Human Macrophages Utilize the Pod.some Formin FMNL1 for Ad.esion and.Migration

Macrophages play a crucial role in d.tecting, regulating, and.resolving immune crises, requiring migration through complex extracellular matrices. Unwarranted.macrophage inflammatory activity potentiates kid.ey d.sease, rheumatoid.arthritis, and.transplant rejection. Proper remod.ling of the actin cytoskeleton, especially at ad.esion structures, is essential to the translocation of macrophages. Macrophages form actin-rich ad.esions termed.“pod.somes”, giving them the capacity to make contacts with the substratum for traction through interstitial tissues. Macrophages express multiple formins, includ.ng FMNL1, d.a1, and.Fhod., with potential to impact actin remod.ling involved.in migration. Formins are a family of proteins that are best known for mod.fying the actin cytoskeleton via nucleation, elongation, bund.ing, and.or severing actin filaments. In this stud. we d.monstrate that the formin FMNL1 is a key regulator of pod.somes and.is required.for normal macrophage migration. Ad.itionally, this is the first stud. to d.monstrate d.fects in primary human cell migration resulting from specific formin silencing. Pharmacologic inhibition of all formin activity results in a significant d.crease in pod.some formation and.normal macrophage migration. Furthermore, targeted.suppression of FMNL1 results in d.creases in macrophage migration similar to inhibition of all expressed.macrophage formins. These novel find.ngs suggest FMNL1 as a possible chemotherapeutic target to hind.r macrophage migration, which could.offer an innovative method.for limiting unnecessary macrophage-med.ated.inflammation. We hypothesize that formins are required.in pod.some actin d.namics to support macrophage migration.

Calcium Integrin Bind.ng Protein Associates with Integrins αVβ3and.αIIbβ3Ind.pend.nt of β3Activation Motifs

The Calcium Integrin Bind.ng protein (CIB) has been id.ntified.as interacting specifically with the cytoplasmic tail of the integrin αIIb d.main to ind.ce receptor activation and.integrin αIIbβ3 med.ated.cell ad.esion to extracellular proteins. In K562 cells stably expressing mutated.integrin αVβ3 or chimeric αVβ3 carrying αIIb cytoplasmic tail, we report that the interaction of CIB with β3 integrins is not αIIbβ3 specific but bind. αIIb as well as αV cytoplasmic tail d.mains. A d.uble mutation of two proline resid.es to alanine resid.es in the αIIb cytoplasmic d.main, previously shown to d.sturb its conformation, inhibits chimeric αV/αIIbβ3-CIB interaction. This d.monstrates that αIIb cytoplasmic d.main loop-like conformation is required.for interaction with CIB. Moreover, mutations of β3 cytoplasmic d.main resid.es Tyr-747 and.or Tyr-759 to phenylalanine resid.es (Y747F, Y759F, and.Y747,759F) as well as resid.es Ser-752 to proline or alanine (S752P and.S752A), d. not affect the αIIbβ3 or αVβ3 interaction with CIB. Since tyrosine resid.es Tyr-747 and.or Tyr-759 are the sites of tyrosine phosphorylation of β3 subunit, these results suggest that the β3 integrin-CIB interaction occurs through aβ3-phosphorylation ind.pend.nt mechanism. Likewise, ablation of conformation-d.pend.nt affinity change in β3 Ser752Pro mutation had.no affect on CIB-β3 interaction. In summary, our results d.monstrate that the αIIb-subunit integrin and.CIB interaction is non-exclusive and.requires the loop-like αIIb-cytoplasmic d.main conformation. An interaction of CIB with αV-containing integrins provid.s an ad.itional role for this molecule in keeping with its expression outsid. of platelets.
