Dual inhibition of COVID-19 spike glycoprotein and main protease 3CLpro by Withanone from Withania somnifera

Objective:To identify the safe and effective natural inhibitors of spike glycoprotein and main protease 3CLpro using potential natural antiviral compounds which are studied under various animal models and viral cell ***:First,compounds were retrieved from the Pub Chem database and predicted for their druggability using the Mol Soft web server,and compounds having drug-like property were predicted for major adverse drug reactions like cardiotoxicity,hepatotoxicity,arrhythmia,myocardial infarction,and nephrotoxicity using *** of nontoxic antiviral compounds with spike glycoprotein and main protease 3CLpro was performed using Auto Dock vina by PyRx 0.8 *** stability of compoundprotein interactions was checked by molecular dynamic(MD)simulation using Schrodinger Desmond ***:Based on the druggable and nontoxic profile,nine compounds were *** them,Withanone from Withania somnifera showed the highest binding affinity and best fit at active sites 1 of spike glycoprotein(glycosylation site)and main protease 3CLpro via interacting with active site amino acid residues before and after MD simulation at 50 ***,which may reduce the glycosylation of SARS-CoV-2 via interacting with Asn343 and inhibit viral ***:The current study reports Withanone as a non-toxic antiviral against SARS-CoV-2 and serve as a potential lead hit for further experimental validation.

Numerical modelling of post-failure behaviors of coal specimens

A modelling approach consisting of best-fit relations to estimate the post-yield strength parameters is presented for simulating post-peak behavior beyond the point of residual strength of coal pillars having different w/h *** model was developed based on back-analysis of the complete stress-strain behavior of specimens belonging to six different Indian coal seams with different w/h ratios of 0.5 *** was found that the simultaneous degradation of the cohesion and friction angle of the Mohr-Coulomb rock material characterizes the post-peak strength behavior of the *** resulting expressions are simplistic as they require parameters that can be easily determined using uniaxial and triaxial compression ***,the developed model was validated by simulating the triaxial tests of coal specimens with different sizes under varying confining stresses and comparing its findings with the published test *** study showed that its implementation in the numerical model could reproduce laboratory-observed mechanical response,deformation behavior,and failure mechanism very closely.

Resistance and resilience of soil biological indicators:A case study with multi-walled carbon nanotube

Soil ecosystem is experiencing stresses due to climate change,and soil inhabitants try to demonstrate their inherent resistance and resilience against those *** of nanomaterials as agricultural inputs could bring shifts in resistance and resilience patterns of soil microbes and associated enzymes,especially under short-term heat *** this background,the impacts of multi-walled carbon nanotube(MWCNT)on the resistance and resilience of soil biological indicators were *** incubation experiment was conducted with varied MWCNT concentrations(0,50,100,250,and 500 mg kg-1soil)for 90 d after 24-h heat stress at 48±2?C to assess the impacts of MWCNT on soil enzyme activities and microbial populations vis-à-vis their resistance and resilience indices under short-term exposure to heat *** activities were reduced after exposure to heat *** indices of enzyme activities were enhanced by MWCNT application on day 1 after heat stress,whereas there was no recovery of enzyme activities after 90-d *** soil enzyme activities,resistance index values of soil microbial populations followed the similar trend and were improved by MWCNT ***-walled carbon nanotube has the potential to improve resistance indices of soil enzyme activities and microbial populations under heat stress,although they could not recover to their original state during periodical incubation after heat *** study helps to understand the relative changes of biological indicators under MWCNT and their ability to withstand heat stress.

Catalytic degradation of methylene blue by Fenton like system: model to the environmental reaction

To develop more efficient chemical methods for the demineralization of organic pollutants from water bodies, which one was also mimic to the nature, a degradation of methylene blue by Fe(Ⅲ) and h 2O 2 in the absence of light instead of Fe(Ⅱ) and h 2O 2 was studied. Results showed that use of Fe (Ⅲ) is more promising than Fe(Ⅱ). The present study reflects that Fenton reaction is more efficient, in the presence of a small amount of salicylic acid is added which is a one of the priority pollutant.

Various 2-arylbenzimidazoles were synthesized from o-phenylenediamine and aldehydes via one-step process using DMP （Dess-Martin-periodinane） reagent as an oxidant. Liquefaction proneness of stratified sand-silt layers based on cyclic triaxial tests

Most studies on liquefaction have addressed homogeneous soil strata using sand or sand with fine content without considering soil *** this study,cyclic triaxial tests were conducted on the stratified sand specimens embedded with the silt layers to investigate the liquefaction failures and void-redistribution at confining stress of 100 kPa under stress-controlled *** loosening of underlying sand mass and hindrance to pore-water flow caused localized bulging at the sand-silt *** is observed that at a silt thickness of 0.2h(h is the height of the specimen),nearly 187 load cycles were required to attain liquefaction,which was the highest among all the silt thicknesses with a single silt ***,0.2h is assumed as the optimum silt thickness(t_(opt)).The silt was placed at the top,middle and bottom of the specimen to understand the effect of silt layer *** to the increase in depth of the silt layer from the top position(capped soil state)to the bottom,the cycles to reach liquefaction(N_(cyc,L))increased 2.18 ***,when the number of silt layers increased from single to triple,there was an increase of about 880%in N_(cyc,L).The micro-characterization analysis of the soil specimens indicated silty materials transported in upper sections of the specimen due to the dissipated pore *** main parameters,including thickness(t),location(z),cyclic stress ratio(CSR),number of silt layers(n)and modified relative density(D_(r,m)),performed significantly in governing the lique-faction *** this,a multilinear regression model is developed based on critical parameters for prediction of N_(cyc,L).Furthermore,the developed constitutive model has been validated using the data from the present study and earlier findings.

Maternal salinity environment affects salt tolerance during germination in Anabasis setifera： A facultative desert halophyte

The effects of maternal salinity and light incubation on the salinity tolerance of the facultative halophyte Anabasis setifera during their germination stages were assessed. Seeds were collected from non-saline habitats in Egypt and saline habitats in the United Arab Emirates （UAE）. The seeds of the two populations were germinated in 0, 100, 200, 400, 600 and 800 mM NaCI, and incubated at 25℃/15℃ in both 12-h light and 12-h darkness regimes and continuous darkness. Significantly more seeds germinated in the Egyptian population than in the UAE population. Salinity tolerance was significantly greater with the Egyptian population than with the UAE population, especially under the conditions of higher salinities. The difference in salinity tolerance between the seeds of two populations was attributed to their seed mass. In addition, germination was significantly faster for the Egyptian population than for the UAE population. Most of the saline treated seeds were able to recover their germination when transferred to distilled water, but this depended on their maternal salinity and light incubation. Recovery from higher salinities was significantly better for the seeds under darkness than for those under light in the UAE population, but the reverse was true for the seeds in the Egyptian population. The higher salinity tolerance for the A. setifera seeds from the non-saline Egyptian population and the lower salinity tolerance for the seeds from the saline UAE population cannot explain their natural distribution. Further studies about other possible roles, such as levels of different promoting and inhibiting phytohormones, are needed to understand the importance of salinity as an environmentally induced maternal effect.

Prospective use of bacteriocinogenic Pediococcus pentosaceus as direct-fed microbial having methane reducing potential

Direct-fed microbials（DFM）, generally regarded as safe status, are successfully used in improving rumen ecology, gastro-intestinal health, feed efficiency, milk production and growth rate in ruminants. On the other hand, methanogenesis in rumen, which accounts for a significant loss of ruminant energy and increased greenhouse gas in environment, is of great concern, therefore, use of DFM for improving productivity without compromising the animal health and ecological sustainability is encouraged. The present study was conducted to investigate the methane reducing potential of bacteriocinogenic strain Pediococcus pentosaceus-34. Since, the culture showed no hemolysis on blood agar and DNase activity, hence, it was considered to be avirulent in nature, a prerequisite for any DFM. The culture also showed tolerance to ph 5.0 for 24 h with 0.5% organic acid mixture, whereas when given a shock for 2 h at different p h and organic acids concentrations, it showed growth at ph 3.0 and 4.0 with 0.1 and 1.0% organic acids, respectively, as having good animal probiotics attributes. The total gas production was significantly（P〈0.05） higher in live pedicoccal culture（LPC） and dead pedicoccal culture（DPC） both with wheat straw, when compared to the control. In sugarcane bagasse, gas production was significantly lower（P〈0.05） with LPC compared to the control and DPC both. Methane was reduced by the inclusion of LPC in sugarcane bagasse（0.07 mL Ch4 mg–1 dry matter digestibility） with no effect on other rumen fermentation parameters. however, with wheat straw and LPC total gas, in vitro dry matter digestibility, total volatile fatty acids increased significantly but no reduction in methane production was observed in comparison to the control. Therefore, further research is warranted in this direction, if the bacteriocinogenic strains can be used as DFM for ruminants to improve the ruminant productivity.

Augmentation cystoplasty in children with stages III and IV chronic kidney disease secondary to neurogenic bladder

Objective:This study examines the outcome of augmentation cystoplasty(AC)in children with stages III and IV chronic kidney disease(CKD)secondary to neurogenic bladder in which transplantation was not ***:hospital records of all children with CKD stages III and IV who underwent AC between 2008 and 2017 were retrieved to study outcome and complications including estimated glomerular filtration rate(eGFR,National Kidney Foundation Calculator),somatic growth(percentiles,compared with population data),and febrile urinary tract infections(fUTIs)requiring *** analysis was performed using ***:AC was performed in 13 children with CKD stages III and IV(10 girls;median 8.0 years)with median follow-up of 51 *** had incontinence(10/13),reflux(7/13),and hydronephrosis(13/13)despite antimuscarinics and intermittent *** capacity was 74%of expected and median compliance was 5 mL/cm h20(inter-quartile range 4 mL/cm h20).All underwent ileocystoplasty(25 cm bowel).One each had nephrectomy and mitrofanoff *** had resolution of *** had acute kidney injury that *** eGFR at presentation(24 mL/min/1.73 m2)improved with conservative management alone(52 mL/min/1.73 m2,p=0.004).This improved further 1 year following AC(61 mL/min/1.73 m2,p=0.036)with stable function at 7 *** was improvement in somatic growth,hydronephrosis,and fUTI despite no ureteric ***:AC carries acceptable morbidity in children with CKD stages III and IV secondary to neurogenic *** is effective with improvements in continence,eGFR,somatic growth,and propensity for *** re-implantation might not be necessary.

Prevalences of oligozoospermia and azoospermia in male partners of infertile couples from different parts of India

Aim： To determine whether there was any regional variation in the prevalence of azoospermia, oligozoospermia and mean sperm counts in male partners of infertile couples from different parts of India. Methods： Data on 16 714 semen samples analyzed over the past five years from six different laboratories located in five cities of India were collated and evaluated. Results： There was a regional variation in the prevalence of azoospermia. The prevalence of azoospermia was extremely high in Kurnool and Jodhpur （38.3% and 37.4%, respectively）. There was also a regional variation in the prevalence of oligozoospermia （51%） in Kurnool. There was no significant difference in the mean sperm counts in normospermic men. Conclusion： There is a regional variation in the prevalence of azoospermia and oligozoospermia in the male partners of infertile couples from different regions of India. The prevalence of azoospermia in Kumool and Jodhpur is higher than any other worldwide reported literature, Further studies need to be carded out to determine the cause of this. （Asian J Androl 2006 Jan; 8： 89-93）
