Imatinib-induced fatal acute liver failure

Imatinib mesylate is a drug that has been approved for treatment of chronic myeloid leukemia (CML) in blast crisis, accelerated or chronic phase, and also for advanced gastrointestinal stromal tumors. Severe hepatic toxicity and three deaths from hepatic failure have been reported. We report the case of a 51-year-old woman who was admitted to our institution with severe acute hepatitis. She was diagnosed with CML and began treatment with imatinib mesylate at a dose of 400 mg/d. Five months after beginning treatment, she developed severe hepatitis associated with coagulopathy, and was admitted to our institution. She had been consuming acetaminophen 500-1000 mg/d after the onset of symptoms. She had a progressive increase in bilirubin level and a marked decrease of clotting factor Ⅴ. Five days after admission, grade Ⅱ encephalopathy developed and she was referred for liver transplantation. Her clinical condition progressively deteriorated, and 48 h after being referred for transplantation she suffered a cardiac arrest and died. This report adds concern about the possibility of imatinib-mesylate-induced hepatotoxicity and liver failure, particularly in the case of concomitant use with acetaminophen. Liver function tests should be carefully monitored during treatment and, with the appearance of any elevation of liver function tests, treatment should be discontinued.

A collection of 368 advanced lines and cultivars of spring wheat（Triticum aestivum L.） from Chile, Uruguay, and CIMMYT（Centro Internacional de Mejoramiento de Maíz y Trigo）, with good agronomic characteristics were evaluated under the Mediterranean conditions of central Chile. Three different water regimes were assayed： severe water stress（SWS, rain fed）, mild water stress（MWS; one irrigation around booting）, and full irrigation（FI; four irrigations： at tillering,flag leaf appearance, heading, and middle grain filling）. Traits evaluated were grain yield（GY）, agronomical yield components,days from sowing to heading, carbon isotope discrimination（△^13C） in kernels, and canopy spectral reflectance. Correlation analyses were performed for 70 spectral reflectance indices（SRI） and the other traits evaluated in the three trials. GY and △^13C were the traits best correlated with SRI, particularly when these indices were measured during grain filling. However,only GY could be predicted using a single regression, with Researchnormalized Difference Moisture Index（nDMI2： 2,200; 1,100）having the best fit to the data for the three trials. For △^13C, only individual regressions could be forecast under FI（r^2： 0.25–0.37）and MWS（r^2： 0.45–0.59） but not under SWS（r^2： 0.03–0.09）.nIR‐based SRI proved to be better predictors than those that combine visible and nIR wavelengths.

Wereport on the colloidal and rheological characterization of suspensions of commercial single-walled carbon nanotubes(SWCnT)and oxidized single walled carbon nanotubes(SWCnT-oxi24h),in water and fetal bovine serum(FBS).The effects of two surfactants,dodecyl-benzene sodium sulfonate(SBDS)and sodium dodecyl sulfate(SDS)at different concentrations(0.5-2.0%)on the zeta potential and the rheology of the suspensions is also *** suspensions were characterized by Transmission Electron Microscopy(TEM)and Atomic Force Microscopy(AFM).The colloidal stability was determined by measurements of zeta potential in the physiological pH range 6.0nts were implemented at concentrations of 1 and 1.5 mg/ml,and temperatures of 25℃,30℃,and 37℃.The results obtained show that SWCnT suspensions in FBS,with and without addition of surfactants,depict a high tendency to aggregate with the bovine serum albumin(BSA),a processwe could further characterize using Molecular Dynamics(MD)*** suspensions in aqueous media show a negative zeta potential between−60 and−80 mV,whereas in FBS the stability is lower,with values of-20 *** observed no significant changes in the zeta potential with the different *** viscosity values obtained are smaller in water than in FBS,demonstrating the higher stability in water.

Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tnGCs). However, the use of tnGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tnGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tnGCs and compared the various features of these improved silk^(+) tnGCs with the tubes without holes(silk^(–) tnGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tnGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tnGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tnGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tnGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. ß-Glucosidases(ßGL)-widely used in the enological field-are enzymes that catalyze the liberation of aromatic volatiles from their glycosidic precursors during *** the present study,aßGL obtained from the Antarctic yeast Mrakia *** 7.1.2016 was produced on a bioreactor scale,purified,characterized,and the enzyme’s properties studied for a potential enological ***-dodecylsulfate-polyacrylamide-gel electrophoresis indicated a high molecular weight for this enzyme,it having at least two subunits of 134 and 14 kDa.ßGL exhibited a pH optimum of 5.0 and retained substantial activity and stability at pH *** temperature range for optimal activity was 50-55℃,with the enzyme demonstrating thermostability up to 50℃ and retaining 87%of residual activity at that temperature after 3 *** respective kinetic constants determined with p-nitrophenyl-ß-D-glucopyranoside and cellobiose were 0.38 and 1.79 mM for Km and 20.1 and 5.65μmol^(-1)mg^(-1)min^(.1) for Vmax,ß-Glucosidase manifested high activity in 10-25%(v/v)ethanol,30.0 mg L^(-1)sulfur dioxide,and 10-200 g L^(-1)fructose;but it was strongly inhibited by glucose,retaining only 6%of residual activity in the presence of 20 g L^(-1).Upon investigating the influence of the enzyme on Muscat-wine glycosidic precursors,we found significant differences in terpene content after 14 days ofßGL treatment at an eightfold increase over control-wine *** enzyme was more active toward the precursors of the monoterpenes nerol and geraniol and oxides of trans-and *** findings contribute to our understanding of the potential of cold-active enzymes in advantageous biotechnological applications to enology.

The quality demanded by consumers in the cocoa sector has led to carrying out the postharvest transformation of cocoa,directing efforts to highlight properties of cocoa beans,such as the content of bioactive compounds,due to its beneficial effects on health,as well as its color,which varies from the typical brown color to the recently developed red-pink *** work aims to establish a correlation between the color parameters of a cocoa matrix obtained during the transformation of cocoa seeds under controlled process conditions(temperature,organic acid,and its concentration,and time)and the flavan-3-ols content(catechin,epicatechin,and epigallocatechin),methylxanthines content(theobromine,caffeine),and total phenolic content(TPC)through image analysis of *** results showed that the content of flavan-3-ols and methylxanthines decreased as the controlled transformation ***,the content of these bioactive compounds is retained by applying mild temperature conditions(30℃)and low concentrations of organic acid(1 g/L)to transform cocoa ***,the color parameters respond to changes in the internal pH of the seeds,and the red-pink hues are favored when using high concentrations of organic acid(30 g/L).Based on these findings,it was possible to establish a correlation between epigallocatechin content and four color parameters(a^(∗),b^(∗),h_(ab)^(∗) and C_(ab)^(∗))through a multiple linear regression *** this way,color image analysis could be an appropriate alternative to predict the concentration of quality-related compounds in cocoa matrices.
