Optimal concentration of mesenchymal stem cells for fracture healing in a rat model with long bone fracture

BACKGROUND There is still no consensus on which concentration of mesenchymal stem cells(MSCs)to use for promoting fracture healing in a rat model of long bone *** To assess the optimal concentration of MSCs for promoting fracture healing in a rat *** Wistar rats were divided into four groups according to MSC concentrations:Normal saline(C),2.5×10^(6)(L),5.0×10^(6)(M),and 10.0×10^(6)(H)*** MSCs were injected directly into the fracture *** rats were sacrificed at 2 and 6 wk *** bone formation[bone volume(BV)and percentage BV(PBV)]was evaluated using micro-computed tomography(CT).Histological analysis was performed to evaluate fracture healing *** protein expression of factors related to MSC migration[stromal cell-derived factor 1(SDF-1),transforming growth factor-beta 1(TGF-β1)]and angiogenesis[vascular endothelial growth factor(VEGF)]was evaluated using western blot *** expression of cytokines associated with osteogenesis[bone morphogenetic protein-2(BMP-2),TGF-β1 and VEGF]was evaluated using real-time polymerase chain *** Micro-CT showed that BV and PBV was significantly increased in groups M and H compared to that in group C at 6 wk post-fracture(P=0.040,P=0.009;P=0.004,P=0.001,respectively).Significantly more cartilaginous tissue and immature bone were formed in groups M and H than in group C at 2 and 6 wk post-fracture(P=0.018,P=0.010;P=0.032,P=0.050,respectively).At 2 wk post fracture,SDF-1,TGF-β1 and VEGF expression were significantly higher in groups M and H than in group L(P=0.031,P=0.014;P<0.001,P<0.001;P=0.025,P<0.001,respectively).BMP-2 and VEGF expression were significantly higher in groups M and H than in group C at 6 wk postfracture(P=0.037,P=0.038;P=0.021,P=0.010).Compared to group L,TGF-β1 expression was significantly higher in groups H(P=0.016).There were no significant differences in expression levels of chemokines related to MSC migration,angiogenesis and cytokines associated

Exploring anharmonic lattice dynamics and dielectric relations in niobate perovskites from first-principles self-consistent phonon calculations

Group I niobates(KNbO_(3) and NaNbO_(3))are promising lead-free alternatives for high-performance energy storage *** their potential,their complex phase transitions arising from temperature-dependent phonon softening and anharmonic effects on dielectric properties remain poorly *** this study,we employ density-functional theory(DFT)and self-consistent phonon(SCP)calculations to investigate finite-temperature phonons in cubic niobate *** include explicit anharmonic vibrational effects,SCP frequencies are shifted by the bubble self-energy correction within the quasiparticle(QP)approximation,providing precise descriptions of phonon softening in these strongly anharmonic *** further calculate the static dielectric constant of KNbO_(3) and NaNbO_(3) as a function of temperature using the Lyddane-Sachs-Teller(LST)relation and QP-corrected phonon *** theoretical results align with experimental data,offering reliable temperature-dependent phonon dispersions while considering anharmonic self-energies and thermal expansion effects,enhancing our understanding of the complex relations between lattice vibrations and phase transitions in these anharmonic oxides.

Estimation of marine winds in and around typhoons using multi-platform satellite observations: Application to Typhoon Soulik (2018)

Estimating horizontal winds in and around typhoons is important for improved monitoring and prediction of typhoons and mitigating their ***,we present a new algorithm for estimating typhoon winds using multiple satellite observations and its application to Typhoon Soulik(2018).Four kinds of satellite remote sensing data,along with their relationship to typhoon intensity,derived statistically from hundreds of historical typhoon cases,were merged into the final product of typhoon wind(MT wind):(1)geostationarysatellite-based infrared images(IR wind),(2)passive microwave sounder(MW wind),(3)feature-tracked atmospheric motion vectors,and (4)scatterometer-based sea surface winds(SSWs).The algorithm was applied to two cases(A and B)of Typhoon Soulik and validated against SSWs independently retrieved from active microwave synthetic aperture radar(SAR)and microwave radiometer(AMSR2)images,and vertical profiles of wind speed derived from reanalyzed data and dropsonde *** Case A(open ocean),the algorithm estimated the realistic maximum wind,radius of maximum wind,and radius of 15 m/s,which could not be estimated using the reanalysis data,demonstrating reasonable and practical ***,for Case B(when the typhoon rapidly weakened just before making landfall in the Korean Peninsula),the algorithm significantly overestimated the parameters,primarily due to the overestimation of typhoon *** study highlights that realistic typhoon winds can be monitored continuously in real-time using multiple satellite observations,particularly when typhoon intensity is reasonably well predicted,providing timely analysis results and products of operational importance.

Scherer et *** the influence of de novo pyrimidine nucleotide synthesis on T-cell responses after antigen stimulation,particularly on effector and memory T-cell development[1].Their work focused on the effect of leflunomide(trade name Avara®),which is a direct inhibitor of dihydroorotate dehydrogenase(DHODH;a key enzyme in the de novo pyrimidine nucleotide pathway and located on the inner mitochondrial membrane).In this pathway,DHODH converts dihydroorotate to orotate and has been implicated in the reduction of ubiquinol to ubiquinone mediated by Complex III in the electron transport *** metabolic effects of DHODH inhibition are unclear[2,3].However,it is known to block S-phase progression by activating *** was originally approved for the treatment of rheumatoid arthritis(RA)in 1998;the mechanism involves the induction of S-phase cell cycle arrest and apoptosis,inhibiting cell proliferation and *** studies have demonstrated the therapeutic potential of leflunomide for cancer[4].Despite the efficacy of leflunomide,the mechanism by which it targets immune cells in the treatment of RA was unknown;however,the work of Scherer and colleagues has shed new light on this mechanism.

Brassinosteroids enhance salicylic acid-mediated immune responses by inhibiting BIN2 phosphorylation of clade I TGA transcription factors in Arabidopsis

Salicylic acid (SA) plays an important role in plant immune response, including resistance to pathogens and systemic acquired resistance. Two major components, NONEXPRESSOR OF PATHOGENESIS-RELATED GENES (NPRs) and TGACG motif-binding transcription factors (TGAs), are known to mediate SA signaling, which might also be orchestrated by other hormonal and environmental changes. Nevertheless, the molecular and functional interactions between SA signaling components and other cellular signaling pathways remain poorly understood. Here we showed that the steroid plant hormone brassinosteroid (BR) promotes SA responses by inactivating BR-INseNSITIVE 2 (BIN2), which inhibits the redox-sensitive clade I TGAs in Arabidopsis. We found that both BR and the BIN2 inhibitor bikinin synergistically increase SA-mediated physiological responses, such as resistance to Pst DC3000. Our genetic and biochemical analyses indicated that BIN2 functionally interacts with TGA1 and TGA4, but not with other TGAs. We further demonstrated that BIN2 phosphorylates ser-202 of TGA4, resulting in the suppression of the redox-dependent interaction between TGA4 and NPR1 as well as destabilization of TGA4. Consistently, transgenic Arabidopsis overexpressing TGA4-YFP with a S202A mutation displayed enhanced SA responses compared to the wild-type TGA4-YFP plants. Taken together, these results suggest a novel crosstalk mechanism by which BR signaling coordinates the SA responses mediated by redox-sensitive clade I TGAs.

Dysregulation of the Hippo pathway has been frequently identified in various human cancers. The Hippo pathway is a highly complex pathway involving multiple types of proteins, and the activation of YAP by LATS kinase is the final effector step in the transcription process. In this study, we linked the roles of the multifunctional adapter proteins β-arrestins (β-arrestin 1 and 2) in cooperation with other signaling pathways such as GPCR and Wnt to essential cellular functions involved in carcinogenesis, including the regulation of cell proliferation, migration, and differentiation as well as stem cell properties.1 Although β-arrestin 1 and 2 have high structural similarities, they have different roles in carcinogenesis as β-arrestin 1 aids cancer cell survival and metastasis2 and β-arrestin 2 inhibits tumor growth.1 In the Hippo signaling pathway, β-arrestins function as scaffold proteins that mediate the phosphorylation of key molecules, and their association with human cancers is a major research topic. In this study, we demonstrated that β-arrestin 2 inhibits YAP activation through the formation of the β-arrestin 2-LATS-YAP trimeric complex, which results in the promotion of the kinase activity of LATS in cancer cell lines and patient-derived colon cancer organoids.

Phytogenic feed additives alleviate pathogenic Escherichia coli-induced intestinal damage through improving barrier integrity and inhibiting inflammation in weaned pigs

Variations in microstructure and bending formability of extruded Mg–Al–Zn–Ca–Y–MM alloy with precompression and subsequent annealing treatment conditions

In this study, the bending formability of an extruded Mg–9Al–1Zn–0.3Mn–0.9Ca–0.6Y–0.5 MM(AZXWMM91100, wt%) alloy at room temperature is significantly improved through application of a combined precompression and subsequent annealing(PCA) treatment. As the amount of precompression applied along the extrusion direction(ED)(i.e., the total strain) increases from 4% to 6%, the area fraction of ED-oriented grains of the PCA-treated alloy increases, which consequently causes an improvement in its bending formability because these grains accommodate larger tensile strain along the ED during bending. As the temperature of the subsequent annealing treatment increases from 350 ℃ to 450 ℃, both the area fraction of the ED-oriented grains and the average grain size increase, and the residual dislocation density decreases owing to the promotion of boundary migration and occurrence of the recovery process at higher ***, the bending formability of both the 4%-precompressed and the 6%-precompressed samples increases with an increase in the annealing temperature. However, when the precompressed samples are annealed at 300 ℃, their bending formability is lower than that of the extruded alloy because the dislocations formed by precompression remain even after the subsequent annealing at this temperature.

Microstructural characteristics and low-cycle fatigue properties of AZ91 and AZ91-Ca-Y alloys extruded at different temperatures

The commercial AZ91 alloy and nonflammable seN9(AZ91-0.3Ca-0.2Y,wt%)alloy are extruded at 300°C and 400°*** microstructure,tensile and compressive properties,and low-cycle fatigue(LCF)properties are investigated,with particular focus on the influence of the extrusion *** the AZ91 and seN9 materials extruded at 300°C(300-materials),numerous fine Mg_(17)Al_(12)particles are inhomogeneously distributed owing to localized dynamic precipitation during extrusion,unlike those extruded at 400°C(400-materials).These fine particles suppress the coarsening of recrystallized grains,decreasing the average grain size of *** the four extruded materials have considerably different microstructures,the difference in their tensile yield strengths is insignificant because strong grain-boundary hardening and precipitation hardening effects in 300-materials are offset almost completely by a strong texture hardening effect in ***,owing to their finer grains and weaker texture,300-materials have higher compressive yield strengths *** the LCF tests,{10-12}twinning is activated at lower stresses in 400-materials than in *** the fatigue damage accumulated per cycle is smaller in 400-materials,they have longer fatigue lives than those of 300-materials.A fatigue life prediction model for the investigated materials is established on the basis of the relationship between the total strain energy density(ΔW_(t))and the number of cycles to fatigue failure(N_(f)),and it is expressed through a simple equation(ΔW_(t)=10·N_(f)-0.59).This model enables fatigue life prediction of both the investigated alloys regardless of the extrusion temperature and strain amplitude.

BACKGROUND Cerebrospinal fluid(CSF)leakage at C1/2 in spontaneous intracranial hypotension(SIH)is *** hematoma(SDH),a serious complication of SIH,may lead to neurological *** report presents a case of SDH after spontaneous C1/2 CSF leakage,which was treated with a targeted epidural blood patch(EBP).CAse SUMMARY A 60-year-old man with no history of trauma was admitted to our hospital with orthostatic headache,nausea,and *** computed tomography imaging revealed bilateral,subacute to chronic *** magnetic resonance imaging(MRI)findings were SDH with dural enhancement in the bilateral cerebral convexity and posterior fossa and mild sagging,suggesting *** the patient underwent burr hole trephination,the patient’s orthostatic headache was *** myelography led to a suspicion of CSF leakage at C1/***,we performed a targeted cervical EBP using an epidural catheter under fluoroscopic *** 5 d after EBP,a follow-up MR myelography revealed a decrease in the interval size of the CSF *** his symptoms improved,the patient still complained of headaches;therefore,we repeated the targeted cervical EBP 6 d after the initial ***,his headache had almost disappeared on the 8th day after the repeated *** Targeted EBP is an effective treatment for SDH in patients with SIH due to CSF leakage at C1/2.
