Pancreatic cancer has a dismal prognosis due to late detection and lack of efficient *** Kirsten rat sarcoma virus(KRAS)oncogene is mutated in up to 90%of all pancreatic ductal adenocarcinomas(PDACs)and constitutes an attractive target for ***,the most common KRAS mutations in PDAC are G12D(44%),G12v(34%)and G12R(20%)that are not amenable to treatment by KRAS G12C-directed cysteine-reactive KRAS inhibitors such as Sotorasib and Adagrasib that exhibit clinical efficacy in lung *** G12C mutant pancreatic cancer has been treated with Sotorasib but this mutation is detected only in 2%–3%of ***,the KRAS G12D-directed MRTX1133 inhibitor has entered clinical trials and more of such inhibitors are in *** other KRAS mutations may be targeted indirectly via inhibition of the cognate guanosine exchange factor(GEF)Son of Sevenless 1 that drives *** agents seem to provide the means to target the most frequent KRAS mutations in PDAC and to improve patient outcomes.

In this paper we will be concerned with the problem -Δu-1/2Δ(a（x）u2)u+v（x）u=f（u）,x∈R2，where v is a potential continuous and f : R → R is a superlinear continuous function with exponential subcritical or exponential critical growth. We use as a main tool the Nehari manifold method in order to show existence of nonnegative solutions and existence of nodal solutions. Our results complement the classical result of solutions for quasilinear Schrodinger equations via the Nehari method due to Jia-quan Liu, Ya-qi Wang and Zhi-Qiang Wang in the sense that in this article we are considering nonlinearity of the exponential type.

Herpes simplex virus-1 (HSv-1) remains a leading cause of viral disease worldwide and is spread by direct contact with infected lesions. There is no vaccine against HSv-1 infections and there remains a need to identify therapeutics that could reduce the spread. In this study various hispolon compounds were analyzed to determine their antiviral potential against HSv-1 infections in cultured vero cells. To determine the effects on infectivity and possible mechanisms of inhibition, the following assays were conducted. In vitro cytotoxicity assays were conducted to determine the effect of the compounds on cell viability and the maximum non-cytotoxic concentrations. Antiviral assays measured cell viability, percent inhibition of infection following treatment with the compounds, and the effect on the viral infection cycle. These effects were visualized using inverted light and fluorescent microscopy. Of the 24 hispolons tested, only hispolon pyrazole-1 (HISP-1) demonstrated antiviral effects. HISP-1 was demonstrated to effect early stages in HSv-1 infection in cultured vero cells (attachment, penetration, and post-penetration). In silico modeling analyses were conducted to analyze the interactions between HISP-1 and viral glycoprotein D (gD). HISP-1 is safe at concentrations tested and is effective in inhibiting infection of HSv-1 in cultured cells. HISP-1 has potential for therapeutic use as an antiviral against HSv-1 infection, could work in synergy with other antivirals that work be a different modality, and could be developed as a component of a topical agent to reduce the spread of HSv-1 infections.

We use a non-relativistic model to study the spectroscopy of a tetraquark composed of [cc]■ in a diquark-antidiquark *** numerically solving the Schr?dinger equation with a Cornell-inspired potential,we separate the four-body problem into three two-body ***-dependent terms(spin-spin,spin-orbit and tensor)are used to describe the splitting structure of the ■ spectrum and are also extended to the interaction between *** experimental data on charmonium states are used to fix the parameters of the model and a satisfactory description of the spectrum is *** find that the spin-dependent interaction is sizable in the diquark-antidiquark system,despite the heavy diquark mass,and also that the diquark has a finite size if treated in the same way as the ■ *** find that the lowest S-wave T 4c tetraquarks might be below their thresholds of spontaneous dissociation into low-lying charmonium pairs,while orbital and radial excitations would be mostly above the corresponding charmonium pair ***,we repeat the calculations without the confining part of the potential and obtain bound diquarks and bound *** might be relevant to the study of exotic charmonium in the quark-gluon *** T 4c states could be investigated in the forthcoming experiments at the LHC and BelleⅡ.
