The ARP2/3 complex, a highly conserved nucleator of F-actin polymerization, and its activator, the sCAR complex, have been shown to play important roles in leaf epidermal cell morphogenesis in Arabidopsis. However, the intracellular site（s） and function（s） of sCAR and ARP2/3 complex-dependent actin polymerization in plant cells remain unclear. We demonstrate that putative sCAR complex subunits BRK1 and sCAR1 are localized to the plasma membrane at sites of cell growth and wall deposition in expanding cells of leaves and roots. BRK1 localization is sCAR-dependent, providing further evidence of an association between these proteins in vivo. Consistent with plasma membrane localization of sCAR complex subunits, cortical F-actin accumulation in root tip cells is reduced in brkl mutants. Moreover, mutations disrupting the sCAR or ARP2/3 complex reduce the growth rate of roots and their ability to penetrate semi-solid medium, suggesting reduced rigidity. Cell walls of mutant roots exhibit abnormal structure and composition at intercellular junctions where BRK1 and sCAR1 are enriched in the adjacent plasma membrane. Taken together, our results suggest that sCAR and ARP2/3 complex-dependent actin polymerization promotes processes at the plasma membrane that are important for normal growth and wall assembly.

BACKGROUND The regulatory factor X6 (RFX6), a member of regulatory factor X family, is known to play a key role in the development and differentiation of pancreatic beta cells as well as insulin production and secretion. However, the potential role of RFX6 in type 2 diabetes (T2D) is still unclear. AIM Recent studies have indicated that RFX6 binding to DNA could be disrupted in diabetes. Therefore, in this study we investigated whether genetic mutations are present in the DNA binding domain of RFX6 gene that could abrogate its function in T2D. METHODs A cohort of T2D patients was enrolled in this study, and the gene encoding the DNA binding domain of RFX6 was amplified by polymerase chain reaction and then analysed by direct DNA sequencing. REsULTs The DNA sequence analysis revealed the absence of any exonic mutation. However, we have identified a new heterozygous single nucleotide polymorphism (IVs6+31 C>T) in the intronic region of DNA binding domain gene that is present in 9.2% and 8.5% of diabetic and control people, respectively (P = 0.97).CONCLUsION We report the absence of any significant genetic variant that could affect the function of RFX6-DNA binding domain in T2D.

This paper presents model problem studies for micropolar thermoviscoelastic solids without memory and micropolar thermoviscous fluid using micropolar non-classical continuum theories (NCCT) based on internal rotations and rotation rates in which rotational inertial physics is considered in the derivation of the conservation and balance laws (CBL). The dissipation mechanism is due to strain rates as well as rotation rates. Model problems are designed to demonstrate and illustrate various significant aspects of the micropolar NCCT with rotational inertial physics considered in this paper. In case of micropolar solids, the translational and rotational waves are shown to coexist. In the absence of microconstituents (classical continuum theory, CCT) the internal rotations are a free field, hence have no influence on CCT. Absence of gradients of displacements and strains in micropolar thermoviscous fluid medium prohibits existence of translational waves as well as rotational waves even though the appearance of the mathematical model is analogous to the solids, but in terms of strain rates. It is shown that in case of micropolar thermoviscous fluids the BAM behaves more like time dependent diffusion equation i.e., like heat conduction equation in Lagrangian description. The influence of rotational inertial physics is demonstrated using BLM as well as BAM in the model problem studies.

Rotational Inertial Physics in Non-Classical Thermoviscous Fluent Continua Incorporating Internal Rotation Rates

In this paper, we derive non-classical continuum theory for physics of compressible and incompressible thermoviscous non-classical fluent continua using the conservation and balance laws (CBL) by incorporating additional physics of internal rotation rates arising from the velocity gradient tensor as well as their time varying rates and the rotational inertial effects. In this non-classical continuum theory time dependent deformation of fluent continua results in time varying rotation rates i.e., angular velocities and angular accelerations at material points. Resistance offered to these by deforming fluent continua results in additional moments, angular momenta and inertial effects due to rotation rates i.e., angular velocities and angular accelerations at the material points. Currently, this physics due to internal rotation rates and inertial effects is neither considered in classical continuum mechanics (CCM) nor in non-classical continuum mechanics (NCCM). In this paper, we present a derivation of conservation and balance laws in Eulerian description: conservation of mass (CM), balance of linear momenta (BLM), balance of angular momenta (BAM), balance of moment of moments (BMM), first and second laws of thermodynamics (FLT, sLT) that include: (i) Physics of internal rotation rates resulting from the velocity gradient tensor;(ii) New physics resulting due to angular velocities and angular accelerations due to spatially varying and time dependent rotation rates. The balance laws derived here are compared with those that only consider the rotational rates but neglect rotational inertial effects and angular accelerations to demonstrate the influence of the new physics. Constitutive variables and their argument tensors are established using conjugate pairs in the entropy inequality, additional desired physics and principle of equipresence when appropriate. Background:National tuberculosis(TB)programs increasingly engage with international non-governmental organizations(INGOs),especially to provide TB care in complex settings where community involvement might be *** Myanmar,however,there is limited data on how such INGO community-based programs are organized and how effective they *** this study,we describe four INGO strategies for providing community-based TB care to hard-to-reach populations in Myanmar,and assess their contribution to TB case ***:We conducted a descriptive study using program data from four INGOs and the National TB Program(NTP)in *** each INGO,we extracted information on its approach and key activities,the number of presumptive TB cases referred and undergoing TB testing,and the number of patients diagnosed with TB and their treatment *** contribution of INGOs to TB diagnosis in their selected townships was calculated as the proportion of INGO-diagnosed new TB cases out of the total NTP-diagnosed new TB cases in the same ***:All four INGOs implemented community-based TB care in challenging contexts,targeting migrants,post-conflict areas,the urban poor,and other vulnerable *** recruited community volunteers via existing community health volunteers or health structures,one via existing community leaderships,and one directly involved TB infected/affected *** INGOs compensated volunteers via performance-based financing,and two provided financial and in-kind *** relied on NTP laboratories for diagnosis and TB drugs,but provided direct observation treatment support and treatment follow-up.A total of 21995 presumptive TB cases were referred for TB diagnosis,with 7383(34%)new TB cases diagnosed and almost all(98%)successfully *** four INGOs contributed to the detection of,on average,36%(7383/20663)of the total new TB cases in their respective townships(range:15-52%).Conclusion:Community-based TB care suppor
