Light perception at dawn plays a key role in coordinating multiple molecular processes and in entraining the plant circadian *** Arabidopsis mutant lacking the main photoreceptors,however,still shows clock entrainment,indicating that the integration of light into the morning transcriptome is not well *** this study,we performed a high-resolution RNA-sequencing time-series experiment,sampling every 2 min beginning at *** parallel experiments,we perturbed temperature,the circadian clock,photoreceptor signaling,and chloroplast-derived light *** used these data to infer a gene network that describes the gene expression dynamics after light stimulus in the morning,and then validated key *** sampling time points at high density,we are able to identify three light-and temperature-sensitive bursts of transcription factor activity,one of which lasts for only about 8 *** and cryptochrome mutants cause a delay in the transcriptional bursts at dawn,and completely remove a burst of expression in key photomorphogenesis genes(HY5 and BBX family).Our complete network is available online(http://***/∼de656/dawnBurst/***).Taken together,our results show that phytochrome and cryptochrome signaling is required for fine-tuning the dawn transcriptional response to light,but separate pathways can robustly activate much of the program in their absence.

The development of high-resolution nanosized photoacoustic contrast agents is an exciting yet challenging technological advance. Herein, antibody （breast cancer-associated antigen I （Brcaal） monoclonal antibody）- and peptide （RGD）- functionalized gold nanoprisms （AuNprs） were used as a combinatorial methodology for in situ photoacoustic imaging, angiography, and localized hyperthermia using orthotopic and subcutaneous murine gastric carcinoma models. RGD-conjugated PEGylated AuNprs are available for tumor angiography, and Brcaal monodonal antibody-conjugated PEGylated AuNprs are used for targeting and for in situ imaging of gastric carcinoma in orthotopic tumor models. In situ photoacoustic imaging allowed for anatomical and functional imaging at the tumor site. In vivo tumor angiography imaging showed enhancement of the photoacoustic signal in a time-dependent manner. Furthermore, photoacoustic imaging demonstrated that tumor vessels were clearly damaged after localized hyperthermia. This is the first proof-of-concept using two AuNprs probes as highly sensitive contrasts and therapeutic agents for in situ tumor detection and inhibition. These smart antibody/peptide AuNprs can be used as an efficient nanotheranostic platform for in vivo tumor detection with high sensitivity, as well as for tumor targeting therapy which, with a single-dose injection, results in tumor size reduction and increases mice survival after localized hyperthermia treatment.

Analysis and Optimization of Blasting Practices at the Sangaredi Mine

Generally, the Mos hardness of bauxite is 2.5 to 3.5. According to the specific conditions of the Sangaredi bauxite deposit, that is, the rock hardness coefficient is between 3 and 6, and there is a clayey zone in the bauxite mining area, it is necessary to carry out blasting work before mining. This article mainly analyzes and optimizes the blasting practice of the Sangaredi open pit bauxite mine. The subject was finally extended to the reduction of nuisances due to blasting, that is to say essentially due to vibrations: this presupposed a study around the vibration data available, and the proposal of methods to limit these vibrations. The bottom conditions of the Sangaredi bauxite ore zone are studied. Bauxite reserves are very rich and the market is huge. The analysis of mine blasting practices, mainly the types of explosives, the selection and analysis of explosives and blasting equipment and choosing the mode of longitudinal blasting, the link of the blasting site of the mining area, further study the optimization of mining blasting practice, the choice of drilling and process to determine reasonable blasting parameters, which improving the drilling method. The experimental results show that this optimization scheme improves efficiency blasting of the bauxite mine but also promotes an increase in production.

Development of an Algorithm for a Triangular Approach to the Sphere Based on the Platonic Solids Using MATLAB Scripts

The sphere is a common object in uncountable engineering problems, which not only appears in structural elements like domes but also in thousands of mechanisms normally used in diverse kinds of machines. To design, calculate and analyze the behaviour on service of spherical elements, it is essential to have a good method to create an ordered group of discrete points of the spherical surface from the parametric equations commonly used to define the sphere continuously. One of the best known and widely used in high-level programming environment is MATLAB. The programming language has thousands of functions, lots of them specially designed for engineering processes. One of these functions generates a sphere knowing a given radius and shows the result. Nevertheless, this function is really imprecise because it is based on parallels and meridians besides the obtained vertices do not keep a constant distance each other. This causes the fact that it would be appropriate to design a new function to generate accurate discrete approximations of the sphere. The objective of this paper is to create a low-level function in MATLAB to obtain a discrete sphere with high regularity and high approximation in order to provide a good base to solve sphere-based engineering problems. To ensure a perfect symmetry and high regularity platonic bodies, MATLAB will be used as a base to divide the continuous spherical surface in a finite number of regular triangles. The obtained results for the different seed bodies will be represented graphically and compared to each other. The accuracy of each method will be evaluated and compared too.

The therapeutic promise of small interfering RNAs （siRNAs） for specific gene silencing is dependent on the successful delivery of functional siRNAs to the cytoplasm. Their conjugation to an established delivery platform, such as gold nanoparticles, offers tremendous potential for treating diseases and advancing our understanding of cellular processes. Their success or failure is dependent on both the uptake of the nanoparticles into the cells and subsequent intracellular release of the functional siRNA. In this study, utilizing gold nanoparticle siRNA-mediated delivery against C-MYC, we aimed to determine if we could achieve knockdown in a cancer cell line with low levels of intracellular glutathione, and determine the influence, if any, of polyethylene glycol （PEG） ligand density on knockdown, with a view to determining the optimal nanoparticle design to achieve C-MYC knockdown. We demonstrate that, regardless of the PEG density, knockdown in cells with relatively low glutathione levels can be achieved, as well as the possible effect of steric hindrance of PEG on the availability of the siRNA for cleavage in the intracellular environment. Gold nanoparticle uptake was demonstrated via transmission electron microscopy and mass spectroscopy, while knockdown was determined at the protein and physiological levels （cells in S-phase） by in-cell westerns and BrdU incorporation, respectively.

知识图谱以图的形式描述客观世界中实体及其之间的关系,是知识的最佳组织形式。目前知识图谱技术已在智能问答、推荐系统等热点研究领域得到了大规模有效应用。短文本类型数据挖掘也是目前互联网信息数据挖掘的一重要研究分支,论文将知识图谱用于短文本情绪感知分析,是基于知识图谱三元组查询方式带来的推理性能优化。论文主要研究内容与研究成果包括:(1)提出了基于推文图表的情绪评估分析方法,以有效评估短文本中具有积极和消极态度的情绪极性。具体地,论文使用推文图表将知识库表示为图,并提取图的属性,以构造用于机器学习模型的特征矩阵,知识图谱表示学习中图表的相似性则设计为与情感预测相关。为增强评估效果论文删除了与霍恩子句和其他逻辑语句对应的图特征(2)构建了基于Skip-Gram的词向量训练模型,以提升短文本词向量训练高效与准确性。论文词向量模型构建基于主流的Word2Vec模型,为获得更好的训练效果,论文比较了不同的语料库训练方法,并对基于不同相似任务的训练效果进行了评估,结果证明使用Word2Vec的Skip-Gram模型获得的词向量训练效果最佳,选其作为本论文建模方案,论文给出了模型的具体实现。最终实验结果表明,论文所设计的词向量模型,在具有更好的有效性和准确性的基础上,还具有较好的可扩展性,如对于网络流行语,也能通过找到合适的同义词去理解潜在的新词义。(3)设计了基于Bi-LSTM模型的情感分析优化模型,以提高短文本情感分析的效果与效率。利用有用信息而无论信息之间的距离与前后关系是双向长短期记忆Bi-LSTM模型的实质,关键是如何定量利用,为此论文设计了两阶段关注分析模型,通过对输入特征的重要性保持关注、对模型编码器对输入序列某些特征的实时收敛性保持关注,并最终以权值方式反映到输出向量的计算中,来提高信息利用的合理性。论文以Facebook和Twitter文本进行了实验验证,样本大小为600,Facebook和Twitter的输入序列长度为分别235和250,实验结果显示FI分数、精度和召回率分别为0.675、0.876和0.812。由此表明了论文所设计模型的有效性。

AIM: To determine the genotypes in Mexican hepatitis Bvirus (HBV) isolates and characterize their precore andcore promoter ***: Forty-nine HBV isolates of Mexico obtainedfrom sera of 15 hepatitis patients, 6 hemodialysis pa-tients, 20 men seeking HIV testing, and 8 AIDS patientswere analyzed. HBV isolates were amplified by PCR,and genotyped by line probe assay (INNO-LiPA HBVGenotyping; INNOGENETICS N V, Ghent, Belgium).HBV genotype confirmation was performed by DNAsequencing part of the sAg region. Precore and core pro-moter mutation characterization was performed by lineprobe assay (INNO-LiPA HBV PreCore; INNOGENETICS NV, Ghent, Belgium).RESULTS: Overall, HBV genotype H was found in 37(75.5%) out of the 49 isolates studied. HBV genotypesG, A, and D were found in 5 (10.2%), 4 (8.2%), and 3(6.1%) isolates, respectively. HBV genotype H was pre-dominant in isolates from hemodialysis patients (100%),hepatitis patients (80%), and men seeking HIV test-ing (75%), and accounted for half of infections in AIDSpatients (50%). Six (12.2%) out of the 49 HBV isolatesshowed both wild type and mutant populations at pre-core codon 28. These mixed wild type and precore mu-tant populations were observed in one HBV genotype Aisolate and in all HBV genotype G isolates. A dual variantcore promoter mutation was observed in 1 (2%) of theisolates, which was genotype ***: HBV genotype H is highly predominantin HBV isolates of Mexico followed by genotypes G, Aand D. A low frequency of precore and core promotermutations is observed in HBV Mexican isolates.

采煤活动产生的采煤裂隙是采煤对土壤重金属迁移影响最明显的迹象之一。研究和揭示受采煤活动影响的废弃地土壤重金属迁移特征和规律,对矿区生态科学有效管理具有重要的科学意义和实用价值。因此,为了了解采煤裂隙对废弃地重金属空间分布和迁移的影响,从受采煤活动影响的废弃地不同深度的土壤中采集样品。以巨野废弃地裂隙对重金属As、Ni、Cr、Cu、Pb、Zn、Hg的空间分布为研究对象,明确了采煤废弃地裂隙周围土壤重金属元素的总量和可交换态,分析了它们的分布和迁移规律,评价了它们的污染、裂隙周围潜在的生态风险和人体健康风险环境,以及采煤裂隙对土壤理化性质的影响,得出以下结论:除Cu、Pb、Hg总量外,As、Ni、Cr、Zn总量均超过各自的背景浓度。荒地中砷的迁移因子最高,而重金属的迁移因子则随着As＞Ni＞Cu＞Zn＞Cr＞Pb逐渐降低。荒地土壤总量重金属的浓度分布在W-0～W-5之间呈现出相似的分布趋势,而可交换态重金属的浓度分布在W-0～W-5之间呈现出不同的分布趋势。除As（w-0-30和w-0-40）、Ni（w-0-40）和Cu在未污染到中度污染范围外,废弃地土壤中重金属的地球化学累积指数表明,各采样深度土壤基本未受污染。各样点（w-0、w-1、w-2和w-5）各重金属的生态风险平均值均为较低的生态风险,说明采煤活动影响下撂荒地土壤重金属的风险指标具有较低的潜在生态风险。儿童危害指数最高,W-0区可能只存在于儿童（口腔危害指数）。重金属不构成非致癌或致癌作用的重大危险,尽管非致癌和致癌的砷的风险接近不可接受,尤其是对儿童。研究结果表明,在受采煤活动影响的废弃地土壤中,除汞外,As、Ni、Cu、Cr、Pb、Zn等金属含量与土壤含水量呈显著正相关。金属汞含量仅与土壤饱和导水率呈正相关。As、Ni、Cu、Cr、Pb、Zn等金属含量与化学性质（速效钾、速效氮、氧化还原电位和电导率）呈正相关。As、Cu、Cr、Pb、Zn等金属含量与化学性质（有效磷）呈显著负相关。金属As、Ni、Cu、Cr、Pb、Zn的浓度与化学性质（速效钾、速效氮、氧化还原电位和电导率）、物理性质（含水量）的迁移路径相似。重金属元素的分布受钾、氮和电导率的影响。论文中有图31张,表20张,参考文献138篇。
