Cytomegalovirus frequency in neonatal intrahepatic cholestasis determined by serology, histology, immunohistochemistry and PCR

AIM: To determine cytomegalovirus (CMV) frequency in neonatal intrahepatic cholestasis by serology, histological revision (searching for cytomegalic cells), immunohistochemistry, and polymerase chain reaction (PCR), and to verify the relationships among these ***: The study comprised 101 non-consecutive infants submitted for hepatic biopsy between March 1982 and December 2005. Serological results were obtained from the patient's f iles and the other methods were performed on paraffin-embedded liver samples from hepatic biopsies. The following statistical measures were calculated: frequency, sensibility, specif ic positive predictive value, negative predictive value, and ***: The frequencies of positive results were as follows: serology, 7/64 (11%); histological revision, 0/84; immunohistochemistry, 1/44 (2%), and PCR, 6/77 (8%). Only one patient had positive immunohistochemical fi ndings and a positive PCR. The following statistical measures were calculated between PCR and serology: sensitivity, 33.3%; specificity, 88.89%; positive predictive value, 28.57%; negative predictive value, 90.91%; and accuracy, 82.35%. CONCLUSION: The frequency of positive CMV varied among the tests. Serology presented the highest positive frequency. When compared to PCR, the sensitivity and positive predictive value of serology were low.

Numerical simulation of non-Darcian flow through a porous medium

This work reports on fluid flow in a fluid-saturated porous medium, accounting for the boundary and inertial effects in the momentum equation. The flow is simulated by Brinkman-Forchheimer-extended Darcy formulation （DFB）, using MAC （Marker And Cell） and Chorin pressure iteration method. The method is validated by comparison with analytic results. The effect of Reynolds number, Darcy number, porosity and viscosity ratio on velocity is investigated. As a result, it is found that Darcy number has a decisive influence on pressure as well as velocity, and the effect of viscosity ratio on velocity is very strong given the Darcy number. Additional key findings include unreasonable choice of effective viscosity can involve loss of important physical information.

Bidirectional effect of Wnt signaling antagonist DKK1 on the modulation of anthrax toxin uptake

LRP6,a co-receptor for the morphogen Wnt,aids endocytosis of anthrax *** we report that Dickkopf1(DKK1)protein,a secreted LRP6 ligand and antagonist,is also a modulator of anthrax toxin ***-mediated gene silencing or TALEN-mediated gene knockout of DKK1 reduced sensitivity of cells to PA-dependent hybrid ***,unlike the solely inhibitory effect on Wnt signaling,the effects of DKK1 overexpression on anthrax toxicity were bidirectional,depending on its endogenous expression and cell *** microscopy and biochemical analyses showed that DKK1 facilitates internalization of anthrax toxins and their receptors,an event mediated by DKK1-LRP6-Kremen2 *** antibodies against DKK1 provided dose-dependent protection to macrophages from killing by anthrax lethal toxin(LT).Our discovery that DKK1 forms ternary structure with LRP6 and Kremen2 in promoting PA-mediated toxin internalization provides a paradigm for bacterial exploitation of mechanisms that host cells use to internalize signaling proteins.

Electric Modulus Analysis of Carbon Black/Copolymer Composite Materials

We have investigated the electrical properties of carbon black (cb) loaded in ethylene butylacrylate copolymer composite (EBA) in the frequency range between 102 and 104 Hz and temperature range between 153 and 353 K. The frequency dependence of electrical data that have been analyzed in two frameworks: the electrical modulus formalism with the Kohlrausch-Williams-Watts stretched exponential function (KWW) and the electrical conductivity by using the Jonscher’s power law in the frequency domain. The stretching exponent βKWW and n are found to be temperature independent for all cb fractions and to be decreased when the cb volume concentrations loaded in copolymer matrix increases. It is found that the activation energy obtained by the modulus method is in good agreement with that obtained by the DC conductivity in the power law which is independent on the cb contents that exist in the copolymer matrix, suggesting that these particles do not interact significantly with the chain segments of the macromolecules in the EBA copolymer.

Study on the structure and composition of aortic valve calcific deposits:Etiological aspects

The structures and chemical compositions of valve calcific deposits were investigated. The deposits was chosen arbitrarily and subjected to chemical analysis, observation with scanning microscope, semi-quantitative determination of Ca, Mg, Na, K, P and C elements by energy dispersive X-ray, X-ray diffraction and Fourier transform infrared spectroscopy carried out. These deposits were found to have non-uniform internal structures composed of layers of a structureless aspidinic inorganic material, substantial amounts of voluminous organic material and in a few samples small spheres were also present. Two groups of deposits with distinctly different chemical compositions were identified: one group with a low Ca/P molar ratio (1.59) and the other group with a high (1.82) Ca/P molar ratio. The deposits belonging to the group with a low Ca/P molar ratio contain higher concentration of magnesium and consist of increased amount of amorphous calcium phosphate. The deposits with a high Ca/P molar ratio contain low concentration of magnesium and consist predominantly of carbonated hydroxyapatite. The inorganic material was identified as a poorly crystalline carbonate hydroxyapatite containing molecular water of the average formula Ca9.1Mg0.4(Na,K)(PO4)5.8(CO3)0.3(OH)2. The actual chemical composition of the apatitic solid phase varies not only from deposit to deposit but also within the same deposit. The non-uniform internal structure of the deposits, the occasional presence of spherical particles and the variable point composition of the individual deposits indicate that their formation did not proceed under more or less constant conditions.

Urinary bladder cancer(UBC)is a heterogeneous disease with highly variable clinical outcomes and responses to *** some advances in the molecular understanding of UBC,this knowledge still has not been translated to the clinic in terms of improvements in the prognosis and treatment of *** urinary bladder tumor models representative of the human disease in terms of histology and behavior are needed to study factors involved in tumor initiation,progression and ***,accurate model systems would facilitate identification of new therapeutic targets and predictive markers that could lead to optimization of existing therapies and development of new *** established cancer cell lines derived from human urinary bladder tumors representing different grades and stages have been used as experimental models for UBC *** cell lines reflect some of the genetic and morphologic alterations observed in human urothelial carcinoma and serve as simplified models to study the behavior of cancer cells in ***,their translational potential is limited due to the artificial conditions,in which the cells are maintained,grown and *** models offer a more complex and realistic model for the establishment,development,and progression of tumors as well as to evaluate new therapeutic *** the years,the authors'group has worked with several UBC cell lines,established and characterized chemically induced UBC models,and patient-derived xenografts *** this study,the authors will provide a summary of the UBC models developed by their group,analyze their translational potential and weaknesses,and define areas that remain to be explored.
