Anticancer,antiobesity,and anti-inflammatory activity of Artemisia species invitro

OBJECTIVE： To investigate the anticancer, antiin flammatory, and antiobesity activity of methanol extracts of eight distinct species： Artemisia Stolon ifera （AST）, Artemisia Selengensis （ASE）, Artemisia .la ponica, Artemisia Montana, Artemisia Capillaris （ACA）, Artemisia Sylvatica （ASY）, Artemisio Keiskeana （AKE）, and Artemisia Scoparia （ASC） in vitro. METHODS： Antiproliferative activity was investigat ed in human breast cancer estrogen receptora pos itive T47D and negative HS578T cell lines exposed to the extracts at various concentrations （5200 mgl ml）for24, 48, and 72 h. For evaluating the antiin flammatory activity of the extracts, inhibition of ni trite synthesis was investigated in lipopolysaccha ride （LPS）stimulated cultures of macrophages cells exposed to 10, 50, 100, and 200 mglml for 24 h. The antiobesity activity of the extracts was deter mined as triglyceride content and by a lipolysis as say in differentiated 3T3LI cells exposed to the extracts for 72 h at the same concentrations de scribed above. RESULTS： All extracts showed similar antiprolifera tive activity in a dose and timedependent man ner in HS578T cells. Although extracts at lower con centrations and shorter times stimulated growth of T47D cells, the antiproliferative effects of the extracts on T47D cells at higher concentrations （〉100 mg/ ml） for 72 h were significantly greater than those of HS578T cells. In case of antiinflammatory activi ty, some extracts （AST, ASE, ACA, and AKE） signifi cantly reduced nitric oxide production at higher concentrations in the presence of LPS compared with that in control cells. Antiobesity activity was showed with reducing lipid accumulation signifi cantly （〉50%） at concentrations above 100 mg/ml in most extracts （except AST and ACA）. Additional ly, AKE and ASC increased lipolysis by 11%24% compared with that in the control. CONCLUSION： Artemisia spp. demonstrates poten tial as bioactive food supplements.

Efficacy and safety of newly developed cross-linked dextran gel injection for glans penis augmentation with a novel technique

There is no safe and effective standard method for glans penis augmentation. Furthermore, there has been scant research on glans penis augmentation due to a poor understanding of glans anatomy, technical difficulty, and a lack of suitable substances for augmentation. Cross-linked dextran gel is a newly developed filler for soft-tissue augmentation. We evaluated the efficacy and safety of using a novel technique to inject cross-linked dextran gel for glans penis augmentation during a 24-week follow-up study. This prospective, single-arm, multicenter study enrolled twenty healthy adult men who underwent glans penis augmentation between June and August 2013. Cross-linked dextran gel was injected into the glans penis using a simple and easy technique. The sizes of the glans penis and individual satisfaction were assessed. Any adverse event was also reported. A total of 18 individuals were analyzed; two of them were lost to follow-up. The mean procedure time and injected volume were about 30 rain and 6.6 ＋- 0.9 ml, respectively. The mean surface areas of the glans at baseline and 24 weeks were 20.0 _＋ 3.5 cm2 and 33.6 -＋ 5.4 cm2, respectively, representing a mean increase of 68.7% _＋ 14.0% （P 〈 0.001）. Sixteen individuals （88.9%） were satisfied with the outcomes, and none were dissatisfied. There were no serious adverse events during the study. Cross-linked dextran gel injection for glans penis augmentation was easy and showed a significant augmentative effect on the glans penis, good durability, and was well tolerated without serious adverse events. Therefore, cross-linked dextran gel injection may be an effective, new technique for glans penis augmentation.

In this study,we investigated the preventive effects of emamectin benzoate 9.7%SL,which was newly developed to reduce the injection volume and number of injection holes required to protect against pine wood *** of the Pinus thunbergii trees injected with emamectin benzoate 9.7%SL at 0.3 ml/cm diameter at breast height(DBH)died within 2 years of inoculation with pine wood *** benzoate 9.7%SL injected at 0.6 ml/cm DBH resulted in no tree mortality for 3 *** residue of emamectin benzoate 9.7%SL in pine twigs injected with 0.3 ml/cm DBH was 0.490μg/g at 1 year after injection and 0.303μg/g after 2 *** residues values are greater than 0.031μg/g,previously determined IC95 value for emamectin benzoate against the pine wood *** field experiment and residue analysis showed that emamectin benzoate 9.7%SL could be a substitute agent for emamectin benzoate 2.15%EC,which is widely used to prevent pine wood nematode in the field and that injection volume and number of injection holes can be greatly educed using this new formulation,which will reduce injury to the cambium,interruption of water movement,and infection of inoculation wounds by wood-decay or blue stain fungi.

Spider angiomas in patients with liver cirrhosis: Role of vascular endothelial growth factor and basic fibroblast growth factor

AIM: To investigate whether vascular endothelial growth factor (VEGF) and basic fibroblastic growth factor (bFGF) are associated wibh spider angiomas in patients wibh liver cirrhosis. METHODS: Eighty-six patients with liver cirrhosis were enrolled and the number and size of the spider angiomas were recorded. Fifty-three healthy subjects were selected as controls. Plasma levels of VEGF and bFGF were measured in both the cirrhotics and the controls. RESULTS: Plasma VEGF and bFGF were increased in cirrhotics compared with controls (122±13 vs. 71±11 pg/ml, P=0.003 for VEGF; 5.1±0.5 vs. 3.4±0.5 pg/ml, P=0.022 for bFGF). In cirrhotics, plasma VEGF and bFGF were also higher in patients with spider angiomas compared with patients without spider angiomas (185±28 vs. 90±10 pg/ml, P=-0.003 for VEGF; 6.8±1.0 vs. 4.1±0.5 pg/ml, P=0.017 for bFGF). Multivariate logistic regression showed that young age and increased plasma levels of VEGF and bFGF were the most significant predictors for the presence of spider angiomas in cirrhotic patients (odds ratio [OR]=6.64, 95% confidence interval[CI]=2.02-21.79, P=0.002; OR=4.35, 95% CI=1.35-14.01,P=0.014; OR=5.66, 95% CI=1.72-18.63, P=0.004, respectively). CONCLUSION: Plasma VEGF and bFGF are elevated inpatients with liver cirrhosis. Age as well as plasma levels of VEGF and bFGF are significant predictors for spider angiomas in cirrhotic patients.

Effect of extracts from Stachys sieboldii Miq. on cellular reactive oxygen species and glutathione production and genomic DNA oxidation

Objective: To evaluate the antioxidant activity of extracts and fractions from Stachys sieboldii Miq., and to examine its effect on the cellular reactive oxygen species(ROS) and glutathione(GSH) production and genomic DNA oxidation in HT-1080 cells. Methods: The ROS generation induced by H2 O2 was measured by the dichlorofluorescein-diacetate assay. GSH levels were measured using a fluorescent method with mBBr. Genomic DNA oxidative damage was measured with levels of oxidative DNA induced by the reaction of ferritin with H2 O2. Results: The n-hexane, 85% aqueous methanol and n-butanol fractions(0.05 mg/ml concentrations) inhibited H2 O2-induced ROS generation by 63%, 35% and 45%, respectively. GSH levels were significantly increased in both acetone+methylene chloride and methanol extracts(Pml(P<0.05). Both the acetone+methylene chloride and methanol extracts, as well as all fractions significantly inhibited oxidative DNA damage(P<0.05). Conclusions: These results indicate that cellular oxidation was inhibited by the n-hexane fraction and this fraction may contain valuable active compounds.

Giant choledocholithiasis treated by mechanical lithotripsy using a gastric bezoar basket

Mechanical lithotripsy （ml） is usually considered as a standard treatment option for large bile duct stones. However, it is impossible to retrieve oversized stones because the conventional lithotripsy basket may not be able to grasp the stone. However, there is no es- tablished endoscopic extraction method for such gi- ant stone removal. We describe a case of successful extraction of a 4-cm large stone using a gastric bezoar basket. A 78-year-old woman had suffered from upper abdominal pain for 20 d. Contrast-enhanced computed tomogram revealed a 4-cm single stone in the distal common bile duct （CBD）. Endoscopic stone retraction was decided upon and endoscopic papillary balloon dilation was performed using a large balloon. An at- tempt to capture the stone using a standard lithotripsy basket failed due to the large stone size. Subsequently, we used a gastric bezoar basket to successfully capturethe stone. The stone was fragmented into small pieces and extracted. The stone was completely removed after two sessions of endoscopic retrograde cholangio- pancreatography; each of which took 30 rain. No com- plications occurred during or after the procedure. The patient was fully recovered and discharged on day 11 of hospitalization. ml using a gastric bezoar basket is a safe and effective retrieval method in select cases, and is considered as an alternative nonoperative option for the management of difficult CBD stones.

Recombinant human fibroblast growth factor-2 promotes nerve regeneration and functional recovery after mental nerve crush injury

Several studies have shown that fibroblast growth factor-2 （FGF2） can directly affect axon regeneration after peripheral nerve damage. In this study, we performed sensory tests and histological analyses to study the effect of recombinant human FGF-2 （rhFGF2） treatment on damaged mental nerves. The mental nerves of 6-week-old male Sprague-Dawley rats were crush-injured for 1 minute and then treated with 10 or 50 μg/ml rhFGF2 or PBS in crush injury area with a mini Osmotic pump. Sensory test using von Frey filaments at 1 week revealed the presence of sensory degeneration based on decreased gap score and increased difference score. However, at 2 weeks, the gap score and difference score were significantly rebounded in the mental nerve crush group treated with 10 μg/ml rhFGF2. Interestingly, treatment with 10 μg/ml rhFGF had a more obviously positive effect on the gap score than treatment with 50 μg/ml rhFGF2. In addition, retrograde neuronal tracing with Dil revealed a significant increase in nerve regeneration in the trigeminal ganglion at 2 and 4 weeks in the rhFGF2 groups （10 μg/ml and 50 μg/ml） than in the PBS group. The 10 μg/ml rhFGF2 group also showed an obviously robust regeneration in axon density in the mental nerve at 4 weeks. Our results demonstrate that 10 μg/ml rhFGF induces mental nerve regeneration and sensory recovery after mental nerve crush injury.

Hepatoprotective effect of manual acupuncture at acupoint GB34 against CCl_4-induced chronic liver damage in rats

AIM： To investigate the hepatoprotective effect of manual acupuncture at Yanglingquan （GB34） on CCl4-induced chronic liver damage in rats. METHODS： Rats were injected intraperitoneally with CCh （1 ml/kg） and treated with manual acupuncture using reinforcing manipulation techniques at left GB34 （Yanglingquan） 3 times a week for 10 wk. A nonacupoint in left gluteal area was selected as a sham point. To estimate the hepatoprotective effect of manual acupuncture at GB34, measurement of liver index, biochemical assays including serum ALT, AST, ALP and total cholesterol, histological analysis and blood cell counts were conducted. RESULTS： Manual acupuncture at GB34 reduced the liver index, serum ALT, AST, ALP and total cholesterol levels as compared with the control group and the sham acupuncture group. It also increased and normalized the populations of WBC and lymphocytes. CONCLUSION： Manual acupuncture with reinforcing manipulation techniques at left GB34 reduces liver toxicity, protects liver function and liver tissue, and normalizes immune activity in CCh-intoxicated rats.
