Assessing bioactivity of environmental water samples filtered using nanomembrane technology and mammalian cell lines

This project reports on the use of a novel nanomembrane filtering technology to isolate and analyze the bioactivity of microplastic(MP)-containing debris from Lake Ontario water *** MPs are a complex mixture of polymers and sorbed chemicals that are persistent and can exhibit a wide range of toxic *** human exposure to MPs is unavoidable,it is necessary to characterize their bioactivity to assess potential health *** work seeks to quantify MP presence in the nearshore waters of Lake Ontario and begin to characterize the bioactivity of the filtrate containing *** utilized silicon nitride(SiN)nanomembrane technology to isolate debris sized between 8 and 20μm from lake water samples collected at various times and *** were identified with Nile red ***-based assays were conducted directly on the filtered debris to test for cell viability,aryl hydrocarbon receptor(AhR)activity,and interleukin 6(IL-6)levels as a measure of proinflammatory *** samples contained *** of the isolated debris impacted cell ***,AhR activity and IL-6 levels varied over ***,no associations were observed between the amount of plastic and *** differences in activity are likely due to variations in the physiochemical properties of debris between *** results highlight the need for increased sampling to fully characterize the bioactivity of MPs in human cells and to elucidate the role that sample physiochemical and spatiotemporal properties play in this activity.

Steroidal glycoalkaloids(SGAs)are specialized metabolites produced by hundreds of Solanum species,including important vegetable crops such as tomato,potato,and *** it has been known that SGAs play important roles in defense in plants and“anti-nutritional"effects(e.g.,toxicity and bitterness)to humans,many of these molecules have documented anti-cancer,anti-microbial,antiinflammatory,anti-viral,and anti-pyretic *** these,α-solasonine andα-solamargine isolated from black nightshade(Solanum nigrum)are reported to have potent anti-tumor,anti-proliferative,and anti-inflammatory ***,α-solasonine andα-solamargine,along with the core steroidal aglycone solasodine,are the most widespread SGAs produced among the Solanum ***,it is still unknown how plants synthesize these bioactive steroidal *** comparative metabolomictranscriptome-guided approach,biosynthetic logic,combinatorial expression in Nicotiana benthamiana,and functional recombinant enzyme assays,here we report the discovery of 12 enzymes from *** that converts the starting cholesterol precursorto solasodine aglycone,and the downstreamα-solasonine,α-solamargine,and malonyl-solamargine SGA *** further identified six enzymes from cultivated eggplant that catalyze the production ofα-solasonine,α-solamargine,and malonyl-solamargine SGAs from solasodine aglycone via glycosylation and atypical malonylation *** work provides the gene tool box and platform for engineering the production of high-value,steroidal bioactive molecules inheterologoushosts usingsyntheticbiology.

In this paper,we present a probabilistic numerical method for a class of forward utilities in a stochastic factor *** this purpose,we use the representation of forward utilities using the ergodic Backward Stochastic Differential equations(eBSDes)introduced by Liang and Zariphopoulou in[27].We establish a connection between the solution of the ergodic BSDe and the solution of an associated BSDe with random terminal time T,defined as the hitting time of the positive recurrent stochastic *** viewpoint based on BSDes with random horizon yields a new characterization of the ergodic cost^which is a part of the solution of the *** particular,for a certain class of eBSDes with quadratic generator,the Cole-Hopf transformation leads to a semi-explicit representation of the solution as well as a new expression of the ergodic cost>.The latter can be estimated with Monte Carlo *** also propose two new deep learning numerical schemes for ***,we present numerical results for different examples of eBSDes and forward utilities together with the associated investment strategies.

Psychological consequences of a diagnosis of infertility in men:a systematic analysis

Almost half of infertility cases involve male *** the consequence of a diagnosis of male infertility,as a sole or partial contributor to the couples'infertility,to the mental health of men is required to ensure clinical care meets their psychological *** aim of this systematic analysis was to synthesize the evidence regarding whether men diagnosed with male factor infertility experience greater psychological distress than(1)men described as fertile and(2)men in couples with other infertility *** databases were searched using a combination of Medical Subject Headings(MeSH)headings and keywords relating to male infertility and psychological *** search yielded 1016 unique publications,of which 23 were included:8 case-control,14 prospective cohort,and 1 data linkage *** aspects of psychological distress were identified depression,anxiety,self-esteem,quality of life,fertility-related stress,general psychological stress or well-being,and psychiatric ***-control studies reported that men with male factor infertility have more symptoms of depression,anxiety and general psychological distress,worse quality of some aspects of life,and lower self-esteem than *** men with male factor infertility were compared to men in couples with other causes of infertility,there were few differences in the assessed aspects of psychological *** methodological limitations within the studies,this systematic analysis suggests that the experience of infertility,irrespective of its cause,negatively affects men's mental health and demonstrates the need for assisted reproduction technology(ART)providers to consider men undergoing assisted reproduction as individuals with their own unique support needs.

This editorial elaborates on the current and future applications of linear endoscopic ultrasound(eUS),a substantial diagnostic and therapeutic modality for various anatomical *** scope of endosonographic assessment is broad and,among other factors,allows for the evaluation of the mediastinal anatomy and related pathologies,such as mediastinal lymphadenopathy and the staging of central malignant lung ***,eUS assessment has proven more accurate in detecting small lesions missed by standard imaging examinations,such as computed tomography or magnetic resonance *** focus on its current uses in the mediastinum,including lung and esophageal cancer staging,as well as evaluating mediastinal lymphadenopathy and submucosal *** editorial also explores future perspectives of eUS in mediastinal examination,including ultrasound-guided therapies,artificial intelligence integration,advancements in mediastinal modalities,and improved diagnostic approaches for various mediastinal lesions.

Objective exercise is Medicine(eIM)is a global initiative encouraging healthcare providers to routinely assess and promote physical activity(PA)among *** objective of this study was to evaluate the feasibility,adoption,implementation and effectiveness of eIM from patient,clinician and healthcare staff perspectives using a combination of electronic health record(eHR),survey and interview *** This study used a combination of the Practical Robust Implementation and Sustainability Model(PRISM)and the Learning evaluation model to implement *** captured from the eHR,including Physical Activity Vital Sign(PAVS)scores,and data collected from qualitative surveys and interviews were used to evaluate the programme’s Reach,effectiveness,Adoption,Implementation,Maintenance(Re-AIM,which is embedded within PRISM)from provider,staff and patient *** Five primary care clinics within a large academic health *** A total of 24443 patients from all participating clinics had at least one PAVS score during the study period.A total of 17 clinicians completed surveys,and 4 clinicians,8 medical assistants and 9 patients completed *** Implementation fidelity metrics varied widely between components and across clinics but were generally consistent over time,indicating a high degree of programme *** was highest during the first 6 months of the COVID-19 pandemic when most visits were *** PAVS scores increased from 57.7(95%CI:56 to 59.4)to 95.2(95%CI:91.6 to 98.8)min per week at 6 months for patients not meeting PA guidelines at baseline and decreased from 253.84(95%CI:252 to 255.7)to 208.3(95%CI:204.2 to 212.4)min per week at 6 months for patients meeting PA guidelines at *** eIM implementation,clinician-estimated time spent discussing PA with patients increased for 35%of providers and stayed the same for 53%.Conclusion Overall,this study established eIM’s feasibility,adoption,implementation

Social learning is a mechanism used by many species to effciently gain information about their *** many animals live in an environment where members of other species are present,little is known about interspecifc social *** and urbanized species provide the opportunity to investigate whether nonhuman animals can learn from heterospecifcs such as humans,who are integral parts of their social *** domestic dogs Canis familiaris have been intensively researched for their ability to learn from humans,most studies have focused on dogs living as ***,free-ranging dogs represent the majority of the world’s dog population,they live alongside humans,scavenge on human refuse,and are subject to natural and sexual ***,free-ranging dogs with extensive exposure to humans and their artifacts provide the opportunity to investigate interspecifc social learning in a naturalistic setting,where learning from humans might be a beneft for *** we tested individual free-ranging dogs in a between-subject design:Dogs in the control group could spontaneously choose between two novel and differently patterned food-delivering *** the experimental group,instead,dogs could frst observe an unfamiliar human approaching and eating from 1 of the 2 *** provide the frst evidence that free-ranging dogs match the choice of an unfamiliar *** results show that at least simple forms of interspecifc social learning might be involved in dogs’success in living alongside humans in a complex urbanized environment.

Spatial dynamics and Palestinian resilience:The lifestyle of Al-Kurshan Bedouins in Bethlehem District

This study explores the role of Bedouins in preserving Palestinian land through their own means of community resilience through the concept of“Art of being”and its entailed“Arts of presence and persistence”.This is examined through the correlation between Bedouin lifestyle and the spatial distribution of structures within“Al-Kurshan”Bedouin clan in Area C in the West *** on a mixed qualitative and spatial methodologies,information was gathered through semi-structured interviews with Al-Kurshan Bedouins and employees of organizations supporting Bedouins,on-site observation,and analyses of maps and photos,in addition to drawings and *** spatial distribution of Bedouin built structures is analyzed in relation to landscape and climate,social aspects within the families,and available service and sectorial *** analyses give insight on how Bedouins struggle against adopting other *** study shows that the Bedouins are slowly forced to negotiate their semi-nomadic lifestyle,but resist giving up their right to exist against the threat of forced *** maintain their“Art of being”which partly represents their own way of safeguarding Palestinian lands from *** study contributes to the wider discussion of spatial dynamics of Bedouins and their community resilience within the Palestinian geopolitical context.
