Discovery and preclinical development of ⅢM-160, a bergenia ciliatabased anti-inflammatory and anti-arthritic botanical drug candidate

Objective: bergenia ciliata(Haw.) Sternb. is used in the Indian traditional system of medicine to treat various ailments including rheumatism and to heal wounds. The objective of the present study was to perform a preclinical characterization of the b. ciliata-based botanical extract ***: ⅢM-160 was chemically standardized and analyzed for heavy metal content, aflatoxins,pesticides and microbial load. The in vitro and in vivo efficacies were determined in suitable models of inflammation, arthritis and nociception. An acute oral toxicity study was performed in Swiss albino mice.A suitable oral formulation was developed and ***: bergenin was found to be the major component(9.1% w/w) of ⅢM-160. The botanical lead displayed inhibition of lipopolysaccharide-induced production of proinflammatory cytokines in THP-1 cells, with selectivity toward interleukin-6(IL-6) and had an excellent safety-window. It showed anti-inflammatory, anti-arthritic and antinociceptive activity in animal models and was not toxic at oral doses up to 2 g/kg in Swiss-albino mice. The gastroretentive, sustained-release capsule formulation showed sustained-release of the bergenin over the period of 24 h, resulting in improved plasma-exposure of bergenin in Sprague–Dawley ***: The dual-activity of IL-6 inhibition and antinociception marks the suitability of ⅢM-160 for treating rheumatoid arthritis. This study will serve as the benchmark for further research on this botanical formulation.

Objective: The current study evaluated various new colchicine analogs for their anticancer activity and to study the primary mechanism of apoptosis and in vivo antitumor activity of the analogs with selective anticancer properties and minimal toxicity to normal ***: Sulforhodamine b(SRb) assay was used to screen various colchicine analogs for their in vitro cytotoxicity. The effect of N-[(7S)-1,2,3-trimethoxy-9-oxo-10-(pyrrolidine-1-yl)5,6,7,9-tetrahydrobenzo[a] heptalene-7-yl] acetamide(ⅢM-067) on clonogenicity, apoptotic induction, and invasiveness of A549 cells was determined using a clonogenic assay, scratch assay, and staining with 4’,6-diamidino-2-phenylindole(DAPI) and annexin V/propidium iodide. Mitochondrial membrane potential(MMP) and reactive oxygen species(ROS) levels were observed using fluorescence microscopy. Western blot analysis was used to quantify expression of proteins involved in apoptosis, cell cycle, and phosphatidylinositol 3-kinase(PI3K)/protein kinase b(AKT)/mammalian target of rapamycin(mTOR) signaling. Pharmacokinetic and in vivo efficacy studies against Ehrlich ascites carcinoma(EAC) and Ehrlich solid tumor models were conducted using Swiss albino ***: ⅢM-067 showed potent cytotoxicity and better selectivity than all other colchicine analogs screened in this study. The selective activity of ⅢM-067 toward A549 cells was higher among other cancer cell lines,with a selectivity index(SI) value of 2.28. ⅢM-067 demonstrated concentration-and time-dependent cytotoxicity against A549 cells with half-maximal inhibitory concentration values of 0.207, 0.150 and0.106 μmol/L at 24, 48 and 72 h, respectively. It also had reduced toxicity to normal cells(SI > 1) than the parent compound colchicine(SI = 1). ⅢM-067 reduced the clonogenic ability of A549 cells in a dose-dependent manner. ⅢM-067 enhanced ROS production from 24.6% at 0.05 μmol/L to 82.1% at 0.4 μmol/L and substantially decreased the MMP(100% in control to 5.6% at 0.4 μmol

Investigation on surface structuring generated by electrochemical micromachining

Surface geometrical features and their function- ality depend on the manufacturing process which is employed for fabrication of surface structures. Maskless electrochemical micromachining （EMM） is used to generate various surface structures for diminishing and controlling friction and wear to increase the lifetime, reliability, and efficiency of mechanical systems. This paper presents a method for the generation of structured surfaces on stainless steel （SS-304） surfaces by using maskless EMM. The micropatterned tool is composed of 800 μm diameter circular holes in a 5 × 5 matrix form. The indigenously developed EMM set up consists of an EMM cell, electrical power supply system, and a controlled vertical cross-flow electrolyte circulation arrangement to control the influence of process parameters during the generation of the micro features of structured surfaces. The single structured cathode tool is used for the mass production of structured surfaces with a short fabrication time in the industrial context by avoiding the use of an individual masking process for each workpiece. The process has been characterized in terms of the effects of predominant process parameters such as machining voltage, electrolyte concentration, duty ratio, pulsed frequency, and machining time on the machined surface characteristics such as current efficiency, machining accuracy, and depth of the circular pattern on the stainless steel surfaces. A mathematical model is also developed to determine the theoretical depth of the dimple pattern and correlate the theoretical depths with actual depths as obtained by experimentation. Moreover, an effort has been made to study the structuringcharacteristics on the basis of micrographs obtained duringthe EMM.

Experimental investigation into fabrication of microfeatures on titanium by electrochemical micromachining

Titanium machining is one of the challenging tasks to modem machining processes. Especially fabricat- ing microfeatures on titanium appear as a potential research interest. Electrochemical micromachining （EMM） is an effective process to generate microfeatures by anodic dis- solution. Machining of titanium by anodic dissolution is different than other metals because of its tendency to form passive oxide layer. The phenomenon of progression of microfeature by conversion of passive oxide layer into transpassive has been investigated with the help of mask- less EMM technique. Suitable range of machining voltage has been established to attain the controlled anodic disso- lution of titanium by converting passive oxide film of titanium into transpassive with nonaqueous electrolyte. The experimental outcomes revealed that the micromachining of titanium with controlled anodic dissolution could be possible even at lower machining voltage in the range of 6-8 V. This work successfully explored the possibility of generation of microfeatures on commercially pure titanium by anodic dissolution process in microscopic domain by demonstrating successful fabrication of various microfea- tures, such as microholes and microcantilevers.

The Narmada Son Lineament(NSL)is a major palaeo-rift system and seismically active intra-plate *** this paper,we processed and analyzed the GPS data from 2009 to 2016 to study the geodynamic characteristics of *** velocities derived from GPS were used to calculate the associated deformation and crustal strain,while the slip deficit was estimated using the horizontal motion of GPS *** investigation reveals that the maximum deformation of western NSL is 1.6 mm per year,and the upper bound of the seismic moment(M;)is 2.0×10;dyn/cm,corresponding to an earthquake of about 6.0 *** study highlights the significance of Aravalli and Satpura mobile *** estimated strain of 0.03μstrain/yr is low but comparable to the stable continental region,and a close association of factors(the fragility,crustal conductors,and compressive tectonics)is considered responsible for seismogenic activities in western NSL.

Novel multistage solid-liquid circulating fluidized bed： Hydrodynamic characteristics

The present work proposes a novel radially cross-flow multistage solid-liquid circulating fluidized bed （SLCFb）. The SLCFb primarily consists of a single multistage column （having an inner diameter of 100 mm and length of 1.40 m）, which is divided into two sections wherein both the steps of utilization or loading （e.g., adsorption and catalytic reaction） and regeneration of the solid phase can be carried out simulta- neously in continuous mode. The hydrodynamic characteristics were studied using ion exchange resin as the solid phase and water as the fluidizing medium. The loading and flooding states were determined for three particle sizes; i.e,. 0.30, 0,42, and 0.61 ram. The effects of the superficial liquid velocity and solid feed rate on the solid hold-up were investigated under loading and flooding conditions. The solid hold-up increases with an increase in the solid feed rate and decreases with an increase in the superficial liquid velocity. An artificial-intelligence formalism, namely the multilayer perceptron neural network （MLPNN）, was employed for the prediction of the solid hold-up. The input space of MLPNN-based model consists of four parameters, representing operating and system parameters of the proposed SLCFb. The developed MLPNN-based model has excellent prediction accuracy and generalization capability.

Galactic transient black hole candidate(bHC)MAXI J1836–194 was discovered on 2011 Aug30,by MAXI/GSC and Swift/*** source activity during this outburst continued for^3 months before entering into the quiescent *** again became active in March 2012 and continued for another^2 *** this paper,3-25 keV RXTE/PCA spectra from the 2011 outburst and 0.5-10.0 keV Swift/XRT data during its 2012 outburst are analyzed with the two-component advective flow(TCAF)model based fits files in *** calculate the X-ray contributions coming from jets/outflow using a newly developed method based on the deviation of the TCAF model *** also study the correlation between observed radio and estimated jet X-ray *** correlation indices(b)are found to be 1.79 and 0.61,when the 7.45 GHz Very Large Array(VLA)radio flux is correlated with the total X-ray and jet X-ray fluxes in 3-25 keV range *** has been found that the jet contributes in X-rays up to a maximum of 86%during its 2011 *** makes the bHC MAXI J1836–194 strongly jet dominated during the initial rising phase.

Characterizing the influence of tide on the physico-chemical parameters and nutrient variability in the coastal surface water of the northern bay of bengal during the winter season

The spatial distribution of physico-chemical parameters（sea surface temperature（SST）, p H, sea surface salinity（SSS）, dissolved oxygen（DO） and Secchi depth） along with filterable nutrients（dissolved inorganic nitrate（DIN）,dissolved inorganic phosphate（DIP） and reactive silicate（DSi）） are measured in the winter months of November,December, January and February for four consecutive years from 2009–2010 to 2012–2013 on the shallow continental shelf（〈20 m bathymetry） of the coastal waters（up to 18 km away from shoreline） of the northern bay of bengal（n bo b） during the highest high tide（HHT） and lowest low tide（LLT） hours for the first time. The variability of the coastal biogeochemical environment is assessed during the HHT and LLT hours and for this purpose, seawater samples are collected from seven different locations of a transect in the coastal region. Physicochemical parameters（except SST） show significant difference in magnitude during the HHT and LLT hours respectively. p H, SSS and DO are found to increase in the HHT hours and vice-versa. The data reveal that during the LLT hours, a relative increase of freshwater input in the n bo b can have elevated the nutrient concentration compared with that observed during the HHT hours. The ratio of nutrient concentration is found to deviate significantly from the Redfield ratio. The abundance of DIP is much higher compared with that of DIN and *** anthropogenic sources of DIP from the upstream flow（especially the domestic effluent of several metropolises） can be mainly attributed behind such an observation. In order to characterize and establish the trend of such variation in such an important bio-climatic region, long-term and systematic ecosystem monitoring in the coastal water of the n bo b northern bay of bengal should be carried out throughout the year.
