The regulatory subunits of PI3k, p85α and p85β, differentially affect BRD7-mediated regulation of insulin signaling

Bromodomain-containing protein 7 (BRD7) has been shown to interact with the regulatory subunit of phosphatidylinositol 3-kinase(PI3k), p85, in the insulin signaling pathway. Here, we show that upregulation of hepatic BRD7 improves glucose homeostasiseven in the absence of either p85 isoform, p85a or p85b. However, BRD7 leads to differential activation of downstream effectorproteins in the insulin signaling pathway depending on which isoform of p85 is present. In the presence of only p85a, BRD7 overexpression increases phosphorylation of insulin receptor (IR) upon insulin stimulation, without increasing the recruitment of p85to IR substrate. Overexpression of BRD7 also increases activation of Akt in response to insulin, but does not affect basal phosphorylation levels of Akt. meanwhile, the phosphorylation of glycogen synthase kinase 3b (Gsk3b) is increased by overexpression ofBRD7. On the other hand, in the presence of only p85b, BRD7 overexpression does not affect phosphorylation levels of IR, and Aktphosphorylation is not affected by insulin stimulation following BRD7 upregulation. However, BRD7 overexpression leads to increased basal phosphorylation levels of Akt and Gsk3b. These data demonstrate that BRD7’s action on glucose homeostasis doesnot require the presence of both p85 isoforms, and p85a and p85b have unique roles in insulin signaling in the liver.

While the primary role of window attachments is often to moderate glare and solar heat gains,they are also able to provide additional daylight to interior *** this purpose,a variety of daylight-redirecting window systems have been developed over the past 150 *** reflective systems(slats/light shelves)or prismatic systems that rely on total internal reflection work well under specific solar conditions,but generally sacrifice performance over a much wider range of incident solar angles and sky *** systems-typically reflective slats-are more responsive to sun angles but have not been able to achieve optimal performance for glare and daylight redirection efficiency.A previous investigation into an adjustable,reflective blind concept first conceived of in the late 1970s showed promise but was not reduced to practice due to lack of adequate simulation and analysis *** this paper,this concept is further developed and its energy and visual comfort performance evaluated for four mid-latitude,temperate climates using ray-tracing simulation *** indicate significant potential lighting energy savings when compared with conventional automated reflective blinds(2.1-4.9 kWh/(m^(2)·a),or 14%-42%,depending on climate and orientation)or,especially,manually-operated matte white Venetian blinds(1.4-7.9 kWh/(m^(2)·a),or 9%-54%,depending on climate and orientation),while maintaining acceptable or better visual comfort conditions throughout the interior space.

Background:To evaluate the association between corneal central endothelial cell count(CECC)with reactivity for hepatitis B virus(HBV),hepatitis C virus(HCV),human immunodeficiency virus(HIV),human T-lymphotropic virus-1(HTLV1),and syphilis from an eye bank ***:Eye bank data included 19,159 donors and 38,318 corneas screened for HBV,HCV,HIV,HTLV1,and syphilis from July 2007-may *** and binary mixed effects models were used to determine the adjusted marginal effect a positive viral screening test had on CECC and morphology,*** models were adjusted for age,race,gender,lens status,and death to *** with missing data were excluded from the *** significance was defined as P valuess and 35,136 corneas were included in the final *** CECC for eyes with negative viral screening was 2,597±436 while the average CECC for eyes screening positive for syphilis,HBV,HCV,HIV,and HTLV1 were 2,638±392(P=0.073),2,569±419(P=0.815),2,603±363(P=0.207),2,615±360(P=0.733),and 2,625±436(P=0.362)***:The presence of HBV,HCV,HIV,HTLV1,and syphilis display no association with a statistically significant difference in CECC when compared to normal non-diseased donors.

The mixed spinel ferrite system mg0.95mn0.05Fe2-2xTizxO4 （0 ≤x ≤ 0.7） was synthesized by the conventional solid-state reaction technique. The effect of Ti^4＋ doping was studied by using the moessbauer spectroscopy measurements at room temperature. From the analysis of the moessbauer spectra, it is observed that s-electron density, electric field gradient （EFG）, quadrupole coupling constant （QCC） and the net hyperfine magnetic fields acting on the moessbauer nuclei-FeA^3＋ and FeB^3＋ change with the increase of Ti^4＋ doping in mg0.95mn0.05Fe2O4. The hyperfine magnetic field decreases with the increase of Ti^4＋ doping.

We report on the first observation of the neutron-rich nucleus^(131)*** isotope was produced via fragmentation reactions of intense secondary radioactive ion beams,including 134,*** secondary beams were produced from induced fission reactions from a stable 238U beam at 345 meV/*** reaction residues were selected by the ZeroDegree spectrometer and identified by measuring their magnetic rigidity,time of flight,energy loss,and total kinetic energy.

The superplastic behavior and associated deformation mechanisms of a fine-grained mg-10.1 Li-0.8Al-0.6Zn alloy(LAZ1011)with a grain size of 3.2μm,primarily composed of the BCCβphase and a small amount of the HCPαphase,were examined in a temperature range of 473 k to 623 *** microstructural refinement of this alloy was achieved by employing high-ratio differential speed *** best superplasticity was achieved at 523 k and at strain rates of 10^(-4)-5×10^(-4)s^(-1),where tensile elongations of 550±600%were *** the heating and holding stage of the tensile samples prior to tensile loading,a significant increase in grain size was observed at temperatures above 573 ***,it was important to consider this effect when analyzing and understanding the superplastic deformation behavior and *** the investigated strain rate range,the superplastic flow at low strain rates was governed by lattice diffusion-controlled grain boundary sliding,while at high strain rates,lattice diffusion-controlled dislocation climb creep was the rate-controlling deformation *** was concluded that solute drag creep is.un.ikely to *** the late stages of deformation at 523 k,it was observed that grain boundary sliding led to the agglomeration of theαphase,resulting in significant strain *** mechanism maps were constructed forβ-mg-Li alloys in the form of 2D and 3D formats as a function of strain rate,stress,temperature,and grain size,using the constitutive equations for various deformation mechanisms derived based on the data of the current tests.

This paper examines the quasi-static cyclic behavior, lateral strength and equivalent damping capacities of a system of post-tensioned segmental bridge columns tied with large diameter martensitic shape memory Alloy （smA） link-bars. moment-curvature constitutive relationships are formulated and analysis tools are developed for the PT column, including a modified four-spring model prepared for the smA bars. The suggested system is exemplified using a column with an aspect ratio of 7.5 and twelve 36.5 mm diameter NiTi martensitic smA bars. A post-tensioning force of 40% to 60% of the tendon yield strength is applied in order to obtain a self re-centering system, considering the residual stress of the martensitie smA bars. The cyclic response results show that the lateral strength remains consistently around 10% of the total vertical load and the equivalent viscous damping ratios reach 10%-12% of critical. When large diameter NiTi superelastic smA bars are incorporated into the column system, the cyclic response varies substantially. The creep behavior of the superelastic smA bar is accounted for since it affects the re-centering capability of the column. Two examples are presented to emphasize the modeling sensitivities for these special bars and quantify their cyclic behavior effects within the column assembly.
