Determination of asenapine in presence of its inactive metabolites in human plasma by LC-Ms/Ms

A highly selective and sensitive liquid chromatography-tandem mass spectrometry （LC-Ms/Ms） assay has been described for the determination of asenapine （AsE） in presence of its inactive metabolites N-desmethyl asenapine （DMA） and asenapine-N-glucuronide （AsG）. AsE, and AsE 13C-d3, used as internal standard （Is）, were extracted from 300 μL human plasma by a simple and precise liquid-liquid extraction procedure using methyl tert-butyl ether. Baseline separation of AsE from its inactive metabolites was achieved on Chromolith Performance RPse （100 mm ×4.6 mm） column using acetonitrile- 5.0 mM ammonium acetate-10% formic acid （90：10：0.1, v/v/v） within 4.5 min. Quantitation of AsE was done on a triple quadrupole mass spectrometer equipped with electrospray ionization in the positive mode. The protonated precursor to product ion transitions monitored for AsE and AsE 13C-d3 were m/z 286.1 → 166.0 and m/z 290.0 → 166.1, respectively. The limit of detection （LOD） and limit of quantitation （LOQ） of the method were 0.0025 ng/mL and 0.050 ng/mL respectively in a linear concentration range of 0.050-20.0 ng/mL for AsE. The intra-batch and inter-batch precision （% CV） and mean relative recovery across quality control levels were 〈 5.8% and 87.3%, respectively. Matrix effect, evaluated as Is-normalized matrix factor, ranged from 1.03 to 1.05. The stability of AsE under different storage conditions was ascertained in presence of the metabolites. The developed method is much simpler, matrix free, rapid and economical compared to the existing methods. The method was suc- cessfully used for a bioequivalence study of asenapine in healthy Indian subjects for the first time.

Probabilistic assessment of surface level seismic hazard in India using topographic gradient as a proxy for site condition

This paper presents spatial variation of seismic hazard at the surface level for India,covering 6-38°N and 68-98°*** most recent knowledge on seismic activity in the region has been used to evaluate the hazard incorporating uncertainties associated with the seismicity parameters using different modeling *** types of seismic source models,*** sources,gridded seismicity model and areal sources,were considered to model the seismic sources and different sets of ground motion prediction equations were used for different tectonic provinces to characterize the attenuation *** hazard estimation at bedrock level has been carried out using probabilistic approach and the results obtained from various methodologies were combined in a logic tree *** seismic site characterization of India was done using topographic slope map derived from Digital Elevation Model *** paper presents estimation of the hazard at surface level,using appropriate site amplification factors corresponding to various site classes based on V_（s30） values derived from the topographic *** variation of surface level peak horizontal acceleration（PHA） for return periods of 475 years and 2475 years are presented as contour maps.

Highly sensitive LC–Ms/Ms method to estimate doxepin and its metabolite nordoxepin in human plasma for a bioequivalence study

A selective, sensitive and rugged liquid chromatography–tandem mass spectrometry(LC–Ms/Ms) assay has been developed for the simultaneous determination of doxepin(Dox) and its pharmacologically active metabolite, nordoxepin(NDox) in human plasma. The analytes and their internal standards(Is)were extracted from 500 m L of human plasma by liquid-liquid extraction using methyl tert-butyl *** separation was achieved on Hypurity C8 column(100 mm ? 4.6 mm, 5 mm) using a mixture of acetonitrile-methanol(95:5, v/v) and 2.0 mM ammonium formate in 93:7(v/v) ratio. Detection was accomplished by tandem mass spectrometry in the positive ionization and multiple reaction monitoring acquisition mode. The protonated precursor to product ion transitions studied for Dox, NDox,and their corresponding Iss, propranolol and desipramine, were m/z 280.1-107.0, 266.0-107.0,260.1-116.1 and 267.1-72.1, respectively. A linear dynamic range of 15.0–3900 pg/mL for Dox and 5.00–1300 pg/mL for NDox was established with mean correlation coefficient(r2) of 0.9991 and 0.9993, respectively. The extraction recovery ranged from 86.6%–90.4% and 88.0%–99.1% for Dox and NDox, respectively. The intra-batch and inter-batch precision(% CV) across quality control levels was r 8.3% for both the analytes. stability evaluated under different storage conditions showed no evidence of degradation and the % change in stability samples compared to nominal concentration ranged from 4.7% to12.3%. The method was successfully applied to a bioequivalence study of 6 mg doxepin hydrochloride orally disintegrating tablet in 41 healthy Indian subjects under fasting and fed conditions.

Low-Frequency Dependence of Conductivity and Dielectric Properties of Polyaniline/ZnFe_(2)O_(4) Nanocomposites

Conducting polyaniline/ZnFe_(2)O_(4) nanocomposites are synthesized by using a simple and inexpensive one-step in-situ polymerization method in the presence of ZnFe2O4 *** structural,morphological and electrical properties of the samples are characterized by x-ray diffraction,Fourier transform infrared spectra and scanning electron *** results reveal the formation of polyaniline/ZnFe2O4 *** morphology of these samples is studied by scanning electron ***,the ac conductivity(σac)of these composites is investigated in the frequency range of 1 kHz–10 *** presence of polarons and bipolarons are responsible for the frequency dependence of ac conductivity in these *** ac conductivity is found to be constant up to 1MHz and thereafter it increases *** ac conductivity of 0.695 s・cm^(−1) at room temperature is observed as the maxima for the polyaniline with 40wt%of the ZnFe2O4 nanocomposite.

Design and optimization analysis of dual material gate on DG-IMOs

An impact ionization MOsFET （IMOs） is evolved for overcoming the constraint of less than 60 mV/decade sub-threshold slope （ss） of conventional MOsFET at room temperature. In this work, first, the device performance of the p-type double gate impact ionization MOsFET （DG-IMOs） is optimized by adjusting the device design parameters. The adjusted parameters are ratio of gate and intrinsic length, gate dielectric thickness and gate work function. secondly, the DMG （dual material gate） DG-IMOs is proposed and investigated. This DMG DG-IMOs is further optimized to obtain the best possible performance parameters. simulation results reveal that DMG DG-IMOs when compared to DG-IMOs, shows better IoN, ION/IoFF ratio, and RF parameters. Results show that by properly tuning the lengths of two materials at a ratio of 1.5 in DMG DG-IMOs, optimized perform- ance is achieved including ION/IoFF ratio of 2.87 × 10^9 A/μm with/ON as 11.87 × 10^-4 A/μm and transconductance of 1.06× 10^-3 s/μm. It is analyzed that length of drain side material should be greater than the length of source side material to attain the higher transconductance in DMG DG-IMOs.

Analysis of Genotoxicity Induced by Food Dyes in Root Meristem of salvia hispanica L.

Food dyes are chemicals either natural or synthetic that were used by humans to give attractive colors to numerous food products. Colorful food products may cause several health-related issues to us because they may be prepared through synthetic food dyes. The color which is intended to attract people to eat is given to them by the use of some artificial food dyes such as Erythrosine, Allura red, Indigostine, Tartrazine, sunset yellow, etc. These artificial food dyes are synthetic chemicals that can cause numerous health-related problems if they were taken in by individuals more than the limited amount. Artificial food dyes are becoming very common in use in different ways to provide an attractive and soothing color to food products such as sweets, drinks, candies, etc. The food products we consume with very good and attractive colors are not as good for our health as they appear. Here in this study, we are observing the cytological effect of two different synthetic food dyes (Brilliant blue and sunset yellow) in the root meristem of salvia hispanica L. (Chia plant) which is a major source of α-linolenic acid, mainly omega 3 fatty acids and dietary fibers found in the seed. For this purpose, germinated seeds of Chia are in the germinator. After the emergence of the young roots, we have given the treatment of the respective dyes of 0.5%, 1%, 1.5% & 2% respectively with a control set. After treatment, the roots were fixed in Carnoy’s fixative and preserved in alcohol for further analysis. When the slides of the treated roots were prepared and analyzed we observed that numerous cytological abnormalities such as (stickiness, fragmentation, bridge, laggards, disorientation, etc.) were there in the root cells and the level of abnormalities are increasing with increasing treatment period due to the exposure of them to respective dyes. These results are enough to conclude that the consumption of these dyes is harmful to the health of humans.

Improved simultaneous quantitation of candesartan and hydrochlorthiazide in human plasma by UPLC–Ms/Ms and its application in bioequivalence studies

A validated ultra-performance liquid chromatography mass spectrometric method （UPLC-Ms/Ms） was used for the simultaneous quantitation of candesartan （CN） and hydrochlorothiazide （HCT） in human plasma. The analysis was performed on UPLC-Ms/Ms system using turbo ion spray interface. Negative ions were measured in multiple reaction monitoring （MRM） mode. The analytes were extracted using a liquid-liquid extraction （LLE） method by using 0.1 mL of plasma volume. The lower limit of quantitation for CN and HCT was 1.00 ng/mL whereas the upper limit of quantitation was 499.15 ng/mL and 601.61 ng/mL for CN and HCT respectively. CN d4 and HCT-13Cd2 were used as the internal standards for CN and HCT respectively. The chromatography was achieved within 2.0 min run time using a C18 Pheno-menex, Gemini NX （100 mm ~ 4.6 mm, 5 mm） column with organic mixture：buffer solution （80：20, v/v） at a flow rate of 0.800 mL/min. The method has been successfully applied to establish the bioequivalence of candesartan cilexetil （CNC） and HCT immediate release tablets with reference product in human subjects.
