Unusual switchable peroxidase-mimicking nanozyme for the determination of proteolytic biomarker

Detection of enzyme biomarkers originating from either bio-fluids or contaminating microorganisms is of utmost importance in clinical diagnostics and food safety. Herein, we pres.nt a simple, low-cost and easy-to-use s.ns.ng approach based on the switchable peroxidase-mimicking activity of plasmonic gold nanoparticles (AunPs) that can catalyse for the oxidation of 3,3’,5’5-tetramethylbenzidine (TMB) for the determination of protease enzyme. The AunP surface is modified with casein, showing dual functionalities. The first function of the coating molecule is to suppress the intrinsic peroxidase-mimicking activity of AunPs by up to 77.1%, due to surface shielding effects. secondly, casein also functions as recognition sites for the enzyme biomarker. In the pres.nce of protease, the enzyme binds to and catalyses the degradation of the coating layer on the AunP surface, resulting in the recovery of peroxidase-mimicking activity. This is shown visually in the development of a blue colored product (oxidised TMB) or spectroscopically as an increase in absorbance at 370 and 650 nm. This mechanism allows for the detection of protease at 44 ng·mL^-1 in 90 min. The nanos.nsor circumvents issues associated with current methods of detection in terms of ease of use, compatibility with point-of-care testing, low-cost production and short analysis time. The s.ns.ng approach has also been applied for the detection of protease spiked in ultra-heat treated (UHT) milk and s.nthetic human urine samples at a limit of detection of 490 and 176 ng·mL^-1, respectively, showing great potential in clinical diagnostics, food safety and quality control.

In this paper,we establish a new algorithm to the non-overlapping schwarz domain decomposition methods with changing transmission conditions for solving one dimensional advection reaction diffusion *** precisely,we first describe the new algorithm and prove the convergence results under several natural assumptions on the sequences of parameters which determine the transmission *** we give a simple method to estimate the new value of parameters in each *** interesting advantage of our method is that one may update the better parameters in each iteration to save the computational cost for optimizing the parameters after many *** some numerical experiments are performed to show the behavior of the convergence rate for the new method.

Defect engineering of water-dispersible g-C_(3)n_(4) photocatalysts by chemical oxidative etching of bulk g-C_(3)n_(4) prepared in different calcination atmospheres

In this study,water-dispersible graphitic carbon nitride(g-C_(3)n_(4))photocatalysts were successively prepared through the chemically oxidative etching of bulk g-C_(3)n_(4) that was polymerized thermally in different calcination atmospheres such as air,CO_(2),and n_(2).The different calcination atmospheres directly influenced the physicochemical and optical properties of both bulk and water-dispersible g-C_(3)n_(4),changing the photocatalytic degradation behavior of methylene blue(MB)and tetracycline hydrochloride(TCHCl)for water-dispersible g-C_(3)n_(4).The bubble-burst process in the thermal polymerization of thiourea produced defective edges containing C=O groups that preferred substituting the C-nHx groups over bulk g-C_(3)n_(4).In the oxygen-free n_(2) atmosphere among the different calcination atmospheres,more C=O functional groups were generated on the defective edges of bulk g-C_(3)n_(4),resulting in the highest n vacancy of the tri-s-triazine *** the successive chemical oxidation,s-or O-containing functional groups were introduced onto water-dispersible g-C_(3)n_(4).The water-dispersible g-C_(3)n_(4) photocatalyst from the oxygen-free n_(2) atmosphere(nTw)contained the most O-and s-functional groups on the g-C_(3)n_(4) ***,nTw exhibited the highest photocatalytic activity in the MB and TC-HCl photodegradation because of its slowest recombination process,which was ascribed to the unique surface properties of nTw such as abundant functional groups on the defective edges and n-deficient property.

Objective:To evaluate the antitumor activity both in vitro and in vivo of saponin–phospholipid complex of Panax ***:The in vitro cytotoxic effect of saponins extract and saponin–phospholipid complex against human lung cancer nCI-H460 and breast cancer cell lines BT474 was examined us.ng MTs *** in vivo evaluation of antitumor potential,saponin and saponin–phospholipid complex were administered orally in rats induced mammary carcinogenesis by 7,12-dimethylbenz(a)anthracene,for 30 ***:Our data showed that saponin–phospholipid complex had stronger anticancer effect compared to saponin *** IC50 values of saponin–phospholipid complex and saponin extract for nCI-H460 cell lines were 28.47 mg/m L and 47.97 mg/mL,respectively and these values for BT474 cells were 53.18 mg/mL and 86.24 mg/mL,*** vivo experiments,administration of saponin,saponin–phospholipid complex and paclitaxel(positive control) effectively suppressed 7,12-dimethylbenz(a) anthracene-induced breast cancer evidenced by a decrease in tumor volume,the reduction of lipid peroxidation level and increase in the body weight,and elevated the enzymatic antioxidant activities of superoxide dismutase,catalase,glutathione peroxidase in rat breast ***:Our study suggests that saponin extract from Panax notogins.ng and saponin–phospholipid complex have potential to prevent cancer,especially breast cancer.

Xanthine oxidase inhibitors from Archidendron clypearia(Jack.) I.C. niels.n: Results from systematic screening of Vietnamese medicinal plants

Objective: To screen Vietnamese medicinal plants for xanthine oxidase(XO) inhibitory activity and to isolate XO inhibitor(s) from the most active plant. Methods: The plants materials were extracted by methanol. The active plant materials were fractionated us.ng different organic solvents, including n-hexane, ethyl acetate, and n-butanol. Bioassay-guided fractionation and column chromatography were used to isolate compounds. The compounds structures were elucidated by analysis of spectroscopic data, including IR, Ms, and nMR. Results: Three hundreds and eleven methanol extracts(CME) belonging to 301 Vietnamese herbs were screened for XO inhibitory activity. Among these plants, 57 extracts displayed XO inhibitory activity at 100 μg/m L with inhibition rates of over 50%. The extracts of Archidendron clypearia, smilax poilanei, Linociera ramiflora and Passiflora foetida exhibited the greatest potency with IC_(50) values below 30 μg/m L. Chemical study performed on the extract of Archidendron clypearia resulted in the isolation of six compounds, including 1-octacos.nol, docos.noic acid, daucosterol, methyl gallate, quercitrin and(-)-7-O-galloyltricetiflavan. The compound(-)-7-O-galloyltricetiflavan showed the most potent XO inhibitory activity with an IC_(50) value of 25.5 μmol/L. Conclusions: From this investigation, four Vietnamese medicinal plants were identified to have XO inhibitory effects with IC_(50) values of the methanol extracts below 30 μg/m L. Compound(-)-7-O-galloyltricetiflavan was identified as an XO inhibitor from Archidendron clypearia with IC_(50) value of 25.5 μmol/L.

In this study,we reported on the concept and practical use of cation exchange res.n(CER)for removing anions in water via pretreating the CER with metal *** cation exchange res.nsupported iron and magnesium oxides/hydroxides composite(FeMg/CER)was s.nthesized and introduced as a new and potential adsorbent for selective removal of nitrate ion in the water *** of FeMg/CER were determined by techniques such as Fouriertransform infrared spectroscopy,scanning electron microscopy,and Xray *** results showed that FeMg/CER material had a high nitrate adsorption capacity of 200 mg nO_(3)^()·g^(1)with a fast equilibrium adsorption time of 30 min at pH *** addition,it had good durability of at least 10 times of regeneration,which could be applied to practical water and wastewater treatment.
