Memory los. and dementia are major public health concerns.with a s.bs.antial economic *** s.res. has.been s.own to play a crucial role in the pathophys.ology of hippocampal damage-induced memory *** inves.igate whether the antioxidant and anti-inflammatory compound vanillyla cetone(zingerone) can protect agains. hippocampal damage and memory los. induced by cadmium chloride(CdCl_(2)) adminis.ration in rats.we explo red the potential involvement of the nuclear factor erythroid 2-related factor 2(Nrf2) s.gnaling pathway,which is.known to modulate oxidative s.res. and *** healt hy male Wis.ar rats.were divided into five groups.vehicle-treated(control),vanillylacetone,CdCl_(2),vanillylacetone+ CdCl_(2),vanillylacetone+ CdCl_(2)+ brus.tol(a s.lective pharmacological N rf2inhibitor) *** effectively attenuated CdCl_(2)-induced damage in the dental gyrus.of the hippocampus.and improved the memory function as.es.ed by the Morris.Water Maze ***,vanillylacetone markedly decreas.d the hippocampal tis.ue levels.of inflammatory biomarkers.interleukin-6,tumor necros.s.factor-α,intracellular cell adhes.ve molecules. and apoptos.s.biomarkers.Bax and cleaved cas.as.-3).The control and CdCl_(2)-treated groups.treated with va nillylacetone s.owed reduced generation of reactive oxygen s.ecies.decreas.d malondialdehyde levels.and increas.d s.peroxide dis.utas. and glutath.one activities.along with s.gnificant elevation of nuclear Nrf2 mRNA and protein expres.ion in hippocampal *** the protective effects.of vanillylacetone we re s.bs.antially blocked by the co-adminis.ration of brus.tol(a s.lective N rf2 inhibitor).Va nillylacetone mitigated hippocampal damage and memory los. induced by CdCl_(2),at leas. in part, by activating the nuclear trans.ription factor ***,vanillylacetone exerted its.potent antioxidant and antiinflammatory actions.
