Background This study aimed to identify survival risk factors in Chinese children with hepatoblastoma(HB)and assess the effectiveness of the new treatment protocol proposed by the Chinese Children's Cancer Group(CCCG)in *** A multicenter,prospective study that included 399 patients with HB from January 2015 to June 2020 was *** demographics,treatment protocols,and other related information were *** regression models and Kaplan-Meier curve methods were *** The 4-year event-free survival(EFS)and overall survival(OS)were 76.9 and 93.5%,*** 4-year EFS rates for the very-low-risk,low-risk,intermediate-risk,and high-risk groups were 100%,91.6%,81.7%,and 51.0%,*** 4-year 0S was 100%,97.3%,94.4%,and 86.8%,*** regression analysis found that age,tumor rupture(R+),and extrahepatic tumor extension(E+)were independent prognostic factors.A total of 299 patients had complete remission,and 19 *** with declining alpha-fetoprotein(AFP)>75%after the first two cycles of neoadjuvant chemotherapy had a better EFS and OS than those≤75%.Conclusions The survival outcome of HB children has dramatically improved since the implementation of CCCG-HB-2016 ***≥8 years,R+,and E+were independent risk factors for *** with a declining AFP>75%after the first two cycles of neoadjuvant chemotherapy had better EFS and OS.

Control of Negative Gravitropism and Tension Wood Formation by Gibberellic Acid and Indole Acetic Acid in Fraxinus mandshurica Rupr. var. japonica Maxim Seedlings

In the present study, we Investigated the role of glbberelllc acid （GA3） and Indole acetic acid （IAA） In the gravity response of stems and tension wood formation using two-year-old stems of Fraxinus mandshurica Rupr. var. Japonica Maxim seedlings. Forty-five seedlings were used and divided Into nine groups that Included five seedlings In each group. Seedlings were treated with applications of GA3 alone at concentrations of 2.89×10^-8 and 2.89×10^-7 μmol/L, IAA alone at concentrations of 5.71×10^-8 and 5.71×10^-7 μmol/L, or their combination to the apical bud of the stem using a mlcroplpette. Seedlings were positioned horizontally after the first treatment. The same treatments were repeated six times per week. At the end of the experiment, all seedlings were harvested. Then, stem segments were cut under a light microscope. Application of exogenous GA3 at the higher concentration stimulated the upward bending of stems, whereas exogenous IAA had no effect. A synergistic effect of GA3 and IAA on upward stem bending was observed following application of the two combinations of GA3 and IAA. Moreover, application of exogenous GA3 at the higher dose stimulated wood formation on both the upper and lower sides of the stems, whereas the mixture of GA3 and IAA had a synerglstic effect on wood formation In horizontal stems. Application of exogenous IAA alone at the lower concentration （5.71×10^-8 μmol/L） or application of a mixture of the higher concentrations of GA3 （2.89×10^-7 μmol/L） and IAA （5.71×10^-7 μmol/L） Inhibited the development of gelatinous fibers （the G-layer） of tension wood on the upper side of the horizontal stems. The differentiation of gelatinous fibers of tension wood was not Inhibited by GA3 when It was applied alone, whereas the development of the gelatinous fibers of tension wood was strongly affected by the application of IAA. Comparative analysis of DNA-SIP and magnetic-nanoparticle mediated isolation (MMI) on unraveling dimethoate degraders

Microorganisms are crucial in the bioremediation of organophosphorus pesticides. However, most functional microorganisms (> 99%) are yet to be cultivated. This study applied two cultivation-independent approaches, DNA-SIP and magnetic-nanoparticle mediated isolation (MMI), to identify the functional microorganisms in degrading dimethoate in agricultural soils. MMI identified five dimethoate degraders: Pseudomonas, Bacillus, Ramlibacter, Arthrobacter, and Rhodococcus, whereas DNA-SIP identified three dimethoate degraders: Ramlibacter, Arthrobacter, and Rhodococcus. Also, MMI showed higher resolution than DNA-SIP in identifying functional microorganisms. Two organic phosphohydrolase (OPH) genes: ophC2 and ophB, were involved in dimethoate metabolism, as revealed by DNA-SIP and MMI. The degradation products of dimethoate include omethoate, O,O,S-trimethyl thiophosphorothioate, N-methyl-2-sulfanylacetamide, O,O-diethyl S-hydrogen phosphorodithioate, O,O,O-trimethyl thiophosphate, O,O,S-trimethyl thiophosphorodithioate, and O,O,O-trimethyl phosphoric. This study emphasizes the feasibility of using SIP and MMI to explore the functional dimethoate degraders, expanding our knowledge of microbial resources with cultivation-independent approaches.

Drug-Target Interactions Prediction Based on Signed Heterogeneous Graph Neural Networks

Drug-target interactions(DTIs) prediction plays an important role in the process of drug *** computational methods treat it as a binary prediction problem, determining whether there are connections between drugs and targets while ignoring relational types information. Considering the positive or negative effects of DTIs will facilitate the study on comprehensive mechanisms of multiple drugs on a common target, in this work, we model DTIs on signed heterogeneous networks, through categorizing interaction patterns of DTIs and additionally extracting interactions within drug pairs and target protein pairs. We propose signed heterogeneous graph neural networks(SHGNNs), further put forward an end-to-end framework for signed DTIs prediction, called SHGNN-DTI,which not only adapts to signed bipartite networks, but also could naturally incorporate auxiliary information from drug-drug interactions(DDIs) and protein-protein interactions(PPIs). For the framework, we solve the message passing and aggregation problem on signed DTI networks, and consider different training modes on the whole networks consisting of DTIs, DDIs and PPIs. Experiments are conducted on two datasets extracted from Drug Bank and related databases, under different settings of initial inputs, embedding dimensions and training modes. The prediction results show excellent performance in terms of metric indicators, and the feasibility is further verified by the case study with two drugs on breast cancer.

A Novel Functional Missense Mutation p.T219A in Type Gaucher＇s Disease

Background： Gaucher＇s disease （GD） is an autosomal recessive disorder caused by a deficiency of acid β-glucosidase （glucocerebrosidase [GBA]） that results in the accumulation of glucocerebroside within macrophages. Many mutations have been reported to be associated with this disorder. This study aimed to discover more mutations and provide data for the genetic pattern of the gene, which will help the development of quick and accurate genetic diagnostic tools for this disease. Methods： Genomic DNA was obtained from peripheral blood leukocytes of the patient and Sanger sequencing is used to sequence GBA gene. Sequence alignments of mammalian β-GBA （GCase） and three-dimensional protein structure prediction of the mutation were made. A construct of this mutant and its compound heterozygous counterpart were used to measure GCase in vitro. Results： GCase is relatively conserved at p.T219A. This novel mutation differs from its wild-type in structure. Moreover, it also causes a reduction in GCase enzyme activity. Conclusion： This novel mutation （c.655A〉G, p.T219A） is a pathogenic missense mutation, which contributes to GD.

Regulation of rhizosphere microenvironment by rice husk ash for reducing the accumulation of cadmium and arsenic in rice

It is important to reduce Cd and As content in brown rice in contaminated paddy *** conducted research on the effects of rice husk ash (RHA) on the Cd and As in the rhizosphere microenvironment (soil,porewater,and iron plaque) and measured the Cd,As,and Si content in rice *** main elements in RHA were Si (29.64%) and O (69.17%),which had the maximum adsorption capacity for Cd was 42.49 mg/kg and for As was 18.62 mg/*** pH and available Si content increased,while soil available Cd and As decreased following application of 0.5%–2%*** promote the transformation of Cd to insoluble fraction,while As was transformed from a poorly soluble form to a more active *** reduced Cd content and increased Si content in porewater,and reduced As only at the later rice growth *** increased the amount of iron plaque,thereby decreasing the Cd content in iron plaque,while increased the As content in *** and inorganic As content in brown rice were decreased,to 0.31 mg/kg and 0.18 mg/kg,*** decrease of Cd in brown rice was due to the decrease of Cd mobility in soil,thereby reducing root accumulation,while the decrease of As in brown rice was affected by the transport from roots to ***,RHA can be considered as a safe and efficient in-situ remediation amendment for Cd and As co-contaminated paddy soil.

A phosphine-catalyzed [4+3] annulation between dinucleophilic indole derivatives and Morita−Baylis−Hillman (MBH) carbonates was discovered by using the N1 and N4′/C4′ nucleophilicities of the indole precursors,in which indoles act as four atom synthons. This protocol provides an efficient and facile access to indole-1,2-fused 1,4-diazepinones and azepines in good to high yields in one step,which illustrates potential synthetic utilities in drug discovery.
