息科技教师培育机制;加快新教材建设步伐,完善课程内容和相关资源体系;重塑信息科技教育价值观,变革教学模式与方法;重视人机协同的教学实践,促进人机融合创新发展;落实“教—学—评”一致性,关注师生数字素养评价。

7075铝合金多场耦合流变行为和软化机理

在应变速率为0.01~10 s^(-1),电流密度为18~27 A·mm^(-2),温度为150~330℃条件下,对7075铝合金进行了脉冲电流辅助拉伸实验。探讨了应变速率与电流密度对7075铝合金力学性能的影响,推导出耦合了电参数的修正Johnson-Cook本构模型,并进行了验证。为解析热、电与力的耦合关系提供了新的研究思路。结果表明,与室温相比,通电试样的断面形貌中出现尺寸更大、分布更规整的韧窝。应变速率0.1~1 s^(-1)和电流密度21~24 A·mm^(-2)对降低7075铝合金的流动应力有着显著影响,说明电流密度的影响存在阈值。并且修正后的Johnson-Cook模型在预测上展现出了较高的可靠性,模型预测与试验数据高度吻合,相关性达到0.9961,平均绝对相对误差仅为3.35%。

Alzheimer's disease （AD） has become an important public health issue worldwide. P-Coumaric acid （CA） and lactoferrin （Lf） possessed antioxidant， anti-inflammatory， and anti-AD activities. Herein， we hypothesized that a combination treatment of CA and Lf would synergistically improve AD symptoms and studied its mechanisms. Twelve-week-old APP/PS1 mice were treated with CA， Lf， or both CA and Lf for 8 weeks. Results showed that individual and combined treatments could ameliorate cognitive deficits to varying degrees， with stronger effects for combined intervention than for CA or Lf alone. Specifically， combined treatment was most effective in improving nesting ability and reducing Aβ1-42deposition in mice. Moreover， combined treatment was more effective in suppressing APP， BACE1， and inflammation; it inhibited the CCAAT-enhancer-binding protein （C/EBPβ）/asparagine endopeptidase （AEP） signaling pathway in the hippocampus and colon and upregulated the expression of tight junction protein ZO-1 in the colon; it also decreased the F/B ratio at the phylum level and increased the relative abundance of Prevotellaceae， Lachnospira， and Eubacterium at the genus level. Overall， the combination of CA and Lf may ameliorate cognitive deficits in APP/PS1 mice by improving inflammation and inhibiting the C/EBPβ/AEP signaling pathway via modulating gut microbiome.

Bietti crystalline corneoretinal dystrophy is an inherited retinal disease caused by mutations in CYP4V2,which results in blindness in the working-age population,and there is currently no available ***,we report the results of the first-in-human clinical trial(NCT04722107)of gene therapy for Bietti crystalline corneoretinal dystrophy,including 12 participants who were followed up for 180-365 *** open-label,single-arm exploratory trial aimed to assess the safety and efficacy of a recombinant adeno-associated-virus-serotype-2/8 vector encoding the human CYP4V2 protein(rAAV2/8-hCYP4V2).Participants received a single unilateral subretinal injection of 7.5×10^(10)vector genomes of rAAV2/***,73 treatment-emergent adverse events were reported,with the majority(98.6%)being of mild or moderate intensity and considered to be procedure-or corticosteroid-related;no treatment-related serious adverse events or local/systemic immune toxicities were *** with that measured at baseline,77.8%of the treated eyes showed improvement in best-corrected visual acuity(BCVA)on day 180,with a mean±standard deviation increase of 9.0±10.8 letters in the 9 eyes analyzed(p=0.021).By day 365,80%of the treated eyes showed an increase in BCVA,with a mean increase of 11.0±10.6 letters in the 5 eyes assessed(p=0.125).Importantly,the patients’improvement observed using multifocal electroretinogram,microperimetry,and Visual Function Questionnaire-25 further supported the beneficial effects of the *** conclude that the favorable safety profile and visual improvements identified in this trial encourage the continued development of rAAV2/8-hCYP4V2(named ZVS101e).

Strategies to improve T cell therapy efficacy in solid tumors such as hepatocellular carcinoma(HCC)are urgently *** common cytokine receptorγchain(γc)family cytokines such as IL-2,IL-7,IL-15 and IL-21 play fundamental roles in T cell development,differentiation and effector *** study aims to determine the combination effects of IL-21 in T cell therapy against HCC and investigate optimized strategies to utilize the effect of IL-21 signal in T cell *** antitumor function of AFP-specific T cell receptor-engineered T cells(TCR-T)was augmented by exogenous IL-21 in vitro and in ***-21 enhanced proliferation capacity,promoted memory differentiation,downregulated PD-1 expression and alleviated apoptosis in TCR-T after activation.A novel engineered IL-21 receptor was established,and TCR-T armed with the novel engineered IL-21 receptors(IL-21R-TCR-T)showed upregulated phosphorylated STAT3 expression without exogenous IL-21 ***-21R-TCR-T showed better proliferation upon activation and superior antitumor function in vitro and in ***-21R-TCR-T exhibited a less differentiated,exhausted and apoptotic phenotype than conventional TCR-T upon repetitive tumor antigen *** novel IL-21 receptor in our study programs powerful TCR-T and can avoid side effects induced by IL-21 systemic *** novel IL-21 receptor creates new opportunities for next-generation TCR-T against HCC.
