Enhanced refolding of lysozyme with imidazolium-based room temperature ionic liquids: Effect of hydrophobicity and sulfur residue

The expression of recombinant proteins in microorganism frequently leads to the formation of insoluble aggregates, inclusion bodies (IBs). Thus, the additional in vitro protein refolding process is required to convert inactive IBs into water-soluble active proteins. This study investigated the effect of sulfur residue and hydrophobicity of imidazolium-based room temperature ionic liquids (RTIls) on the refolding of lysozyme as a model protein in the batch dilution method which is the most commonly used refolding method. When lysozyme was refolded in the refolding buffer containing [BF4]-based RTIls with a systematic variety of alkyl chain on cations varying from two to eight, less hydrophobic imidazolium cations having shorter alkyl chains were effective to facilitate lysozyme refolding. compared to the conventional refolding buffer, 2 times higher lysozyme refolding yield was obtained in 1-ethyl-3-methylimidazolium tetrafluoroborate ([EMIM][BF4]) containing refolding buffer. The refolding yield of lysozyme was even more increased by 2.5 times when 1-butyl-3-methylimidazolium methylsulfate ([BMIM][MS]) containing sulfur residue on anion was used. The sulfur residue in [BMIM][MS] is supposed to improve the refolding yield of lysozyme which has 4 intramolecular disulfide bonds. For dilution-based refolding of lysozyme, the optimum concentrations of RTIls in refolding buffer were found to be 1.0 M [EMIM][BF4] and 0.5 M [BMIM][MS], respectively. The optimum temperate for dilution-based refolding of lysozyme with RTIls was 4 °c.

Improving the capacitance and energy density is a significant challenge while developing practical and flexible energy storage system(ESS).Redox mediators(RMs),as redox-active electrolyte additives,can provide additional energy storing capability via electrochemical faradaic contribution on electrodes for high-performance flexible ***,determining effective material combinations between electrodes and RMs is essential for maximizing surface faradaic redox reactions for energy-storage *** this study,an electrode-RM system comprising heterostructured hybrid(carbon fiber(cF)/MnO_(2)) faradaic electrodes and iodine RMs(I-RMs) in a redox-active electrolyte is *** cF/MnO_(2)with the 1-RMs(cF/MnO_(2)-I) induces dominant catalytic faradaic interaction with the I-RMs,significantly enhancing the surface faradaic kinetics and increasing the overall energy-storage *** cF/MnO_(2)-I ESSs show a 12.6-fold(or higher) increased volumetric energy density of 793.81 mWh l^(-1)at a current of 10 μA relative to ESSs using cF/MnO_(2)without I-RMs(cF/MnO_(2)).Moreover,the cF/MnO_(2)-I retains 93.1% of its initial capacitance after 10,000 cycles,validating the excellent ***,the flexibility of the ESSs is tested at different bending angles(180° to 0°),demonstrating its feasibility for flexible and high-wear ***,cF/MnO_(2)electrodes present a practical material combination for high-performance flexible energy-storage devices owing to the catalytic faradaic interaction with I-RMs.

Effects of pulsed radiofrequency on spasticity in patients with spinal cord injury:a report of two cases

Spasticity following spinal cord injury（ScI） results in functional deterioration and reduced quality of life. Herein, we report two ScI patients who presented with good response to pulsed radiofrequency（PRF） for the management of spasticity in the lower extremities. Patient 1（a 47-year-old man） had complete thoracic cord injury and showed a phasic spasticity on the extensor of both knees（3–4 beats clonus per every 30 seconds） and tonic spasticity（Modified Ashworth Scale： 3） on both hip adductors. Patient 2（a 64-yearold man） had incomplete cervical cord injury and showed a right ankle clonus（approximately 20 beats） when he walked. After the application of PRF to both l2 and l3 dorsal root ganglion（DRG）（patient 1） and right S1 DRG（patient 2） with 5 Hz and 5 ms pulsed width for 360 seconds at 45 V under the c-arm guide, all spasticity disappeared or was reduced. Moreover, the effects of PRF were sustained for approximately 6 months with no side effects. We believe that PRF treatment can be useful for patients with spasticity after ScI.

In plane micro-supercapacitors that are miniaturized energy storage components have attracted significant attention due to their high power densities for various ubiquitous and sustainable device systems as well as their facile integration on various flexible/wearable *** implement the micro-supercapacitors in various practical applications that can accompany solid state or gel electrolyte and flexible substrates,ions must be readily transported to electrodes for achieving high power ***,we show large enhancement in electrochemical properties of flexible,inplane micro-supercapacitor using sharp-edged interdigitated electrode design,which was simply fabricated through direct laser scribing *** sharp-edged electrodes allowed strong electric field to be induced at the corners of the electrode fingers which led to the greater accumulation of ions near the surface of electrode,significantly enhancing the energy storage performance of *** electric field-enhanced in-plane micro-supercapacitor showed the volumetric energy density of 1.52 Wh l^(−1)and the excellent cyclability with capacitive retention of 95.4%after 20000 *** further showed various practicability of our sharp-edged design in micro-supercapacitors by showing circuit applicability,mechanical stability,and air *** results present an important pathway for designing electrodes in various energy storage devices.

A straight, non-sporulating, Gram-variable bacillus (HKU24T) was recovered from the blood culture of a patient with metastatic breast carcinoma. After repeated subculturing in BAcTEc Plus Anaerobic/F blood culture broth, HKU24T grew on brucella agar as non-hemolytic, pinpoint colonies after 96 h of incubation at 37 °c in an anaerobic environment and aerobic environment with 5% cO2. Growth was enhanced with a streak of Staphylococcus aureus. HKU24T was non-motile and catalase-negative, but positive for alkaline phosphatase, β-glucosidase, and α-glucosidase. It hydrolyzed phenylphosphonate and reduced resazurin. 16S rRNA, groEl, gyrB, recA, and rpoB sequencing showed that HKU24T occupies a distinct phylogenetic position among the leptotrichia species, being most closely related to leptotrichia trevisanii. Using HKU24T groEl, gyrB, recA, and rpoB gene-specific primers, fragments of these genes were amplified from one of 20 oral specimens. Based on phenotypic and genotypic characteristics, we propose a new species, leptotrichia hongkongensis sp. nov., to describe this bacterium.

This paper studies the discrete minus one norm least-squares methods for the stress formulation of pure displacement linear elasticity in two dimensions. The proposed least-squares functional is defined as the sum of the l2- and H-1-norms of the residual equations weighted appropriately. The minus one norm in the functional is replaced by the discrete minus one norm and then the discrete minus one norm least-squares methods are analyzed with various numerical results focusing on the finite element accuracy and multigrid convergence performances.

Impacts of low level aflatoxin in feed and the use of modified yeast cell wall extract on growth and health of nursery pigs

This study was to investigate the effect of corn naturally contaminated with aflatoxins(AF)under the regulatory level on the growth performance and health of nursery pigs,and the efficiency of yeast cell wall based feed additive(Yc)mainly composed ofβ-glucans and mannan oligosaccharide(MOS)(Integral A+,Alltech,lexington,KY)in prevention of ***(60 barrows and 60 gilts at 6.02 ± 0.83kg BW)were randomly allotted to 4 treatments in a randomized complete block design based on a 2×2 factorial arrangement with 10 pens(5 barrow and 5 gilt pens)per treatment and 3 pigs per *** were fed experimental diets for 5 *** factor was AF(0 or 20 mg/kg in feed)and the second factor was Yc(0 or 2 g/kg in feed).Feed intake and body weight were measured weekly,and blood samples were used to measure blood cell counts,immunoglobulin G(Ig G),tumor necrosis factor-a(TNF-a),oxidative damage status,and serological evaluation related to liver *** decreased(Platelet count(247.4 to 193.5×10~3/μl),and it also tended to increase the level of albumin(P=0.055,3.46 to 3.63 g/d l),albumin:globulin ratio(P=0.050,2.09 to 2.37),and ca(P=0.080,10.79 to 10.97 mg/d l).Yeast cell wall based feed additive increased(P cted,and it also tended to increase(P=0.055)albumin level(3.46 to 3.63 g/d l).Interactions(P lobin,hematocrit,and platelet count indicated that Yc further increased their levels when pigs were eating AF contaminated ***(P lood urea N to creatinine ratio indicated that Yc further decreased their levels when feed were contaminated with *** conclusion,low level of 20 mg AF/kg under the regulatory level had minor effects on hematology without affecting growth performance,however the supplementation of 2 g/kg Yc as a source ofβ-glucans and MOS in feed can improve feed intake and therefore the growth of pi

Introduction Using the information obtained from analysis of 16S rRNA gene sequences,Tsukamurella was first proposed as a genus in 1988,1 despite the fact that the first strain of this group of bacteria was described in 1941,2 and the first human isolate was reported in 1971.3 Similar to related genera of the order corynebacteriales,such as Nocardia,Rhodococcus,and Gordonia,members of Tsukamurella are Gram-positive,aerobic,catalase-positive,and partially acid-fast as a result of the presence of mycolic acid in the cell *** to their similar phenotypic properties,differentiation of these genera and speciation within these genera is difficult in most clinical microbiology laboratories.

Electrical and corrosion Properties of Titanium Aluminum Nitride Thin Films Prepared by Plasma-Enhanced Atomic layer Deposition

Titanium-aluminum-nitride（TiAlN） films were grown by plasma-enhanced atomic layer deposition（PEAlD）on 316 l stainless steel at a deposition temperature of 200 °c. A supercycle, consisting of one AlN and ten TiN subcycles, was used to prepare TiAlN films with a chemical composition of Ti（0.25）Al（0.25）N（0.50）. The addition of AlN to TiN resulted in an increased electrical resistivity of TiAlN films of 2800 μΩ cm, compared with 475 μΩ cm of TiN films, mainly due to the high electrical resistivity of AlN and the amorphous structure of TiAlN. However, potentiostatic polarization measurements showed that amorphous TiAlN films exhibited excellent corrosion resistance with a corrosion current density of 0.12 μA/cm^2, about three times higher than that of TiN films, and about 12.5 times higher than that of 316 l stainless steel.
