Two major bacterial blight （BB） resistance genes （Xa21 and xa13） and a major gene for blastresistance （Pi54） were introgressed into an Indian rice variety MTU1010 through marker-assistedbackcross breeding. Improved samba Mahsuri （possessing Xa21 and xa13） and NLR145 （possessingPi54） were used as donor parents. Marker-assisted backcrossing was continued till BC2 generationwherein PCR based functional markers specific for the resistance genes were used for foregroundselection and a set of parental polymorphic microsatellite markers were used for background selectionat each stage of backcrossing. selected BC2F1 plants from both crosses, having the highest recoveriesof MTU1010 genome （90% and 92%, respectively）, were intercrossed to obtain intercross F1 （ICF1） plants,which were then selfed to generate 880 ICF2 plants possessing different combinations of the BB andblast resistance genes. Among the ICF2 plants, seven triple homozygous plants （xa13xa13Xa21Xa21Pi54Pi54）with recurrent parent genome recovery ranging from 82% to 92% were identified. All the seven ICF2plants showed high resistance against the bacterial blight disease with a lesion lengths of only 0.53–2.28 cm, 1%–5% disease leaf areas and disease scoring values of ‘1’ or ‘3’. The seven ICF2 plants wereselfed to generate ICF3, which were then screened for blast resistance, and all were observed to behighly resistant to the diseases. several ICF3 lines possessing high level of resistance against BB andblast, coupled with yield, grain quality and plant type on par with MTU1010 were identified and advanced forfurther selection and evaluation.

The kinetics study of the reduction of Fast Green dye with cetylpyridinum chloride as cationic surfactant

The kinetics of the reaction of Fast Green dye(FG)with cetylpyridinum chloride was studied in alkaline medium by UV-Visible *** of Fast Green dye was carried out by varying the fast green dye concentration,cetylpyridinum chloride concentration and concentration of sodium *** the present study the reduction of dye was carried out in order to reduce the color *** interaction of dye was carried out with reducing analyte(cetylpyridinum chloride).The rate of the reaction was determined by varying the above parameters at different *** was observed that the reduction followed pseudo first-order kinetics with respect to dye,surfactant,OH–ion concentration according to the following reaction *** like entropy of activation(Δs)and free energy of activation(ΔG)showed the extremely solvated states of transient complex which was less disorderly arranged than the oxidized form of dye,whereas Ea values reflects a high amount of energy required for the reduction of dye with cetylpyridinum chloride.

Effect of foreign ions for the determination of metal ion in the presence of surfactants

Aluminum is the third most abundant metal in the Earth’s *** its ubiquitous nature it is present in small amount in living *** toxicity has been implicated in the pathogenesis of renal distinct clinical syndromes,including progressive and fatal encephalopathy and bone *** the present study,Al was selected for the analysis by complexometric *** method was based on the formation of a red colored ternary complex by the reaction of Aluminum with Aluminon(Aurin tricarboxylic acid triammonium salt)in the presence of micellar *** ternary complex of Aluminum with the surfactant Triton X-100 shows a maximum absorbance at 530 nm wavelength at pH 4.0 while with the sodium dodecyl sulfate it shows a maximum absorbance at 525 nm and at pH *** reaction was proceeded by the variation in pH and concentrations of surfactants,aluminon,*** effects on the reaction of aluminum with aluminon complex in micellar media were recorded by UV-visible *** reaction was found to be extremely rapid at room *** system obeys Lambert Beer’s law between 0.24 and 21.74μg/mL concentrations with Triton *** values of slope,intercept and correlation coefficients were 0.07,0.348 and 0.989,*** concentration varied between 0.24 and 24.14μg/mL with sodium dodecyl sulfate and the values of slope,intercept and correlation coefficients were 0.029,0.148 and 0.962,*** foreign ion effect was also tested by keeping the constant concentration of metal ion and determining its concentration in the presence of different foreign *** method was also applied for the determination of Al(III)in pharmaceutical formulations and water samples,which showed an excellent resemblance between reported and obtained results.

Dragon’s blood(DB)has shown a protective effect on neurological ***(MG)or simulated MG(sMG)can induce blood-brain barrier(BBB)dysfunction,which is a characteristic feature of neurological *** study’s purpose was to evaluate the effect of DB on sMG-induced BBB dysfunction and explore its signaling *** DB and vitamin C(Vc)were administered orally for tail-suspended rats within 3 *** and Vc solutions were added to human brain microvascular endothelial cells(HCMEC/D3)cells,which were then exposed to sMG for 24 *** protective effect of DB was assessed by hematoxylin and eosin and Nissl staining,ultrastructure observation,and permeability in *** apoptosis and the distribution of tight junction(TJ)and adherens junction(AJ)proteins and filamentous actin(F-actin)were examined in HCMEC/*** oxidative stress and inflammation,and TJ and AJ protein expressions were determined in rat brain and HCMEC/*** focal adhesion kinase(FAK)signaling pathway proteins were *** protected sMG-induced rat BBB disruption by improving neuronal apoptosis,repairing widened intercellular space,and decreasing BBB *** effectively relieved sMG-induced HCMEC/D3 damage by inhibiting cell apoptosis and restoring F-actin spindle *** doses of DB upregulated TJ and AJ protein expressions and decreased oxidative stress and proinflammatory cytokine levels in rat brain and HCMEC/*** enhanced the expressions of FAK signal transduction proteins and F-actin/globular actin(G-actin)ratio in rat brain and HCMEC/D3,suggesting that DB promotes actin cytoskeleton polymerization,benefits the endothelial cell-cell and cell-extracellular matrix adhesion,and,in consequence,contributes to BBB integrity.

Development of diabetic cataract is mainly associated with the accumulation of sorbitol via the polyol pathway through the action of Aldose reductase(AR).Hence,AR inhibitors are considered as potential agents in the management of diabetic *** study explored the AR inhibition potential of Hemidesmus indicus *** root extract by in silico and ex vivo *** docking studies(Auto Dock tool)betweenβ-sitosterol,hemidesminine,hemidesmin-1,hemidesmin-2,and AR showed thatβ-sitosterol(−10.2 kcal/mol)and hemidesmin-2(−8.07 kcal/mol)had the strongest affinity to AR *** vivo studies were performed by incubating isolated goat lenses in artificial aqueous humor using galactose(55 mM)as cataract inducing agent at room temperature(pH 7.8)for 72 *** treatment with Vitamin E acetate−100μg/mL(standard)and test extract(500 and 1000μg/mL)separately,the estimation of biochemical markers showed inhibition of lens AR activity and decreased sorbitol ***,extract also normalized the levels of antioxidant markers like sOD,CAT,*** results showed evidence that *** *** root was able to prevent cataract by prevention of opacification and formation of polyols that underlines its potential as a possible therapeutic agent against diabetic complications.

A single-Tube, Functional Marker-Based Multiplex PCR Assay for simultaneous Detection of Major Bacterial Blight Resistance Genes Xa21, xa13 and xa5 in Rice

In marker-assisted breeding for bacterial blight(BB) resistance in rice, three major resistance genes, viz., Xa21, xa13 and xa5, are routinely deployed either singly or in combinations. As efficient and functional markers are yet to be developed for xa13 and xa5, we have developed simple PCR-based functional markers for both the genes. For xa13, we designed a functional PCR-based marker, xa13-prom targeting the In Del polymorphism in the promoter of candidate gene Os8N3 located on chromosome 8 of rice. With respect to xa5, a multiplex-PCR based functional marker system, named xa5 FM, consisting of two sets of primer pairs targeting the 2-bp functional nucleotide polymorphism in the exon II of the gene TFIIA5(candidate for xa5), has been developed. Both xa13-prom and xa5 FM can differentiate the resistant and susceptible alleles for xa13 and xa5, respectively, in a co-dominant fashion. Using these two functional markers along with the already reported functional PCR-based marker for Xa21(p TA248), we designed a single-tube multiplex PCR based assay for simultaneous detection of all the three major resistance genes and demonstrated the utility of the multiplex marker system in a segregating population.
