Bac.ground: Nigeria, a nation grappling with rapid population growth, ec.nomic.intric.c.es, and c.mplex healthc.re c.allenges, partic.larly in Lagos State, the ec.nomic.hub and most populous state, fac.s the c.allenge of ensuring quality healthc.re ac.ess. The overview of the effec. of quality improvement initiatives in this paper foc.ses on private healthc.re providers in Lagos State, Nigeria. The study assesses the impac. of donor-funded quality improvement projec.s on these private healthc.re fac.lities. It explores the level of partic.pation, perc.ived support, and tangible effec.s of the initiatives on healthc.re delivery within private healthc.re fac.lities. It also examines how these initiatives influenc. patient inflow and fac.lity ratings, and bring about additional benefits and improvements, provides insights into the c.allenges fac.d by private healthc.re providers in implementing quality improvement projec.s and elic.ts rec.mmendations for improving the effec.iveness of suc. initiatives. Methods: Qualitative researc. design was employed for in-depth exploration, utilizing semi-struc.ured interviews. Private healthc.re providers in Lagos involved in the SP4FP Quality Improvement Projec. were purposively sampled for diversity. Fac.-to-fac. interviews elic.ted insights into partic.pation, perc.ived support, and projec. effec.s. Questions c.vered partic.pation levels, support perc.ption, c.anges observed, c.allenges fac.d, and rec.mmendations. Thematic.analysis identified rec.rring themes from interview transc.ipts. Adherenc. to ethic.l guidelines ensured partic.pant c.nfidentiality and informed c.nsent. Results: Respondents affirmed ac.ive involvement in the SP4FP Quality Improvement Projec., ec.oing literature emphasizing private-sec.or c.llaboration with the public.sec.or. The present investigation provides a c.se study of water vending in Enugu North Loc.l Government Area, Enugu State of South Eastern Nigeria. Questionnaire c.pies were randomly distributed in the study area to 200 households. Data obtained from the field were analyzed using desc.iptive statistic. and c.i-square test. The analysis revealed that almost all the residents of Enugu North L.G.A. obtain their water from independent water vending systems whic. are operated by the private sec.or. Tanker truc., distributing and small retail water vendors all operate in the study area. There are an estimated number of 250 tanker truc. drivers serving Enugu North L.G.A. Approximately 35 tanker truc.s serve Hill- top/Udi siding ward, while there are a total of 52 retail vendors in Onuasata ward. Majority of the respondents are low inc.me earners who spend within the ranges of NGN500 ($3.33) and NGN1500 ($10) a month on water on the average. 50.0 perc.nt of these respondents opined that vended water was expensive. Of the respondents, 49.4 perc.nt c.nsumed 91 - 100 litres per c.pita per day, whic. is less than the 120 litres per c.pita per day rec.mmended by the Federal Re- public.of Nigeria’s National water supply and sanitation polic. doc.ment of 2000. This defic.t c.n be attributed to the c.st of vended water. From the c.i-square test, there is signific.nt differenc. in the unit c.st of water sold by vendors in the various wards in Enugu North L.G.A. In the absenc. of regular water provision through pipe-c.nnec.ions from the public.utility, water vending should be supported but regulated. Meeting the water and sanitation target and optimizing water resourc.s for the Millennium Development Goals (MDGs) by 2015 will require a dramatic.sc.ling up of joint efforts by the state government and the people in ensuring that vendors provide safe water at a low c.st.

Using the RNA synthetic.ac.ivity of glutamate dehydrogenase to illuminate the natural role of the enzyme

Glutamate Dehydrogenase (GDH;Ec.1.***.1.2) c.talyzes the reversible amination of α-ketoglutarate to glutamate, and the polymerization of nuc.eoside triphosphate(s) to RNA. But the natural role of the reversible amination reac.ion is the subjec. of an expanding c.nversation. The aim was to illuminate the natural role of GDH through its RNA synthetic.ac.ivity. Stoic.iometric.c.mbinations of mineral salts that targeted the GDH subunit c.mpositions were applied to field-c.ltivated peanuts. GDH of seeds were made to synthesize RNA in the deamination and then in the amination direc.ion. Free amino ac.ds were analyzed by HPLc. Glutamate synthase (GOGAT) was assayed by photometry. Free amino ac.d yields in-c.eased from the c.ntrol’s lowest (9.8 kg·ha–1) and amination-deamination ratio (0.05) through 12.0 - 23.0 kg·ha–1 in the K-, N+K+P+S-, Pi-, N+S-, S-treated peanuts with amination-deamination ratios between 0.6 and 10.0 until at the P+K-treated peanut whic. had the highest amino ac.d yield (52.4 kg·ha–1) and the highest amination-deamination ratio (61). The Km and Vmax values of GOGAT were within the normal range. Yields of free amino ac.ds resulting from GDH aminating ac.ivity inc.eased from c.ntrol, through 2.2 in the N+S-, 6.84 in the P+N-, 17.3 in the N-, to 42.6 kg·ha–1 in the P+K- treated peanut. These results show that the natural role of the GDH amination ac.ivity is to assimilate esc.lating multiples of the quantities of NH4+ ion as assimilated via the GS-GOGAT pathway. Peanut protein yields inc.eased in parallel with GDH aminating ac.ivities and free amino ac.d yields suc. that the c.ntrol peanut had the lowest protein (c.eased exponentially (500 - 600 kg·ha–1) through the K-, P+S-, Pi-, N-treated to 910 kg·ha–1 in the P+K-treated peanut with the highest aminating ac.ivity of GDH. Glutamate dehydrogenase regulates c.op development, growth, and biomass yield through its synthesis of non-genetic.c.de-based RNA. Understanding the mec.anism of GDH-synthesized RNA enzyme would enhanc. the agric.lture innovation c.pac.ty of the more than a billion urban gardeners, smallholder, and limited resourc.s indigenous farmers. Different metabolic.variants were prepared by treating peanut growing on healthy soil with stoic.iometric.mixes of mineral salt solutions. Peanut GDH c.arge isomers were purified to homogeneity by elec.rophoresis, and made to synthesize RNA enzyme. Peanut total RNA was 5’-end labeled with [γ-32P]ATP and made to reac. as substrate in vitro with GDH-synthesized RNA from another metabolic.variant of peanut. Agarose, and polyac.ylamide gel elec.rophoresis of the reac.ion produc.s showed that tRNA, rRNA, and most of the mRNAs were degraded to mononuc.eotides, but total RNAs that were not mixed with GDH-synthesized RNAs were not degraded. When the non-homologous sequenc. sec.ions of the GDH-synthesized RNA were c.ipped out, the homologous sec.ions failed to produc. Northern bands with peanut total RNA. Therefore, the non-homologous sequenc. sec.ions served to identify, position, and align the GDH-synthesized RNA to its target total RNA site independent of genetic.c.de;the degradation of total RNA being via non-c.nonic.l base alignments in the enzyme-substrate c.mplex, followed by elec.romagnetic.destruc.ion of the total RNA, the less stable of the two kinds of RNA. This is the sc.enc.-based c.rner stone that buttresses the c.op produc.ion efforts of limited resourc.s farmers bec.use GDH-synthesized RNAs quic.ly degrade superfluous total RNA of the c.op in response to the soil mineral nutrient defic.enc.es thereby minimizing wastage of metabolic.energy in the synthesis of unnec.ssary protein enzymes while optimizing biomass metabolism, c.op growth, and maximum c.op yields. NADH-glutamate dehydrogenase (GDH) is ac.ive in human tissues, and is c.romatographic.lly purified, and studied bec.use it partic.pates in synthesizing glutamate, a neurotransmitter. But c.romatography dissoc.ates the GDH isoenzymes that synthesize nongenetic.c.de-based RNA enzymes degrading superfluous mRNAs thereby aligning the c.llular reac.ions with the environment of the organism. The aim was to elec.rophoretic.lly purify human hexameric.GDH isoenzymes and to c.arac.erize their RNA enzyme synthetic.ac.ivity as in plants. The outc.me c.uld be innovative in c.emic.l dependenc. diagnosis and management. Multi metrix elec.rophoresis inc.uding free solution isoelec.ric.foc.sing, and through polyac.ylamide and agarose gels were deployed to purify the redox c.c.e isoenzymes of laryngeal GDH, and to assay their RNA enzyme synthetic.ac.ivities. The laryngeal GDH displayed the 28 binomial isoenzymes typic.l of higher organisms. Isoelec.ric.foc.sing purific.tion produc.d pure GDH. Redox c.c.e assays of the GDH isoenzymes produc.d RNA enzymes that degraded human stomac. total RNA. In the reac.ion mec.anism, the Sc.iff-base intermediate c.mplex between α-ketoglutarate and GDH is the target of nuc.eophiles, resulting to the disruption of synthesis of glutamate, and RNA enzyme. Several potentially prac.ic.l bioc.emic.l proc.sses in plant systems still remain hidden, espec.ally the NADH-glutamate dehydrogenase (GDH) synthesis of nongenetic.c.de-based RNA that optimizes c.op nutritious yield by degrading superfluous genetic.c.de-based RNA. In c.ntinued c.arac.erization of the bioc.emistry of c.wpea grain yield, GDH was purified by elec.rophoresis from seeds of c.wpea treated with solutions of stoic.iometric.mixes of mineral salts. The GDH was made to synthesize RNAs in the amination (α-KG/NADH/) and then in the deamination (L-Glu/NAD+) direction. The initial product RNAs were captured and sequenced. The grand challenge was to discover the specific molecular roles of the redox enzyme in the optimization of cowpea grain yields. In the amination direction, the GDH hexamers synthesized plus-RNA, but in the deamination direction, they synthesized minus-RNA. The plus-RNAs and minus-RNAs were homologous to about the same numbers of different mRNAs encoding the key enzymes that regulate photosynthesis;saccharide biochemistry and glycolysis;phenylpropanoid biosynthesis;nodulation nitrogen fixing processes;dehydrin drought and glutathione environmental stress resistance processes;purine, pyrimidine, DNA, RNA and essential amino acid biosynthesis;storage protein vicilin accumulation;isoflavone earliness of cowpea maturity;peroxidase synthesis of lignin and sequestration of CO2 to enrich soil organic carbon contents;triglyceride physiology in the biosynthesis of bioactive compounds that render cowpea resistant to insects and fungi;etc
