NKX6.3 protects against gastric mucosal atrophy by downregulatingβ-amyloid production

BACKGROUND Atrophic gastritis is characterized by loss of appropriate glands and reduction in gastric secretory function due to chronic inflammatory processes in gastric mucosa. Moreover, atrophic gastritis is considered as a precancerous condition of gastric cancer. However, little is known about the molecular mechanism underlying gastric mucosal atrophy and its contribution to gastric ***, we hypothesized that transcription factor NKX6.3 might be involved in maintaining gastric epithelial homeostasis by regulating amyloid β(Aβ)*** To determine whether NKX6.3 might protect against gastric mucosal atrophy by regulating Aβ *** We identified NKX6.3 depletion induced cell death by cell count and Western blot assay. Production and mechanism of Aβ oligomer were analyzed by enzymelinked immunosorbent assay, Western blot, immunoprecipitation, real-timequantitative polymerase chain reaction and immunofluorescence analysis. We further validated the correlation between expression of NKX6.3, Helicobacter pylori CagA, Aβ oligomer, apolipoprotein E(ApoE), and β-secretase 1(Bace1) in 55 gastric *** NKX6.3 depletion increased both adherent and floating cell populations in HFE-145 cells. Expression levels of cleaved caspase-3,-9, and poly ADP ribose polymerase were elevated in floating HFE-145^(shNKX6.3) cells. NKX6.3 depletion produced Aβ peptide oligomers, and increased expression of ApoE, amyloid precursor protein, Aβ, Bace1, low-density lipoprotein receptor, nicastrin, high mobility group box1, and receptor for advanced glycosylation end product proteins. In immunoprecipitation assay, γ-secretase complex was stably formed only in HFE-145^(shNKX6.3) cells. In gastric mucosae with atrophy, expression of Aβpeptide oligomer, ApoE, and Bace1 was detected and inversely correlated with NKX6.3 expression. Intelligent vision-based surveillance systems are designed to deal with the gigantic volume of videos captured in a particular environment to perform the interpretation of scenes in form of detection,tracking,monitoring,behavioral analysis,and *** addition to that,another evolving way of surveillance systems in a particular environment is human gait-based *** the existing research,several methodological frameworks are designed to use deep learning and traditional methods,nevertheless,the accuracies of these methods drop substantially when they are subjected to covariate *** covariate variables disrupt the gait features and hence the recognition of subjects becomes *** handle these issues,a region-based triplet-branch Convolutional Neural Network(CNN)is proposed in this research that is focused on different parts of the human Gait Energy Image(GEI)including the head,legs,and body separately to classify the subjects,and later on,the final identification of subjects is decided by probability-based majority voting ***,to enhance the feature extraction and draw the discriminative features,we have added soft attention layers on each branch to generate the soft attention *** proposed model is validated on the CASIA-B database and findings indicate that part-based learning through triplet-branch CNN shows good performance of 72.98%under covariate conditions as well as also outperforms single-branch CNN models.

AIM： To determine intestinal permeability, the serum tumor necrosis factor （TNF）-α level and urine nitric oxide （NO） metabolites are altered in liver cirrhosis （LC） with or without ascites. METHODS： Fifty-three patients with LC and 26 healthy control subjects were enrolled in the study. The intestinal permeability value is expressed as the percentage of polyethylene glycol （PEG） 400 and 3350 retrieval in 8-h urine samples as determined by high performance liquid chromatography. Serum TNF-α concentrations and urine NO metabolites were determined using an enzyme-linked immunosorbent assay （ELISA） and Greiss reaction method, respectively. RESULTS： The intestinal permeability index wassignificantly higher in patients with LC with ascites than in healthy control subjects or patients with LC without ascites （0.88 ± 0.12 vs 0.52 ± 0.05 or 0.53 ± 0.03, P 〈 0.05） and correlated with urine nitrite excretion （r = 0.98）. Interestingly, the serum TNF-α concentration was significantly higher in LC without ascites than in control subjects or in LC with ascites （198.9 ± 55.8 pg/mL vs 40.9 ± 12.3 pg/mL or 32.1 ± 13.3 pg/mL, P 〈 0.05）. Urine nitrite excretion was significantly higher in LC with ascites than in the control subjects or in LC without ascites（ 1170.9± 28.7 μmol/L vs 903.1 ± 55.1 μmol/L or 956.7 ± 47.7 μmol/L, P 〈 0.05）. COMCLUSIOM： Increased intestinal macromolecular permeability and NO is probably of importance in the pathophysiology and progression of LC with ascites, but the serum TNF-α concentration was not related to LC with ascites.

Endoscopic fibrin glue injection for closure of pancreatocutaneous fistula following transgastric endoscopic necrosectomy

Transgastric endoscopic necrosectomy has been recently introduced as the effective and alternative management of infected pancreatic necrosis and pancreatic abscess. However,up to 40% of patients who undergo endoscopic necrosectomy may need an additional percutaneous approach for subsequent peripancreatic fluid collection or non-resolution of pancreatic necrosis. This percutaneous approach may lead to persistent pancreatocutaneous fistula,which remains a serious problem and usually requires prolonged hospitalization,or even open-abdominal surgery. We describe the first case of pancreatocutaneous fistula and concomitant abdominal wall defect following transgastric endoscopic necrosectomy and percutaneous drainage,which were endoscopically closed with fibrin glue injection via the necrotic cavity.

Highly efficient reduction of aqueous Cr(VI) with novel ZnO/SnS nanocomposites through the piezoelectric effect

In this work, novel ZnO/SnS nanocomposites were successfully synthesized via a hydrothermal approach, which is developed for piezoelectric catalytic reduction of hexavalent chromium(Cr(VI)) in an aqueous solution. The constructed ZnO/SnS nanocomposites exhibited higher catalytic efficiency for Cr(VI) reduction under a mechanical force(e.g., ultrasonic vibration) compared to pristine ZnO and SnS. In particular, the ZnO/SnS(with 30 wt.% of SnS) heterojunctions revealed an optimal degradation activity among all the prepared samples, which completely removed the Cr(VI)(20 mg/L) solution within 35 min. Moreover, the piezoelectric catalytic activity of ZnO/SnS remained stable after four consecutive cycling experiments. The results of the morphology observations indicated that the SnS nanoparticles adhere to the surface of the ZnO nanorods. The improved piezoelectric catalytic performance of the ZnO/SnS heterojunctions can be attributed to the formation of an intimate interfacial between ZnO and SnS, which effectively inhibits the electron-hole recombination and speeds up the rate of charge transfer. The study reveals a new design of ZnO/SnS heterojunctions as a high-performance and eco-friendly piezoelectric catalyst and provides a promising strategy for addressing environmental problems and energy crises.

Ultra-narrow-linewidth mode-locked lasers with wide wavelength tunability can be versatile light sources for a variety of newly emergent ***,it is very challenging to achieve the stable mode locking of substantially long,anomalously dispersive fiber laser cavities employing a narrowband spectral filter at the telecom ***,we show that a nearly dispersion-insensitive dissipative mode-locking regime can be accessed through a subtle counterbalance among significantly narrowband spectral filtering,sufficiently deep saturable absorption,and moderately strong in-fiber Kerr *** achieves ultra-narrow-linewidth(a few gigahertz)nearly transform-limited self-starting stable dissipative soliton generation at low repetition rates(a few megahertz)without cavity dispersion management over a broad tuning range of wavelengths covering the entire telecom *** unique laser may have immediate application as an idealized pump source for high-efficiency nonlinear frequency conversion and nonclassical light generation in dispersion-engineered tightly light-confining microphotonic/nanophotonic systems.

Background Central blood pressure （BP） is pathophysiologically more important than peripheral BP for the pathogenesis of cardiovascular disease. Arterial stiffness is also a good predictor of cardiovascular morbidity and mortality. The effects of benidipine, a unique dual L-IT-type calcium channel blocker, on central BP have not been reported. This study aimed to compare the effect of benidipine and Iosartan on the central BP and arterial stiffness in mild to moderate essential hypertensives. Methods This 24 weeks, multi-center, open label, randomized, active drug comparative, parallel group study was designed as a non-inferiority study. The eligible patients （n=200） were randomly assigned to receive benidipine （n=101） or Iosartan （n=99）. Radial artery applanation tonometry and pulse wave analysis were used to measure the central BP, pulse wave velocity （PWV） and augmentation index （AIx）. We also measured the metabolic and inflammatory markers. Results After 24 weeks, the central BP decreased significantly from baseline by （16.8±14.0/10.5±9.2） mmHg （1 mmHg =0.133 kPa）（systolic/diastolic BP; P 〈0.001） in benidipine group and （18.9±14.7/12.1±10.2） mmHg （P 〈0.001） in Iosartan group respectively. Both benidipine and Iosartan groups significantly lowered peripheral BP （P 〈0.001） and AIx （P 〈0.05）, but there were no significant differences between the two groups. The mean aortic, brachial and femoral PWV did not change in both groups after 24-week treatment. There were no significant changes of the blood metabolic and inflammatory biomarkers in each group. Conclusion Benidipine is as effective as Iosartan in lowering the central and peripheral BP, and improving arterial stiffness.
