Boldness is defined as the tendency of an individual to take risks when exposed to novel objects or situations. The main aim of this work was to dilucidate if boldness was related to the development of the secondary sexual traits （ssTs） in the male of the fiver blenny salariafluviatilis, a freshwater fish which takes care of eggs. As a second objective ssTs effect on pa rental status adoption was also be explored. Wild young fish were caught in the segre River （Ebro basin） in November 2010. Fish were kept in five aquaria in groups of eight males and eight females with artificial nests. Boldness （boldness score and hesitancy） of males （initially with little ssTs development） was tested as the time required to exit a refuge when exposed to a new environ ment. Fish were mantained in the same aquaria for several months, and males＇ ssTs development and reproductive behaviour were monitored. Boldness score was negatively associated to cephalic crest development. Hesitancy was negatively related to male length and was also influenced by the interaction between male length and cephalic crest. Parental status acquisition was positively related to the male＇s length, marginally related to anal gland development, and nonrelated to cephalic crest. These re sults suggest an intrasexual role of the crest as an informative trait of boldness （honest signal） which might not necessarily be chosen by the females. It is also discussed that correlational selection could be responsible for linking boldness and growth rate in this species. sexual selection, in addition to having directed the evolution of secondary sexual traits, might have favored certain personality traits （e.g. boldness） associated with the achievement of a larger body length .

The recently developed RNA-guided clustered regularly interspaced short palindromic repeat （CRIsPR）/CRIsPR-associated 9 （Cas9） nuclease system has progressed to be an invaluable technology for genome manipulation in somatic cell types and germline model organisms. While the unprecedented advance in human embryo gene editing research has great potential in next-generation therapeutics, it raises various ethical concerns that need to be addressed before being translated for clinical use. Here, we discuss the current and potential applications of CRIsPR/Cas9 technology and its limitations in clinical applications, as well as ethical and legal considerations in the treatment, disease prevention or disability in somatic cells or human embryo via gene editing.

Quantifying post logging biomass loss using satellite images and ground measurements in southeast Cameroon

Forest logging in the Congo basin has led to forest fragmentation due to logging infrastructures and felling gaps. In the same vein, forest concessions in the Congo basin have increasing interest in the REDD＋ mechanism. However, there is little information or field data on carbon emissions from forest degradation caused by logging. To help fill this gap, Landsat 7 and 8 and sPOT 4 images of the East Region of Cameroon were processed and combined with field measurements （measurement of forest roads widths, felling gaps and log yards） to assess all disturbed areas. Also, measurements of different types of forest infrastructures helped to highlight emission factors. Forest contributes to 5.18 % of the degradation of the annual allowable cut （AAC）（84.53 ha） corresponding to 4.09 % of forest carbon stock （6.92 t ha^-1）. Felling gaps constitute the primary source of degradation, represented an estimated area of 32.41 ha （2 % of the cutting area） far ahead of primary roads （18.44ha） and skid trails （16.36 ha）. Assessment of the impact of degradation under the canopy requires the use of high resolution satellite images and field surveys.

Pancreatic cancer cells render tumor-associated macrophages metabolically reprogrammed by a GARP and DNA methylation-mediated mechanism

How tumor-associated macrophages transit from a predominant antitumor M1-like phenotype to a protumoral M2-like phenotype during the development of pancreatic ductal adenocarcinoma (PDA) remains to be elucidated. We thus conducted a study by employing a PDA-macrophage co-culture system, an “orthotopic” PDA syngeneic mouse model, and human PDA specimens, together with macrophages derived from GARP knockout mice and multiple analytic tools including whole-genome RNA sequencing, DNA methylation arrays, multiplex immunohistochemistry, metabolism measurement, and invasion/metastasis assessment. Our study showed that PDA tumor cells, through direct cell–cell contact, induce DNA methylation and downregulation of a panel of glucose metabolism and OXPHOs genes selectively in M1-like macrophages, leading to a suppressed glucose metabolic status in M1-like but not in M2-like macrophages. Following the interaction with PDA tumor cells, M1-like macrophages are reprogrammed phenotypically to M2-like macrophages. The interaction between M1-like macrophages and PDA cells is mediated by GARP and integrin αV/β8, respectively. Blocking either GARP or integrin would suppress tumor-induced DNA methylation in Nqo-1 gene and the reprogramming of M1-like macrophages. Glucose-response genes such as Il-10 are subsequently activated in tumor-educated M1-like macrophages. Partly through Il-10 and its receptor Il-10R on tumor cells, M1-like macrophages functionally acquire a pro-cancerous capability. Both exogenous M1-like and M2-like macrophages promote metastasis in a mouse model of PDA while such a role of M1-like macrophages is dependent on DNA methylation. Our results suggest that PDA cells are able to reprogram M1-like macrophages metabolically and functionally through a GARP-dependent and DNA methylation-mediated mechanism to adopt a pro-cancerous fate.
