Vast grasslands are found in the walnut-fruit forest region of southern Kyrgyzstan,Middle *** above the worldwide unique walnutfruit forests and used for grazing,they play a pivotal role in the mixed mountain agriculture of local ***,these pastures are subject to an increasing utilization pressure reflecting the changing political and social conditions in the transformation process from a Soviet republic to an independent state.A first detailed analysis of mountain pasture vegetation in the Ferghana Range answers the following questions:What are the main plant community types among Kyrgyzstan's mountain pastures? What are the main environmental gradients that shape their species composition? Which phytogeographical distribution types are predominant? How does grazing affect community composition and species richness in these grasslands? Cellular homeostasis requires a tightly controlled balance between protein synthesis, folding and degradation. Especially long-lived, post-mitotic cells such as neurons depend on an efficient proteostasis system to maintain cellular health over decades. Thus, a functional decline of processes contributing to protein degradation such as autophagy and general lysosomal proteolytic capacity is connected to several age-associated neurodegenerative disorders, including Parkinson＇s, Alzheimer＇s and Huntington＇s diseases. These so called proteinopathies are characterized by the accumulation and misfolding of distinct proteins, subsequently driving cellular demise. We recently linked efficient lysosomal protein breakdown via the protease cathep- sin D to the Ca2＋/calmodulin-dependent phosphatase calcineurin. In a yeast model for Parkinson＇s disease, functional calcineurin was required for proper trafficking of cathepsin D to the lysosome and for recycling of its endosomal sorting receptor to allow further rounds of shuttling. Here, we discuss these findings in relation to present knowledge about the involvement of cathepsin D in proteinopathies in general and a possible connection between this protease, calcineurin signalling and endosomal sorting in particular. As dysregulation of Ca2＋ homeostasis as well as lysosomal impairment is connected to a plethora of neurode- generative disorders, this novel interplay might very well impact pathologies beyond Parkinson＇s disease.

Awareness during emergence from anesthesia: Features and future research directions

The anesthesia awareness with recall(AAWR) phenomenon represents a complication of general anesthesia consisting of memorization of intraoperative events reported by the patient immediately after the end of surgery or at a variable distance from it. Approximately 20% of AAWR cases occur during emergence from anesthesia. Clinically, these unexpected experiences are often associated with distress especially due to a sense of paralysis. Indeed, although AAWR at the emergence has multiple causes, in the majority of cases the complication develops when the anesthesia plan is too early lightened at the end of anesthesia and there is a lack of use, or misuse, of neuromuscular monitoring with improper management of the neuromuscular block. Because the distress caused by the sense of paralysis represents an important predictor for the development of severe psychological complications, the knowledge of the phenomenon, and the possible strategies for its prophylaxis are aspects of considerable importance. Nevertheless, a limited percentage of episodes of AAWR cannot be prevented. This paradox holds also during the emergence phase of anesthesia which represents a very complex neurophysiological process with many aspects yet to be clarified.

Concentrations of disinfection by-products in swimming pool following modifications of the water treatment process:An exploratory study

The formation and concentration of disinfection by-products（DBPs） in pool water and the ambient air vary according to the type of water treatment process used. This exploratory study was aimed at investigating the short-term impact of modifications of the water treatment process on traditional DBP levels（e.g., trihalomethanes（THMs）, chloramines） and emerging DBPs（e.g., Halonitromethanes, Haloketones, NDMA） in swimming pool water and/or air. A sampling program was carried to understand the impact of the following changes made successively to the standard water treatment process： activation of ultraviolet（UV）photoreactor, halt of air stripping with continuation of air extraction from the buffer tank,halt of air stripping and suppression of air extraction from the buffer tank, suppression of the polyaluminium silicate sulfate（PASS） coagulant. UV caused a high increase of Halonitromethanes（8.4 fold）, Haloketones（2.1 fold）, and THMs in the water（1.7 fold） and, of THMs in the air（1.6 fold） and contributed to reducing the level of chloramines in the air（1.6fold） and NDMA in the water（2.1 fold）. The results highlight the positive impact of air stripping in reducing volatile contaminants. The PASS did not change the presence of DBPs, except for the THMs, which decrease slightly with the use of this coagulant. This study shows that modifications affecting the water treatment process can rapidly produce important and variable impacts on DBP levels in water and air and suggests that implementation of any water treatment process to reduce DBP levels should take into account the specific context of each swimming pool.

Neuroendocrine and squamous colonic composite carcinoma:Case report with molecular analysis

Composite colorectal carcinomas are rare. There are amodest number of cases in the medical literature,with even fewer cases describing composite carcinoma with neuroendocrine and squamous components. There areto our knowledge no reports of composite carcinomamolecular alterations. We present a case of composite carcinoma of the splenic flexure in a 33 year-old Cau-casian male to investigate the presence and prognos-tic significance of molecular alterations in rare coloniccarcinoma subtypes. Formalin-fixed paraffin-embedded(FFPE) tissue was hematoxylin and eosin- and mucicar-mine-stained according to protocol,and immuno-stained with cytokeratin(CK) 7,CK20,CDX2,AE1/AE3,chromo-granin-A and synaptophysin. DNA was extracted from FFPE tissues and molecular analyses were performed according to lab-developed methods,followed by capil-lary electrophoresis. Hematoxylin and eosin staining showed admixed neuroendocrine and keratinized squamous cells. Positive nuclear CDX2 expression confirme dintestinal derivation. CK7 and CK20 were *** cells stained positively for synaptophysin and AE1/AE3 and negatively for chromogranin and mucicarmine. Hepatic metastases showed a similar immunohistochemical profile. Molecular analysis revealeda G13D KRAS mutation. BRAF mutational testing wasnegative and microsatellite instability was not *** patient had rapid disease progression on chemotherapy and died 60 d after presentation. Although theG13D KRAS mutation normally predicts an intermed iateoutcome,the aggressive tumor behavior suggests other modifying factors in rare types of colonic carcinomas.

绩效考核是组织人力资源优化发展的一项基本活动,也是衡量员工工作绩效的行之有效的基本工具之一。绩效考核的实施与组织特定的发展环境、文化背景等因素紧密相关,其考核结果的有效性是组织实施工资、津贴等物质激励,以及员工发展、培训等精神激励的依据与前提,最终将影响一个组织对其员工的激励效果。同时,开展员工的绩效管理也是管理者的一项重要职能。对员工实施绩效考核,旨在设立对个人和组织发展目标的预期,并通过考核的过程,评价员工对组织的贡献度,同时帮助员工发现自身的优势和问题,进而为推动员工的个人发展提供依据。绩效考核的有效性有赖于管理者与员工对组织目标的认知,以及为实现目标而制定的具体方案和实施的努力。马达加斯加反腐败局(BIANCO)是马达加斯加的一个独立的反腐败组织,其使命是预防腐败,帮助公民了解腐败的有害影响,发展公众支持,以及有效执行反腐败法等。BIANCO服务于全社会,并为经济发展、加强政府治理和建立法治做出贡献,其目标是大幅减少腐败,使马达加斯加变得更为公平和繁荣,以实现该国的可持续发展与和谐发展。本论文以BIANCO为研宄对象,通过问卷调查与访谈的方式,对该组织的绩效考核系统实施评价,进而为完善BIANCO的员工绩效考核体系提出建议。论文研宄的具体目标包括:-一是评价组织员工对BIANCO绩效考核体系的看法,发现影响员工对绩效考核体系满意程度的因素;二是考察绩效考核的目标和有效性;三是发现BIANCO现行的员工考核体系所存在的问题;四是提出针对性建议,改进BIANCO的绩效考核体系。本研究在对己有文献进行梳理的基础上,确定研宂对象和抽样程序,实施数据收4,并主要通过对数据的定性分析与定量分析开展具体研究。定性分析过程中主要使用描述性调查,即对所面临的不同因素、不同方面现状实施调査研宄,对有关资料数据的采集和记录更侧重于对客观事实的静态描述,其数据通常通过问卷调查、访谈或观察收集。本论文研宄过程中对BIANCO的207名工作人员发放了问卷,回收有效问卷111份,同时实施访谈,就BIANCO不同部门员工对组织绩效考核的认知进行了研究。本研究采用的问卷主要由两部分构成,一是有关员工服务年限、年龄、性别的信息,这有助于分析不同类群员工对组织绩效考核的观点差异。二是员工对组织绩效考核的具体评价,例如询问受访者是否有参与绩效考核,组织实施绩效考核的频率,以及组织实施绩效考核的必要性等。这部分内容主要是一个与BIANCO绩效考核有效性相关的Likert量表。员工在在回答问卷时需要依据自身感受对相关描述进行打分,由此构成本研究的主要数据来源。本研宂在对原始数据进行整理的基础上,利用SPSS软件对相关数据进行分析,具体考察影响员工绩效考核体系满意度的因素,并提出具有针对性的改进建议。本论文共分为六章。第一章包括研宄的背景、问题陈述、研宄意义和目标。第二章为理论基础与文献综述。第三章为研宄设计和研宄方法。第四章对BIANCO进行了介绍,包括其使命、资源、员工构成等内容。第五章主要为问卷调查的主要结果。第六章为研究结.论,并提出相关改进建议。问卷调査的结果显示:第一,BIANCO能够开展员工的绩效考核。大多数员工表示,他们每年至少接受一次绩效评估。第二,BIANCO的绩效考核在一定程度上是有意义的。大多数员工或多或少都认为绩效考核是有意义的,但也有部分员工不认可绩效考核的价值。因此,我们认为,BIANCO有必要对所有员工解释绩效考核的重要性,这是组织实施有效激励的前提。第三,绩效考核的工具使用不够合理。超过半数的受访者认为,目前的评估方法十分复杂且效率低下。第四,考核体系的有效性存疑。受访者对实际的绩效考核体系存在不同看法。部分员工认为,这个体系是有效的,能够促进组织目标实现。但也有部分员工认为,现有系统效率低下、操作复杂,引发一些负面影响。我们认为,如果员工不认同绩效考核体系,那么该系统的实施效果将十分有限,不能实现预期的评估、激励并开发员工潜力的功能。第五,现有绩效考核体系的设置目标不够明确。根据调查结果,并非所有的BIANCO员工都清楚该体系的作用与目标。对于部分员工而言,他们只能部分了解考核体系设置的目标,尤其是有关奖励与晋升目标的设立,多数员工表示并不清楚。第六,多数受访者对绩效考核给出了积极评价,但也有部分受访者表示无法做出评价或持保留意见。大部分员工表示,他们参与制定了自身绩效考核的目标,这一点值得肯定。但也有员工反映,其薪酬、福利和晋升等激励性内容的确定并非由绩效考核的结果来决定,其间还掺杂了一些影响因素。而且员工认为,BIANCO绩效考核目标的制定更多反映了组织发展的需求,员工个人的发展需求并没有被很好的考虑与反映。除了Likert量表之外,问卷还设计了一些开放式问题,以考察BIANCO员工对组织绩效考核的实际想法。员工的反馈表明,

Wrong place at the wrong time: how retrograde trafficking drives cancer metastasis through receptor mislocalization

Retrograde trafficking is a well-regulated, multi-component pathway that can result in endosomal trafficking to the trans-Golgi network, the perinuclear space, or the nucleus. Either clathrin or the retromer complex can travel with proteins endocytosed from the plasma membrane, guided by Rabs (including 5, 6, 7, 9, 22A), interacting with a host of sorting nexin proteins, and fusing with Golgi-specific anchors to allow transport of activated receptor tyrosine kinases to a potential end within the nucleus. Amplification in these constituents is common in cancer, leading to increased retrotranslocation and a reduction in degradation of receptor tyrosine kinases, an event highly associated with cancer metastasis. Here, we review the role of retrograde trafficking in altering transmembrane receptor localization and activity and the relationship to metastasis, focusing on all four members of the ErbB family, with comparison to other receptor tyrosine kinases including the insulin receptor and fibroblast growth factor receptor, as well as other transmembrane proteins dysregulated in metastasis. By examining how these receptors are being alternatively trafficked and the cancer-associated events resulting from this process, we hope to identify novel therapeutic targets.
