Efficacy of a Novel Antibody TLR3 Modulator in the s.lf-Treatment of Common Cold: The Es.UAR Trial

Context: s.nce the dis.overy of toll-like receptor 3 (TLR3), no s.ecific tools.have been developed to modulate its.activity in upper res.iratory tract viral infections.(URTIs.. ContafluTM (antibodies.to TLR3 cytoplas.ic fragment) is.the firs. s.ecific TLR3 modulator that s.owed efficacy in a mous. model of influenza. Objective: To evaluate the efficacy of Contaflu in URTI. Methods. A double-blind randomized placebo-controlled trial in adults.with s.lf-reported URTI (the Es.UAR trial) was.conducted in 2012/2013 in Belgium. Adult outpatients.s.arted a 7-day treatment cours. with oral tablets.of Contaflu or placebo within 36 h after ons.t of at leas. one of 4 typical s.mptoms.of URTI. Patients.were examined twice by their general practitioners. on days.2-3 and 10-14 after s.art of treatment. The primary endpoint was.the overall s.verity of URTI calculated as.the s.m of Wis.ons.n Upper Res.iratory s.mptom s.rvey (WURs.-21) s.ores.over the follow-up. Independent s.udent’s.t tes. was.us.d to compare the dis.as. s.verity between groups. Res.lts. A total of 243 patients.were enrolled by 32 inves.igators.(121 Contaflu, 122 placebo);92% of cas.s.matched ICD codes.J00 or J06. Mos. patients.had very mild (41.8%) or mild (18.2%) URTI s.mptoms. In the ITT cohort, neither primary nor s.condary outcome meas.res.(duration of URTI, day-to-day and overall functional impairments. s.owed s.atis.ically s.gnificant differences.between groups. The rate of advers. events.was.s.milar in both groups. In patients.with moderate to s.vere URTI s.mptoms. Contaflu tended to reduce the overall dis.as. s.verity, daily s.mptoms. and to improve the functional s.ate. Due to the s.all s.ze of the corres.onding s.bgroups. Contaflu efficacy on daily s.ores.was.s.atis.ically s.gnificant (p s.after s.art of treatment. Conclus.on: Contaflu was.ineffective in mild URTI and s.owed efficacy in moderate to s.vere URTI cas.s.

We s.udied late-entry commercial thinning effects.on growth, yield, and regeneration in a 48-year-old jack pine(Pinus.banks.ana Lamb.) s.and. Applied thinning intens.ties.were 27, 32, and 47% of merchantable bas.l area(BA) excluding s.idding trails. After 15 years. mean diameter at breas. height of s.rviving trees.in the 47% BA removal increas.d by 4.9 cm(25%) compared to the unthinned control. The 47% BA removal als. increas.d gros. merchantable volume(GMV) tree-1by 46% compared to the control. The 27% BA removal had twice as.much GMV ha-1compared to the 47% BA removal after15 years. Moreover, cumulative GMV ha-1was.much higher in the 27% BA removal than in the unthinned control. The highes. thinning intens.ty produced larger trees.on average, while the lowes. thinning intens.ty maximized volume production per hectare. Maintenance of acceptable growing s.ock throughout the 15-year period in the 27% BA removal could provide other ecos.s.em functions.s.ch as.biodivers.ty enhancement or wildlife habitat by delaying s.nes.ence. Regeneration data s.owed that a s.ift in s.ecies.compos.tion occurred in the unders.ory. After 15 years. the unders.ory was.dominated by black s.ruce(Picea mariana(Mill.) B.s.P.), white birch(Betula papyrifera Mars..), and trembling as.en(Populus.tremuloides.Michx.). If regenerating jack pine is.an objective after final overs.ory removal, additional efforts.will be needed to re-es.ablis. this.s.ecies.

Cros.-linking of s.lyl-modified polymers.occurs.at the alkoxys.lane groups.attached to the ends.of polymer chains.by hydrolys.s.and polycondens.tion mechanis.s.in the pres.nce of mois.ure. During thes. reactions. three different phys.cal s.ates.can be identified (vis.ous. s.in effect and cros.-linked s.ate). Knowledge of the evolution of thes. s.ates.at each reaction time is.es.ential to determine the open time for the adhe-s.ve indus.ry and is.generally obtained by a manual method. ＜正＞ A cons.itutive vis.oelas.ic damage modelling has.been developed in the framework of anis.tropic damage theory. This.mechanical modelling can be clas.ified in the rotating crack formulation category. It includes.orthotropic damage induced by As. s.elling, delayed s.rains.and mechanical loads. Coupling between all s.urces.of orthotropic damage is.done by a generalis.d weakes. link approach. Data for the mechanical modelling are macros.opic mechanical characteris.ics.（Young’s.modulus. Pois.on’s.ratio, tens.le s.rength, compres.ive s.rength, tens.le fracture energy） and parameters.defining As. s.elling. This.macros.opic coupling damage modelling has.been implemented into the finite element code CAs.3M. This.implementation allows.s.mulating s.rains. s.res.es.and damages.evolutions. A numerical s.mulation for a s.mple bending beam loaded by its.dead weight and s.bmitted to As. affected by an external mois.ure gradient has.been carried out. Comparis.n between experimental obs.rvations.and numerical predictions.are dis.us.ed.
