In addition to susceptibility to infections,conventional primary immunodeficiency disorders(PIDs)and inborn errors of immunity(IeI)can cause immune dysregulation,manifesting as lymphoproliferative and/or autoimmune *** can be the prominent phenotype of PIDs and commonly includes cytopenias and rheumatological diseases,such as arthritis,systemic lupus erythematosus(sLe),and sjogren’s syndrome(sjs).Recent advances in understanding the genetic basis of systemic autoimmune diseases and PIDs suggest an at least partially shared genetic background and therefore common pathogenic ***,we explore the interconnected pathogenic pathways of autoimmunity and primary immunodeficiency,highlighting the mechanisms breaking the different layers of immune tolerance to self-antigens in selected IeI.

The use of computer animation in behavioral research is a state-of-the-art method for designing andpresenting animated animals to live test animals. The major advantages of computer animations are：（1） the creation of animated animal stimuli with high variability of morphology and even behavior; （2）animated stimuli provide highly standardized, controlled and repeatable testing procedures; and （3）they allow a reduction in the number of live test animals regarding the 3Rs principle. But the use of ani-mated animals should be attended by a thorough validation for each test species to verify that behaviormeasured with live animals toward virtual animals can also be expected with natural stimuli. Here wepresent results on the validation of a custom-made simulation for animated 3D sailfin mollies Poecilialatipinna and show that responses of live test females were as strong to an animated fish as to a videoor a live male fish. Movement of an animated stimulus was important but female response was stron-ger toward a swimming 3D fish stimulus than to a ＂swimming＂ box. Moreover, male test fish wereable to discriminate between animated male and female stimuli; hence, rendering the animated 3D fisha useful tool in mate-choice experiments with sailfin mollies.

Magnesium as biodegradable biomaterial could serve as bone augmentation material in implant *** knowledge about the predictability of the biodegradation process is essential as this process needs to go hand in hand with the formation of new bone to gradually replace the augmentation ***,this work aimed to assess if the electrochemistry(eC)measurements of the corrosion process correlate with the surface features at various time points during the surface degradation,in order to describe the degradation process of Mg and Mg alloys more reliably,under the assumption that differences in eC behavior can be detected and related to specific patterns on the *** this test setup,a special optical chamber was used for electrochemical measurements on Mg and Mg-alloys(Mg2Ag,Mg4Ag,and Mg6Ag).specimens were investigated using different circulating cell culture solutions as electrolytes,these were minimum essential medium(MeM),Hank’s Balanced salt solution(HBss),and MeM+(MeM with added sodium hydrogen carbonate)at 37℃.Open circuit potential measurements(OCP)over 30 min followed by cyclic polarization were *** electrochemistry data,including OCP,exchange current density and corrosion potential,were compared with visible changes at the surface during these treatments over *** results show that the addition of silver(Ag)leads to a“standardization”of the degradation regardless of the selected test *** is currently difficult to correlate the visible microscopic changes with the data taken from the ***,further investigations are ne.es.ary.

Aging is a complex process that can be characterized by functional and cognitive decline in an individual. Aging can be assessed based on the functional capacity of vital organs and their intricate interactions with one another. Thus, the nature of aging can be described by focusing on a specific organ and an individual itself. However, to fully understand the complexity of aging,one must investigate not only a single tissue or biological process but also its complex interplay and interdependencies with other biological processes. Here, using RNA-seq, we monitored changes in the transcriptome during aging in four tissues(including brain, blood, skin and liver) in mice at9 months, 15 months, and 24 months, with a final evaluation at the very old age of 30 *** identified several ge.es.and processes that were differentially regulated during aging in both tissue-dependent and tissue-independent manners. Most importantly, we found that the electron transport chain(eTC) of mitochondria was similarly affected at the transcriptome level in the four tissues during the aging process. We also identified the liver as the tissue showing the largest variety of differentially expressed ge.es.DeGs) over time. Lcn2(Lipocalin-2) was found to be similarly regulated among all tissues, and its effect on longevity and survival was validated using its orthologue in Caenorhabditis elegans. Our study demonstrated that the molecular processes of aging are relatively subtle in their progress, and the aging process of every tissue depends on the tissue’s specialized function and environment. Hence, individual gene or process alone cannot be described as the key of aging in the whole organism.

The successful application of magnesium(Mg)alloys as biodegradable bone substitutes for critical-sized defects may be comprised by their high degradation rate resulting in a loss of mechanical *** study investigates the degradation pattern of an open-porous fluoride-coated Mg-based scaffold immersed in circulating Hanks’Balanced salt solution(HBss)with and without in situ cyclic compression(30 N/1 Hz).The changes in morphological and mechanical properties have been studied by combining in situ high-resolution X-ray computed tomography mechanics and digital volume *** in situ cyclic compression induced acceleration of the corrosion rate,probably due to local disruption of the coating layer where fatigue microcracks were formed,no critical failures in the overall scaffold were observed,indicating that the mechanical integrity of the Mg scaffolds was *** changes,due to the accumulation of corrosion debris between the scaffold fibres,resulted in a significant increase(pe material volume fraction from 0.52±0.07 to 0.47±0.03 after 14 days of ***,despite an increase in fibre material loss,the accumulated corrosion products appear to have led to an increase in Young’s modulus after 14 days as well as lower third principal strain(εp3)accumulation(-91000±6361μεand-60093±2414μεafter 2 and 14 days,respectively).Therefore,this innovative Mg scaffold design and composition provide a bone replacement,capable of sustaining mechanical loads in situ during the postoperative phase allowing new bone formation to be initially supported as the scaffold resorbs.

Rapid technical advances in the field of computeropened new avenues in animal behavior *** （CA） and virtual reality （VR） haveAnimated stimuli are powerful tools as theyoffer standardization, repeatability, and complete control over the stimulus presented, thereby＂reducing＂ and ＂replacing＂ the animals used, and ＂refining＂ the experimental design in line withthe 3Rs. However, appropriate use of these technologies raises conceptual and technical *** this review, we offer guidelines for common technical and conceptual considerations related tothe use of animated stimuli in animal behavior research. Following the steps required to create ananimated stimulus, we discuss （I） the creation, （11） the presentation, and （111） the validation of CAsand VRs. Although our review is geared toward computer-graphically designed stimuli, consider-ations on presentation and validation also apply to video playbacks. CA and VR allow both new be-havioral questions to be addressed and existing questions to be addressed in new ways, thus weexpect a rich future for these methods in both ultimate and proximate studies of animal behavior.

Aims: Assessment of N- terminal brain natriuretic peptide(NTBNP) as a screening tool for heart failure in patients with a permanent pacemaker. Methods and results: Consecutive patients undergoing a routine permanent pacemaker assessment were enrolled. Patients underwent medical history and examination, echocardiography and blood sampling for NT- BNP. Analysis was performed on 261 patients(132 DDD, 121 VVI, eight others), mean age 73± 12 years, range 34- 99 years. seventy two subjects(27% ) had heart failure as defined by left ventricular ejection fraction(LVeF) ≤ 40% and symptoms of heart failure(NYHA class II, III, or IV). screening with NT- BNP gave a sensitivity of 73% and specificity of 72% for detecting heart failure in all patients [area under the curve(AUC) 0.76, Ps increased in subjects with a DDD type pacemaker(sensitivity 80% , specificity 66% , AUC=0.8, CI 0.7- 0.90) and reduced in subjects with a VVI type pacemaker(sensitivity 66% , specificity 61% , AUC 0.68 CI 0.57- 0.78). Conclusion: symptoms of heart failure are common in patients with pacemakers. screening with NT- BNP is feasible and assists in the detection of important cardiac co- morbidity, particularly in patients with a DDD type pacemaker.
