＜正＞ We investigated the changes in the binding properties of sedative and non - sedative antihistamines by histamine（HA） - induced intemalization of histamine H1 receptors （H1Rs） in intact human U373 MG astrocytoma cells. Intemalization of H1Rs was induced without their degradation by treatment with 0.1 mM HA for 30 min at 37℃. The binding properties of [3H]mepyramine. a cell - penetrating radioligandfor H1Rs. were not changed by the HA pretreatment. The displacement curves for sedative antihistamines （6 drugs tested） against [3H]mepyramine binding were not changed by the HA pretreatment. In contrast, the displacement analyses for non - sedative antihistamines （5 drugs tested） showed that their affinities for H1Rs were reduced by the HA pretreatment. which was prevented under hy- pertonic conditions where the clathrin - mediated receptor intemalization was inhibited. These results suggest that non - sedative antihistamines have the lower affinities for the intracellular H1Rs than for the cell surface H1Rs. possibly due to their less accessibility through the biomembrane. This intact cell binding assay might provide us a novel method for screening sedative and non - sedative antihistamines in vitro.

AIM: To analyze the relationship between the glycated albumin (GA) to glycated hemoglobin (HbA1c) ratio and the histological grading of liver ***: The study retrospectively included consecutive hepatitis C virus positive chronic liver disease patients (n = 142) who had undergone percutaneous liver biopsy between January 2008 and March 2010 at our institution. The ratios of GA/HbA1c were calculated in all patients to investigate the relationship with the degree of the liver fibrosis. The values of the aspartate aminotransferase-to-platelet ratio index (APRI), an excellent marker for the evaluation of liver fibrosis, were also calculated. In addition, we combined the ratio of GA/HbA1c and the APRI in order to improve our ability to detect the presence of significant liver fibrosis. RESULTS: Sixty-one (43%) patients had either no fibrosis or minimal fibrosis (METAVIR score: F0-F1), while 25 (17%) had intermediate fibrosis (F2). Fifty-six (39%) patients had severe fibrosis (F3-F4) and 27 of them had cirrhosis (F4). The mean values of the GA/HbA1c increased with the progression of the fibrosis (F0-1: 2.83 ± 0.24, F2: 2.85 ± 0.24, F3: 2.92 ± 0.35, F4: 3.14 ± 0.54). In the present study,we show that time-consuming manual tuning of parameters in the Rietveld method,one of the most frequently used crystal structure analysis methods in materials science,can be automated by considering the entire trial-and-error process as a blackbox optimisation *** automation is successfully achieved using Bayesian optimisation,which outperforms both a human expert and an expert-system type automation despite the absence of *** approach stabilises the analysis quality by eliminating human-origin variance and bias,and can be applied to various analysis methods in other areas which also suffer from similar tiresome and unsystematic manual tuning of extrinsic parameters and human-origin variance and bias.

Background: Although Child–Pugh grade A patients with hepatocellular carcinoma(HCC) are candidates for curative resection, some may have a poor prognosis. The albumin-bilirubin(ALBI) grade, a measure of liver function based on albumin and bilirubin, has the potential to detect Child–Pugh grade A HCC patients with poor prognosis. Because components of the ALBI grade can be measured easily even after surgery, we explored the predictive values of ALBI in patient prognosis after HCC resection. Methods: In this retrospective case-control study, we included 136 HCC patients who underwent curative resection between January 2004 and December 2013 at our hospital. ALBI grade was calculated from laboratory data recorded the day before surgery and at post-operative day 5. Results: Pre-and post-operative ALBI grade predicted patients’ long-term outcomes( P = 0.020 and P < 0.001, respectively, for overall survival, and P = 0.012 and P = 0.015, respectively, for recurrence-free survival). Post-operative ALBI grade was associated with patients’ surgical factors of repeated hepatic resection( P = 0.012), intra-operative bleeding( P = 0.006), and surgery duration( P = 0.033). Furthermore, post-operative ALBI grade, rather than pre-operative ALBI grade, was an independent predictive factor of long-term outcome of Child–Pugh grade A patients with HCC. Conclusions: Post-operative ALBI grade is useful to predict the prognosis in patients after HCC resection.

Measurement of Ion Cyclotron Emissions by Using High-Frequency Magnetic Probes in the LHD

Two pairs of high-frequency magnetic probes were installed in the Large Helical Device （LHD）. During the injection of a perpendicular neutral beam, ion cyclotron emissions （ICEs） with the fundamental frequency corresponding to the ion cyclotron frequency at the plasma edge were detected, which are the same type of ICE as measured with the former spare ion cyclotron range of frequencies （ICRF） heating antennas. This type of ICE was further investigated with regard to the phase and intensity of signals. Another type of ICE was found in the LHD, and these ICEs were synchronized with bursts of toroidicity induced Alfv^n eigenmodes （TAE） and the rise of intensity of lost ion flux. Therefore the source of these ICEs was thought to be the particles transferred from the core to the outer region of plasma by the TAE bursts. The frequency of ICEs induced by the TAE bursts increases linearly with the magnetic field strength, since the ion cyclotron frequency increases with the magnetic field strength.

Multilayer polarization elements and their applications to polarimetric studies in 20～400 eV region are mainly reviewed. General principle of selecting material combinations to get high linear polarizance multilayers of reflection type is given with practical examples,with periodic or non-periodic layer structures depending on the usage. Transmission type is introduced as linear polarizer and phase shifter. Their applications include polarization diagnosis of laboratory optical systems and synchrotron radiation beamlines of linear and circular polarization,magnetic rotation experiments such as Faraday rotation and magnetic Kerr rotation on magnetic films and multilayers,and ellipsometry to measure optical constants of thin films precisely. Polarization analysis of soft X-ray fluorescence using multilayer-coated grating is also mentioned. Finally this review is summarized with outlook of further developments.

Plant metabolomics developed as a powerful tool to examine gene functions and to gain deeper insight into the physiology of the plant cell. In this study, we screened Arabidopsis lines overexpressing rice full-length （FL） cDNAs （rice FOX Arabidopsis lines） using a gas chromatography-time-of-flight mass spectrometry （GC-TOF/MS）-based technique to identify rice genes that caused metabolic changes. This screening system allows fast and reliable identification of candi- date lines showing altered metabolite profiles. We performed metabolomic and transcriptomic analysis of a rice FOX Ara- bidopsis line that harbored the FL cDNA of the rice ortholog of the Lateral Organ Boundaries （LOB） Domain （LBD）/ Asymmetric Leaves2-1ike （ASL） gene of Arabidopsis, At-LBD37/ASL39. The investigated rice FOX Arabidopsis line showed prominent changes in the levels of metabolites related to nitrogen metabolism. The transcriptomic data as well as the results from the metabolite analysis of the Arabidopsis At-LBD37/ASL39-overexpressor plants were consistent with these findings. Furthermore, the metabolomic and transcriptomic analysis of the Os-LBD37/ASL39-overexpressing rice plants indicated that Os-LBD37/ASL39 is associated with processes related to nitrogen metabolism in rice. Thus, the combination of a metabolomics-based screening method and a gain-of-function approach is useful for rapid characterization of novel genes in both Arabidopsis and rice.

AIM： To develop the effective technology for reconstruction of a liver organ in vitro using a bio-artificial liver. METHODS： We previously reported that a radial-flow bioreactor （RFB） could provide a three-dimensional highdensity culture system. We presently reconstructed the liver organoid using a functional human hepatocellular carcinoma cell line （FLC-5） as hepatocytes together with mouse immortalized sinusoidal endothelial cell （SEC） line M1 and mouse immortalized hepatic stellate cell （HSC） line A7 as non parenchymal cells in the RFB. Two × 10^7 FLC-5 cells were incubated in the RFB. After 5 d, 2 × 10^7 A7 cells were added in a similar manner followed by another addition of 10^7 M1 cells 5 d later. After three days of perfusion, some cellulose beads with the adherent cells were harvested. The last incubation period included perfusion with 200 nmol/L swinholide A for 2 h and then the remaining cellulose beads along with adherent cells were harvested from the RFB. The cell morphology was observed by transmission electron microscopy （TEM） and scanning electron microscopy （SEM）. To assess hepatocyte function, we compared mRNA expression for urea cycle enzymes as well as albumin synthesis by FLC-5 in monolayer cultures compared to those of single-type cultures and cocultures in the RFB. RESULTS： By transmission electron microscopy, FLC-5, M1, and A7 were arranged in relation to the perfusion side in a liver-like organization. Structures resembling bile canaliculi were seen between FCL-5 cells. Scanning electron microscopy demonstrated fenestrae on SEC surfaces. The number of vesiculo-vacuolar organelles （WO） and fenestrae increased when we introduced the actin-binding agent swinholide-A in the RFB for 2h. With respect to liver function, urea was found in the medium, and expression of mRNAs encoding arginosuccinate synthetase and arginase increased when the three cell types were cocultured in the RFB. However, albumin synthesis decreased. CONCLUSION： Co-culture
