BACKGROUND the trans-fat containing AMLN(amylin liver non-alcoholic steatohepatitis,NASH)diet has been extensively validated in C57BL/6J mice with or without the Lep^ob/Lep^ob(ob/ob)mutation in the leptin gene for reliably inducing metabolic and liver histopathological changes recapitulating hallmarks of *** to a recent ban on trans-fats as food additive,there is a marked need for developing a new diet capable of promoting a compatible level of disease in ob/ob and C57BL/6J *** to develop a biopsy-confirmed mouse model of NASH based on an obesogenic diet with trans-fat substituted by saturated *** Male ob/ob mice were fed AMLN diet or a modified AMLN diet with trans-fat(Primex shortening)substituted by equivalent amounts of palm oil[Gubra amylin NASH,(GAN)diet]for 8,12 and 16 wk.C57BL/6J mice were fed the same diets for 28 *** and GAN diets had similar caloric content(40%fat kcal),fructose(22%)and cholesterol(2%)*** the GAN diet was more obesogenic compared to the AMLN diet and impaired glucose ***-confirmed steatosis,lobular inflammation,hepatocyte ballooning,fibrotic liver lesions and hepatic transcriptome changes were similar in ob/ob mice fed the GAN or AMLN diet.C57BL/6J mice developed a mild to moderate fibrotic NASH phenotype when fed the same *** Substitution of Primex with palm oil promotes a similar phenotype of biopsyconfirmed NASH in ob/ob and C57BL/6J mice,making GAN diet-induced obese mouse models suitable for characterizing novel NASH treatments.

Prediction and Validation of Promoters Involved in the Abscisic Acid Response in Physcomitrella patens

Detection of cis-regulatory elements, such as transcription factor binding sites （tFBS）, through utilization of ortholog conservation is possible only if genomic data from closely related organisms are available. An alternative approach is the detection of tFBS based on their overrepresentation in promoters of co-regulated genes. However, this approach usually suffers from a high rate of false-positive prediction. Here, we have conducted a case study using promoters of genes known to be strongly induced by the phytohormone abscisic acid （ABA） in the model plant Physcomitrella patens, a moss. Putative tFBS were detected using three de novo motif detection tools in a strict consensus approach. the resulting motifs were validated using data from microarray expression profiling and were able to predict ABA-induced genes with high specificity （90.48%） at mediocre sensitivity （33.33%）. In addition, 27 genes predicted to contain ABA-responsive tFBS were validated using real-time PCR. Here, a total of 37% of the genes could be shown to be induced upon ABA treatment, while 70% were found to be regulated by ABA. We conclude that the consensus approach for motif detection using coregulation information can be used to identify genes that are regulated under a given stimulus. In terms of evolution, we find that the ABA response has apparently been conserved since the first land plants on the level of families involved in transcriptional regulation.

In the present paper the edge stabilization technique is applied to a convection-diffusion problem with exponential boundary layers on the unit square, using a Shishkin mesh with bilinear finite elements in the layer regions and linear elements on the coarse part of the mesh. An error bound is proved for ‖πu-u^h‖Е, where πu is some interpolant of the solution u and uh the discrete solution. this supercloseness result implies an optimal error estimate with respect to the L2 norm and opens the door to the application of postprocessing for improving the discrete solution.

A new recovery operator P ：Qn^disc（t）→Qn＋1^disc（M） for discontinuous Galerkin is derived. It is based on the idea of projecting a discontinuous, piecewise polynomial solution on a given mesh t into a higher order polynomial space on a macro mesh M. In order to do so, we define local degrees of freedom using polynomial moments and provide global degrees of freedom on the macro mesh. We prove consistency with respect to the local L2-projection, stability results in several norms and optimal anisotropic error estimates. As an example, we apply this new recovery technique to a stabilized solution of a singularly perturbed convection-diffusion problem using bilinear elements.

Surgical strategies in the therapy of non-small cell lung cancer

Lung cancer represents the leading cause of cancer mortality worldwide. Despite improvements in preoperative staging, surgical techniques, neoadjuvant/adjuvant options and postoperative care, there are still major difficulties in significantly improving survival, especially in locally advanced non-small cell lung cancer(NSCLC). to date, surgical resection is the primary mode of treatment for stage?Ⅰ?and Ⅱ NSCLC and has become an important component of the multimodality therapy of even more advanced disease with a curative intention. In fact, in NSCLC patients with solitary distant metastases, surgical interventions have been discussed in the last years. Accordingly, this review displays the recent surgical strategies implemented in the therapy of NSCLC patients.

Effect of Different Calcination temperatures and Post Annealing on the Properties of Acetic Acid Based Sol-Gel (Na_(0.5)K_(0.5))NbO_3(NKN) thin Films

（Na0.5K0.5）NbO3 （NKN） lead free thin films were synthesized by means of an acetic acid based sol-gel process on Pt/ti/SiO2/Si substrates. Na-acetate, K-acetate and Nb-pentaethoxide were used as metal precursors and acetic acid as the solvent. the effect of different calcination temperatures on the properties of the NKN films was investigated by X-ray diffraction, scanning electron microscopy, leakage current and hysteresis measurements. Low calcination temperatures led to low currents at high electric fields whereas high calcination temperatures led to low currents at low electric fields. Based on these findings calcination at low temperature was combined with a post annealing treatment. Low leakage currents of 4×10^-4 A/cm2 at 150 kV/cm and 2Pr and 2Ec values of 28 μC/cm2 and 150 kV/cm, respectively, could be obtained. All films were single phase NKN with random crystal orientations and no crack or pore formation was visible on the surface.

Synthesis and Characterization of(Na_(0.5)K_(0.5))NbO_3(NKN) thin Films Formed by a Diol-based Sol-gel Process

Lead-free （***0.5）Nb03 （NKN） thin films were fabricated by spin coating on Pt/ti/SiO2/Si substrates by a diol-based sol-gel process. Na-acetate, K-acetate, Nb-pentaethoxide and 1,3 propanediol were used to prepare the NKN precursor solution. thermal analysis showed two characteristic temperatures of 360 and 600℃. Based on these temperatures, a heat treatment program with pyrolysis at 360℃ and calcination at 600℃ after every layer was used. to avoid inhomogeneities and secondary phases, an excess of sodium and potassium was necessary. to evaluate the proper excess amount of sodium and potassium secondary ion mass spectrometry （SIMS） lateral element maps and X-ray diffraction （XRD） patterns were recorded. An excess amount of 20% led to homogeneous distribution of the elements and to single phase perovskite NKN films with random crystal orientation. Scanning electron microscopy （SEMI images showed a pore free surface with 100 nm grains. the leakage current measurements showed a current of lx10-3 A/cm2 at 150 kV/cm.
