Parkinson’s disease-associated VPs35 mutant reduces mitochondrial membrane potential and impairs PINK1/Parkinmediated mitophagy

Background:Mitochondrial dysfunction plays a prominent role in the pathogenesis of Parkinson’s disease(PD),and several genes linked to familial PD,including PINK1(encoding PTEN-induced putative kinase 1[PINK1])and PARK2(encoding the E3 ubiquitin ligase Parkin),are directly involved in processes such as mitophagy that maintain mitochondrial *** dominant p.D620N variant of vacuolar protein sorting 35 ortholog(VPs35)gene is also associated with familial PD but has not been functionally connected to PINK1 and ***:To better mimic and study the patient situation,we used CRIsPR-Cas9 to generate heterozygous human sH-sY5Y cells carrying the PD-associated D620N variant of *** cells were treated with a protonophore carbonyl cyanide m-chlorophenylhydrazone(CCCP)to induce the PINK1/Parkin-mediated mitophagy,which was assessed using biochemical and microscopy ***:Mitochondria in the VPs35-D620N cells exhibited reduced mitochondrial membrane potential and appeared to already be damaged at steady *** a result,the mitochondria of these cells were desensitized to the CCCPinduced collapse in mitochondrial potential,as they displayed altered fragmentation and were unable to accumulate PINK1 at their surface upon this ***,Parkin recruitment to the cell surface was inhibited and initiation of the PINK1/Parkin-dependent mitophagy was ***:Our findings extend the pool of evidence that the p.D620N mutation of VPs35 causes mitochondrial dysfunction and suggest a converging pathogenic mechanism among VPs35,PINK1 and Parkin in PD.

Relationship between Tropical forest Distribution and soils on Different Types of Mother Rocks in the Republic of Congo

The factors influencing the distribution of forests and their development are important in order to better understand the bio-functioning of tropicals ecosystems forests. The Republic of the Congo has an important forest area of 23.5 million ha subdivided into three large massifs with different forest units from the north until the south of the country. The present study proposes to highlight the relationship between the edaphic and pedological factors and the distribution of the floristic species of some tropical forests of the Congo. To achieve this aim, a principal component analysis (PCA) was to identify similarities or oppositions between variables and to locate the most correlated variables. Also, the indices of biodiversities were used to assess the biodiversity between forest plot and forest sites. A total of 238 species distributed in 46 families were counted. We noted a Cs similarity between Mbomo-Kellé and fMU Mokabi-Dzanga of 50%. However, there was considerable variability between the forests of the Impfondo-Dongou axis and of the forest of other localities. The main component analysis carried out showed that the distribution of floristic species in the studied forests is determined by the edaphic factors.

The Effect of an Acute farmers Walk Exercise Bout on Muscle Damage and Recovery in Recreationally Trained Adults

Purpose The farmer’s Walk(fW)may supplement resistance training through functional tasks like lifting and carrying weight over various *** information exists concerning the intramuscular responses resulting from fW per-formance,possibly impacting its application in exercise ***,the purpose of the study was to investigate Creatine Kinase(CK)and myoglobin(Mb)responses following the farmers Walk Condition(fWC)compared to a control protocol(NWC).Methods fifteen participants(Mean±sEM;age:21.6±0.5 years;height:172.5±2.4 cm;body weight:81.8±4.0 kg)completed an initial session to measure body composition,lower body power,and *** then completed two counter-balanced exercise protocols consisting of a 20-m walk performed within 5 sets of 2 repetitions while either carrying weight(fWC;average wt:85.15±25.55 kg)or not(NWC)with collection of Visually Perceived Muscle soreness(VPMs),Ratings of Perceived Exertion(RPE),and blood ***,blood samples,and Countermovement Jump(CMJ)height were also collected during recovery from each exercise protocol with significance of Psignificant differences were observed between exercise protocols performed including participant RPE(Ps(Psignificant differences were noted for Mb or CMJ *** Training variables implemented during the fWC may have indirectly minimized muscle damage and neuro-muscular inhibitions in *** participants reported mild soreness,the negligible physiological damage suggests the fWC is a safe and appropriate functional movement exercise.

Impact of urinary tract and pulmonary infection on mortality after intracerebral hemorrhage in Brazzaville

Objective: To evaluate the impact of urinary tract and pulmonary infection on mortality after cerebral hemorrhage. Method: We conducted at the University Hospital of Brazzaville, a cross-sectional study from January to August2012 inthe emergency department, neurology and intensive care unit. It included patients admitted for cerebral hemorrhage confirmed by CT-scan. A statistical analysis by logistic regression was carried out to evaluate the correlation between infection and death. Result: Among total of 261 patients for stroke, 82 admitted for cerebral hemorrhage (31.4%). The mean age was 55 ± 11 years (range 26 to 83 years). The sex ratio men/women was 1.7. Hypertension was the most important risk factor to 80.5%. The average intake in neurology time was 28 ± 13 hours. The average time for completion of the CT-scan was 2.4 ± 2 days. Thirty-eight (46.3%) patients had a fever linked to an infectious cause from the third day of hospitalization. The most frequent infectious complications were sepsis (n = 16%;42%), pulmonary infection (n = 14%;37%) and urinary tract infection (n = 8%;21%). specific mortality of infection was 31.7% (n = 26). The multivariate analysis showed a positive correlation between the occurrence of infection and mortality (p = 0.002), specifically between sepsis and mortality (p = 0.0004), and an association between the time of admission late in neurology and the occurrence of infectious complications (p = 0.0001). Conclusion: Infection is one of the dreaded complications of cerebral hemorrhage. It is often associated with delayed care in specialized areas, and is thereby a preventable cause of death.

A Power and Area Optimization Approach of Mixed Polarity Reed-Muller Expression for Incompletely specified Boolean functions

The power and area optimization of Reed-Muller （RM） circuits has been widely concerned. However, almost none of the exiting power and area optimization approaches can obtain all the Pareto optimal solutions of the original problem and are efficient enough. Moreover, they have not considered the don＇t care terms, which makes the circuit performance unable to be further optimized. In this paper, we propose a power and area optimization approach of mixed polarity RM expression （MPRM） for incompletely specified Boolean functions based on Non-Dominated sorting Genetic Algorithm II （NsGA-II）. firstly, the incompletely specified Boolean function is transformed into zero polarity incompletely specified MPRM （IsMPRM） by using a novel IsMPRM acquisition algorithm. secondly, the polarity and allocation of don＇t care terms of IsMPRM is encoded as chromosome. Lastly, the Pareto optimal solutions are obtained by using NsGA-II, in which MPRM corresponding to the given chromosome is obtained by using a chromosome conversion algorithm. The results on incompletely specified Boolean functions and MCNC benchmark circuits show that a significant power and area improvement can be made compared with the existing power and area optimization approaches of RM circuits.
