Chitosan tubes enriched with fresh skeletal muscle fibers for delayed repair of peripheral nerve defects

Nerve regeneration after delayed nerve repair is often unsuccessful. indeed, the expression of genes associated with regeneration, including neurotrophic and gliotrophic factors, is drastically reduced in the distal stump of chronically transected nerves; moreover, schwann cells undergo atrophy, losing their ability to sustain regeneration. in the present study, to provide a three-dimensional environment and trophic factors supporting schwann cell activity and axon re-growth, we combined the use of an effective conduit(a chitosan tube) with a promising intraluminal structure(fresh longitudinal skeletal muscle fibers). This enriched conduit was used to repair a 10-mm rat median nerve gap after 3-month delay and functional and morphometrical analyses were performed 4 months after nerve reconstruction. Our data show that the enriched chitosan conduit is as effective as the hollow chitosan conduit in promoting nerve regeneration,and its efficacy is not statistically different from the autograft, considered the "gold standard" technique for nerve reconstruction. since hollow tubes not always lead to good results after long defects(> 20 mm), we believe that the conduit enriched with fresh muscle fibers could be a promising strategy to repair longer gaps, as muscle fibers create a favorable three-dimensional environment and release trophic factors. All procedures were approved by the Bioethical Committee of the University of Torino and by the italian Ministry of Health(approval number: 864/2016/PR) on september 14, 2016.

Lithium(Li)metal with high theoretical capacity and low electrochemical potential is the most ideal anode for next-generation high-energy ***,the practical implementation of Li anode has been hindered by dendritic growth and volume expansion during cycling,which results in low Coulombic efficiency(CE),short lifespan,and safety ***,we report a highly stable and dendrite-free Li metal anode by utilizing N-doped hollow porous bowl-like hard carbon/reduced graphene nanosheets(CB@rGO)hybrids as three-dimensional(3D)conductive and lithiophilic scaffold *** lithiophilic carbon bowl(CB)mainly works as excellent guides during the Li plating process,whereas the rGO layer with high conductivity and mechanical stability maintains the integrity of the composite by confining the volume change in long-range order during ***,the local current density can be reduced due to the 3D conductive ***,CB@rGO presents a low lithium metal nucleation overpotential of 18 mV,high CE of 98%,and stable cycling without obvious voltage fluctuation for over 600 cycles at a current density of 1 mA cm^(-2).Our study not only provides a good CB@rGO host and pre-Lithiated CB@rGO composite anode electrode,but also brings a new strategy of designing 3D electrodes for those active materials suffering from severe volume expansion.

Dear Editor,Anaplastic lymphoma kinase-positive(ALK+)anaplastic large cell lymphoma(ALCL)is an aggressive peripheral Tcell lymphoma representing 10%-15%of pediatric lymphoid *** constitutively activated nucleophosmin-ALK(NPM-ALK)fusion protein is the main driver of ALK+ALCL oncogenesis[1].Current clinical protocols can achieve high cure rates,with 75%event-free survival(EFs)at 5 years[2],but the patients’outcome with relapsed/refractory disease still remains poor[3].Liquid biopsy is widely applied for the prognostic and therapeutic stratification of onco-hematological *** a non-invasive procedure,it can be used to identify several biomarkers,such as small extracellular vesicles(s-EVs)[4].s-EVs are stable carriers of molecules that mediate genetic exchange between distant cells,alter the microenvironment,facilitate cancer cell progression and promote drug resistance[5].

Effect of intravenous ibuprofen on ischemia-Reperfusion injury Following Perioperative Tourniquet in Patients Undergoing Total Knee Arthroplasty

Oxidative stress occurs in the organism with ischemia due to tourniquet use and subsequent reperfusion. Oxidative stress increases postoperative morbidity. some Non-steroidal Anti-inflammatory Drugs (NsAiDs) perform their anti-inflammatory effects in part by binding or inhibiting their formation of active oxygen radicals at the site of inflammation. in this study, we aimed to evaluate the effect of iV ibuprofen on ischemia-reperfusion injury (iRi) in patients undergoing total knee arthroplasty over oxidative stress parameters. The patients were randomly divided into two groups. Each patient’s protocol number, age, sex, body mass index (BMI), additional disease, drug use, tourniquet time, hemoglobin value, additional analgesic requirement and application, adverse reaction development on the first postoperative day were recorded in the research follow-up form. Both groups of patients;before anesthesia, 45 minutes after tourniquet application, 5 minutes after tourniquet lowering, 20 minutes after tourniquet lowering and at 24th-hour post-op;TOS, TAS, paraoxonase, arylesterase, myeloperoxidase, catalase, ceruloplasmin, albumin, IMA, thiol-disulfide balance tests were studied. Statistical analysis of test results was performed. We observed that antioxidants decreased and oxidants increased on the first postoperative day in both groups in patients who underwent total knee arthroplasty. The decrease in antioxidant parameters was higher in IV ibuprofen doses compared to the control group in the case group;these doses indicate that the drug adversely affects the organism in the fight against oxidative stress, which is an undesirable effect. To evaluate this negative effect of IV ibuprofen which is increasingly used in postoperative analgesia, studies with different doses of drugs and different surgeries may be needed.

胰岛素B链中C-端十四肽的衍生物,即Cbz·Leu·Val·（s-Bz）Cys·Gly·（γ-OCH3）·Glu·（NG-NO2）Arg·Gly·Phe·Phe·Tyr·Thr·Pro·（ε-tos）Lys·Ala·OCH3(B17-30)分别从Cbz·Leu·Val·（s-Bz）Cys·Gly·ONP(B17-20ONP)和H·（γ-OCH3）Glu·（NG-NO2）Arg·Gly·Phe·Phe·Tyr·Thr·Pro·（ε-tos）Lys·Ala·OCH3(B21-30b)直接缩合或Cbz·Leu·Val·（s-Bz）Cys·Gly·（γ-OCH3）Glu·（NG-NO2）Arg·Gly·OH(B17-23a)和H·Phe·Phe·Tyr·Thr·Pro·（ε-tos）Lyg·Ala·OCH3(B24-30b)借DCCi法缩合而得。B21-30b由Cbz·（γ-OCH3）Glu·（NG-NO2）Arg·Gly·Phe·Phe·Tyr·Thr·Pro·（ε-tos）Lys·Ala·OCH3(B21-30)经溴化氢的三氟醋酸溶液处理后获得;B17-20ONP系由Cbz·Leu·Val·（s-Bz）Cys·Gly·OH和对硝基苯酚用碳二亚胺法缩合而成。按新的方案（3+7）将Cbz·（γ-OCH3）Glu·（NG-NO2）Arg·Gly·OH(B21-23a)和B24-30b用DCCi法缩合生成B21-30,其物理常数与以前用（2+8）方案合成的十肽衍生物的数据相同。B21-23a由Cbz·（γ-OCH3）·Glu·ONP和（NG-NO2）Arg·Gly·OH缩合生成;B24-30b系由Cbz·Phe·Phe·Tyr·Thr·Pro·（ε-tos）Lys·Ala·OCH3经催化氢化而得。该七肽衍生物又由Cbz·Phe·Phe·Tyr·Thr·OH(B24-27a)和H·Pro·（ε-tos）Lys·Ala·OCH3以DCCi法缩合而得。B17-23a则由B17-20ONP和H·（γ-OCH3）Glu·（NG-NO2）Arg·Gly·OH 缩合而成。

Ⅲ型结核病结核菌株原发耐药性测定

目的为初治肺结核选用合理化疗药物提供科学依据．方法采用改良罗氏培养基对Ⅲ型初治肺结核分离出的９２株结核菌做百种常用抗结核药物的药敏试验．结果９２株结核菌耐药发生率为７２．８％，耐药率依次是ＳＭ５０．０％、ＩＮＨ４４．６％、ＲＦＰ２５．０％、ＥＭＢ２１．７％、ＫＭ９．８％、ＰＡＳＳ．７％、ＣＰＭ４．３％和１３２ｌｔｈ２．２％．单耐１药和同时耐２药之和占９２株的５２．２％，耐ＨＲ２０株占２１．７％。结论Ｓ、Ｈ、Ｒ、Ｅ耐药发生率较高，初治结核病做耐药规定是必要的；耐１、２种药物的菌株比例较大．故应联合用药。
