Prepartum dietary energy intake alters adipose tissue transcriptome profiles during the periparturient period in Holstein dairy cows

Background:The aim of the study was to investigate the effect of energy overfeeding during the dry period on adipose tissue transcriptome profiles during the periparturient period in dairy ***:Fourteen primiparous Holstein cows from a larger cohort receiving a higher-energy diet(1.62 Mcal of net energy for lactation/kg of dry matter;15%crude protein)for ad libitum intake to supply 150%(OVR)or 100%(CTR)of energy requirements from dry off until parturition were *** calving,all cows received the same lactation *** adipose tissue(SAT)biopsies were collected at−14,1,and 14d fromparturition(d)and used for transcriptome profiling using a bovine oligonucleotide *** mining of differentially expressed genes(DEG)between treatments and due to sampling time was performed using the Dynamic Impact Approach(DIA)and Ingenuity Pathway Analysis(IPA).Results:There was a strong effect of over-feeding energy on DEG with 2434(False discovery rate-corrected Ply 340 and 538 at 1 and 14 *** most-impacted and activated pathways in the Kyoto Encyclopedia of Genes and Genomes(KEGG)database that were highlighted by DIA analysis at−14 d in OVR *** included 9 associated with carbohydrate metabolism,with‘Pyruvate metabolism’,‘Glycolysis/gluconeogenesis’,and‘Pentose phosphate pathway’among the *** surprisingly,OVR led to marked activation of lipid metabolism(e.g.‘Fatty acid biosynthesis’and‘Glycerolipid metabolism’).Unexpected metabolic pathways that were activated at−14 d in OVR included several related to metabolism of amino acids(*** chain)and of cofactors and vitamins(thiamin).Among endocrine and immune system pathways,at−14 d OVR led to marked activation of‘PPAR signalling’and‘Antigen processing and presentation’.Among key pathways affected over time in OVR,a number were related to translation(*** signaling),endocrine/immune signaling(CXCR4 and IGF1),and lipid metabolism(oxidative

Transcriptional changes in mesenteric and subcutaneous adipose tissue from Holstein cows in response to plane of dietary energy

Background： Dairy cows can readily overconsume dietary energy during most of the prepartum period, often leading to higher prepartal concentrations of insulin and glucose and excessive body fat deposition. The end result of these physiologic changes is greater adipose tissue lipolysis post-partum coupled with excessive hepatic lipid accumulation and compromised health. Although transcriptional regulation of the adipose response to energy availability is well established in non-ruminants, such regulation in cow adipose tissue depots remains poorly ***： Effects of ad-libitum access to high [HIGH; 1.62 Mcal/kg of dry matter（DM）] or adequate（CON; 1.35 Mcal/kg of DM） dietary energy for 8 wk on mesenteric（MAT） and subcutaneous（SAT） adipose tissue transcript profiles were assessed in non-pregnant non-lactating Holstein dairy cows using a 13,000-sequence annotated bovine oligonucleotide microarray. Statistical analysis revealed 409 and 310 differentially expressed genes（DEG） due to tissue and diet. Bioinformatics analysis was conducted using the Dynamic Impact Approach（DIA） with the KEGG pathway database. Compared with SAT, MAT had more active biological processes related to adipose tissue accumulation（adiponectin secretion） and signs of pro-inflammatory processes due to adipose tissue expansion and macrophage infiltration（generation of ceramides）. Feeding the HIGH diet led to changes in m RNA expression of genes associated with cell hypertrophy（regucalcin）, activation of adipogenesis（phospholipid phosphatase 1）,insulin signaling activation（neuraminidase 1） and angiogenesis（semaphorin 4 G, plexin B1）. Further, inflammation due to HIGH was underscored by m RNA expression changes associated with oxidative stress response（coenzyme Q3, methyltransferase）, ceramide synthesis（N-acylsphingosine amidohydrolase 1）, and insulin signaling（interferon regulatory factor 1, phosphoinositide-3-kinase regulatory subunit 1, retinoic acid receptor alpha）. Background:In Alzheimer disease(AD),hyperphosphorylation of tau proteins results in microtubule destabilization and cytoskeletal *** prior ultra-morphometric studies documented a clear reduction in microtubules in pyramidal neurons in AD compared to controls,however,this reduction did not coincide with the presence of paired helical *** latter suggests the presence of compensatory mechanism(s)that stabilize microtubule dynamics despite the loss of tau binding and *** are composed of tubulin dimers which are subject to posttranslational modifications that affect the stability and function of ***:In this study,we performed a detailed analysis on changes in the posttranslational modifications in tubulin in postmortem human brain tissues from AD patients and age-matched controls by immunoblot and ***:Consistent with our previous study,we found decreased levels ofα-tubulin in AD *** of tubulin with various posttranslational modifications such as polyglutamylation,tyrosination,and detyrosination were also proportionally reduced in AD brain,but,interestingly,there was an increase in the proportion of the acetylatedα-tubulin in the remainingα-*** distribution was changed from predominantly in the processes to be more accumulated in the cell *** number of processes containing polyglutamylated tubulin was well preserved in AD *** there was a cell autonomous detrimental effect of NFTs on tubulin,this is likely a gradual and slow process,and there was no selective loss of acetylated or polyglutamylated tubulin in NFT-bearing ***:Overall,we suggest that the specific changes in tubulin modification in AD brain likely represent a compensatory response.

Potentiality of Vermicompost Humic Acids in Banana in Vitro Micropropagation Clone： Enano Guantanamero

The research was done at Havana Biofabric, Mayabeque Province. The vitro plants of banana （Musa sp） used came from the clone Enano Guantanamero （viand type）. They come from multiplication phase in a Murashige and MS （Skoog medium）. The objective of this work was to study the potentialities of HA （humic acids） for stimulation of rooting and cell division process, searching for completely synthetic hormone substitution due to its high cost. The multiplication medium contained thiamine （2 mg·l^-1）, myo-inositol （100 mg·l^-1）, sugar （30mg·l^-1）. By the use of HA, five treatments were used with total substitution of auxin and cytokinin （TI： HA （10 ***）; （T2： HA （20 mg·l^-1）; （T3： HA （30mg·l^-1）; （T4： HA （40 mg·l^-1）; （T5： HA （50 mg·l^-1） and the control with 6BAP （benzil amino purine）（4 mg·l^-1）＋ IBA （indol butyric acid）（0.65 mg·l^-1）, pH of the medium was adjust just before agar were add. The results showed that in the explants under HA treatments, elongation and cell multiplication were favored confirmed by explants great high, root number and length due to dry mass. The total protein content, reduced carbohydrate and peroxidases enzymatic activity were determine. The use of Vermicompost HA in the in vitro micropropagation of banana allow the elimination of rooting phase saving materials and be able to pass explants directly to Acclimatization phase.

The objective of this work was to describe the predominant cell forms in the phases of growth kinetics in a suspension culture of the Bougainvillea glabra Choisy variety *** in suspension with Murashige and Skoog(MS)basal culture medium were supplemented with six different concentrations and combinations of vegetable growth regulator(VGR)of type auxin(2,4-D and NAA)and cytokinin(BAP)(0.2-5.3 mg/l)and a *** callus was obtained from leaf explants ex vitro to in vitro culture in solid MS medium using PhytagelTM as gelling agent to 2.2 g/l.A portion of callus(1.0 g)was used as cellular inoculum and grown under controlled conditions(50 ml,120 rpm,25°C and luminous intensity of 48μmol m2/s).The best treatment with significant differences(p≤0.05)were with a hormonal relation of 2:1 of BAP and NAA,respectively,with a fresh weight yield ranging from 1.7905 g to 5.8340 g,which represents around 70%.An adaptation phase was observed from day 0 to day 14 on the curve of fresh weight;an exponential phase at day 14 to day 19 and a declination phase at day *** forms in the adaptation phase were elongated cells,a few globular and a few with kidney *** the exponential phase,these cells formed small aggregates of globular *** the death phase,brown,elongated,damaged and fragmented cells were *** this data obtained it is possible to establish the subculture time in fresh medium.
