With the invention of Internet-enabled devices,cloud and blockchain-based technologies,an online voting system can smoothly carry out election *** pandemic situations,citizens tend to develop panic about mass gatherings,which may influence the decrease in the number of *** urges a reliable,flexible,transparent,secure,and cost-effective voting *** proposed online voting system using cloud-based hybrid blockchain technology eradicates the flaws that persist in the existing voting system,and it is carried out in three phases:the registration phase,vote casting phase and vote counting phase.A timestamp-based authentication protocol with digital signature validates voters and candidates during the registration and vote casting *** smart contracts,third-party interventions are eliminated,and the transactions are secured in the blockchain ***,to provide accurate voting results,the practical Byzantine fault tolerance(PBFT)consensus mechanism is adopted to ensure that the vote has not been modified or ***,the overall performance of the proposed system is significantly better than that of the existing *** performance was analyzed based on authentication delay,vote alteration,response time,and latency.

Prostate cancer risk-associated genetic markers and their potential clinical utility

Prostate cancer （PCa） is one of the most common cancers among men in Western developed countries and its incidence has increased considerably in many other parts of the world, including China. The etiology of PCa is largely unknown but is thought to be multifactorial, where inherited genetics plays an important role. In this article, we first briefly review results from studies of familial aggregation and genetic susceptibility to PCa. We then recap key findings of rare and high-penetrance PCa susceptibility genes from linkage studies in PCa families. We devote a significant portion of this article to summarizing discoveries of common and Iow-penetrance PCa risk-associated single-nucleotide polymorphisms （sNPs） from genetic association studies in PCa cases and controls, especially those from genome-wide association studies （GWAss）. A strong focus of this article is to review the literature on the potential clinical utility of these implicated genetic markers. Most of these published studies described PCa risk estimation using a genetic score derived from multiple risk-associated sNPs and its utility in determining the need for prostate biopsy. Finally, we comment on the newly proposed concept of genetic score; the notion is to treat it as a marker for genetic predisposition, similar to family history, rather than a diagnostic marker to discriminate PCa patients from non-cancer patients. Available evidence to date suggests that genetic score is an objective and better measurement of inherited risk of PCa than family history. Another unique feature of this article is the inclusion of genetic association studies of PCa in Chinese and Japanese populations.

The subeconomic Mount Novit Zn-Pb-Ag deposit is located approximately 20 km south of Mount Isa,*** contrast to the nearby Mount Isa,Hilton and George Fisher Zn-Pb-Ag deposits,mineralisation at Mount Novit is situated to the west of the regional-scale Mount Isa Fault and is hosted in the Moondarra siltstone as opposed to the Urquhart ***-grade(sation primarily replaces pre-existing carbonate alteration and veining and consists of pyrrhotite,pyrite and sphalerite with lesser ***-grade(>10 wt.%Zn+Pb)mineralisation occurs as a matrix supported breccia dominated by sphalerite and pyrrhotite with galena,pyrite,and ***-situ U-Pb geochronology was completed on apatite and two textural varieties of ***-grained(subhedral to anhedral monazite is located within highly foliated biotite alteration directly adjacent Zn-Pb-Ag mineralisation and yields a mean weighted^(207)Pb/^(206)Pb age of 1527±18 Ma(MsWD=1.06).This age is consistent with the formation of highly foliated biotite alteration during D;deformation of the Isan *** from the same fabric yields a lower intercept age of1443±29 Ma(MsWD=1.30).Consistent with previous studies,this age is interpreted to represent the age of a major thrusting event along the Mount Isa Fault that resulted in the cooling of the Mount Novit area below~375℃.Coarse-grained monazite is coeval with Zn-Pb-Ag mineralisation and yields a mean weighted^(207)Pb/^(206)Pb age of 1457±11 Ma(MsWD=0.28).sphalerite from Mount Novit has low concentrations(10 ppm),possibly reflecting the leaching of the metals from an underlying basement *** GGIMFis geothermometer(Frenzel et al.,2016)produced a mean formation temperature of 345±52℃.The timing and temperature of Zn-Pb-Ag mineralisation is consistent with the age and cooling temperature of apatite presented in this *** on these correlations,we

An Efficient schema Transformation Technique for Data Migration from Relational to Column-Oriented Databases

Data transformation is the core process in migrating database from relational database to NosQL database such as column-oriented database. However,there is no standard guideline for data transformation from relational database toNosQL database. A number of schema transformation techniques have been proposed to improve data transformation process and resulted better query processingtime when compared to the relational database query processing time. However,these approaches produced redundant tables in the resulted schema that in turnconsume large unnecessary storage size and produce high query processing timedue to the generated schema with redundant column families in the transformedcolumn-oriented database. In this paper, an efficient data transformation techniquefrom relational database to column-oriented database is proposed. The proposedschema transformation technique is based on the combination of denormalizationapproach, data access pattern and multiple-nested schema. In order to validate theproposed work, the proposed technique is implemented by transforming data fromMysQL database to MongoDB database. A benchmark transformation techniqueis also performed in which the query processing time and the storage size arecompared. Based on the experimental results, the proposed transformation technique showed significant improvement in terms query processing time and storagespace usage due to the reduced number of column families in the column-orienteddatabase.

Impact of building regulations on the perceived outdoor thermal comfort in the mixed-use neighbourhood of Chennai

The population in urban areas is increasing rapidly around the world in most of the *** India,this growth has forced the local governments to review the planning norms regularly with the main focus of making an affordable urban *** resulted in increasing the built density without evaluating the effect of such developments on human thermal *** microclimate is one of the significant factors that determine the quality of out-door *** study investigates the potential impact of the built geometry guided by the newly published development control rules of Chennai,India on the various parameters that influence *** existing and future scenario has been modelled for three locations in a typical mixed-use neighborhood of ***,air temperature and relative humid-ity were measured in three locations on a typical summer month of May *** recorded data was used for validating the simulated model and calibrating the model *** the studies on outdoor thermal comfort compare a base case scenario and project future *** paper is a more realistic comparison of outdoor thermal comfort between the actual resultant built environment guided by the new Tamil Nadu Combined Development and Building Rules,2019 for selected locations and the existing built geometry which is the outcome of revised development control rules of *** study found a significant reduction of 18℃in mean radiant temperature(Tmrt)and a reduction of 12℃in Physiological Equiv-alent Temperature(PET)between the 2013 and the predicted built geometry as per 2019 build-ing *** the duration of extreme heat stress in the Physiological Equivalent Temperature(PET)scale show a reduction of 3 h during the day time,The study will assist ur-ban planners and designers to include outdoor thermal comfort also as an important factor while developing building rules.

Population-standardized genetic risk score： the sNP-based method of choice for inherited risk assessment of prostate cancer

several different approaches are available to clinicians for determining prostate cancer （PCa） risk. The clinical validity of various PCa risk assessment methods utilizing single nucleotide polymorphisms （sNPs） has been established; however, these sNP-based methods have not been compared. The objective of this study was to compare the three most commonly used sNP-based methods for PCa risk assessment. Participants were men （n = 1654） enrolled in a prospective study of PCa development. Genotypes of 59 PCa risk-associated sNPs were available in this cohort. Three methods of calculating sNP-based genetic risk scores （GRss） were used for the evaluation of individual disease risk such as risk allele count （GRs-RAC）, weighted risk allele count （GRs-wRAC）, and population-standardized genetic risk score （GRs-Ps）. Mean GRss were calculated, and performances were compared using area under the receiver operating characteristic curve （AUC） and positive predictive value （PPV）. All sNP-based methods were found to be independently associated with PCa （all P 〈 0.05; hence their clinical validity）. The mean GRss in men with or without PCa using GRs-RAC were 55.15 and 53.46, respectively, using GRs-wRAC were 7.42 and 6.97, respectively, and using GRs-Ps were 1.12 and 0.84, respectively （all P 〈 0.05 for differences between patients with or without PCa）. All three sNP-based methods performed similarly in discriminating PCa from non-PCa based on AUC and in predicting PCa risk based on PPV （all P 〉 0.05 for comparisons between the three methods）, and all three sNP-based methods had a significantly higher AUC than family history （all P 〈 0.05）. Results from this study suggest that while the three most commonly used sNP-based methods performed similarly in discriminating PCa from non-PCa at the population level, GRs-Ps is the method of choice for risk assessment at the individual level because its value （where 1.0 represents average population risk） can be easily interpreted regardless of

Inflammatory bowel disease(IBD),including Crohn’s disease(CD)and ulcerative colitis(UC),is a chronic inflammatory disease of the digestive tract with increasing prevalence *** venous thromboembolism(VTE)is a major complication in IBD patients,it is often underappreciated with limited tools for risk *** To estimate the proportion of VTE among IBD patients and assess genetic risk factors(monogenic and polygenic)for *** Incident VTE was followed for 8465 IBD patients in the UK Biobank(UKB).The associations of VTE with F5 factor V leiden(FVL)mutation,F2 G20210A prothrombin gene mutation(PGM),and polygenic score(PGs003332)were tested using Cox hazards regression analysis,adjusting for age at IBD diagnosis,gender,and genetic background(top 10 principal components).The performance of genetic risk factors for discriminating VTE diagnosis was estimated using the area under the receiver operating characteristic curve(AUC).REsULTs The overall proportion of incident VTE was 4.70%in IBD patients and was similar for CD(4.46%),UC(4.49%),and unclassified(6.42%),and comparable to that of cancer patients(4.66%)who are well-known at increased risk for *** carriers of F5/F2 had a significantly increased risk for VTE compared to non-mutation carriers,hazard ratio(HR)was 1.94,95%confidence interval(CI):*** contrast,patients with the top PGs decile had a considerably higher risk for VTE compared to those with intermediate scores(middle 8 deciles),HR was 2.06(95%CI:1.57-2.71).The AUC for differentiating VTE diagnosis was 0.64(95%CI:0.61-0.67),0.68(95%CI:0.66-0.71),and 0.69(95%CI:0.66-0.71),respectively,for F5/F2 mutation carriers,PGs,and *** similar to cancer patients,VTE complications are common in IBD *** provides more informative risk information than F5/F2 mutations(FVL and PGM)for personalized thromboprophylaxis.

An efficient method to enhance recovery and detection of sARs-CoV-2 RNA in wastewater

Wastewater surveillance(Ws)of sARs-CoV-2 currently requires multiple steps and suffers low recoveries and poor ***,we report an improved analytical method with high sensitivity and recovery to quantify sARs-CoV-2 RNA in *** improve the recovery,we concentrated sARs-CoV-2 viral particles and RNA from both the solid and aqueous phases of wastewater using an electronegative membrane(EM).The captured viral particles and RNA on the EM were incubated in our newly developed viral inactivation and RNA preservation(VIP)***,the RNA was concentrated on magnetic beads and inhibitors removed by *** eluting,the RNA on the magnetic beads was directly detected using reverse transcription quantitative polymerase chain reaction(RT-qPCR).Analysis of sARs-CoV-2 pseudovirus(sARs-CoV-2 RNA in a noninfectious viral coat)spiked to wastewater samples showed an improved recovery of 80%.Analysis of 120 wastewater samples collected twice weekly between May 2021 and February 2022 from two wastewater treatment plants showed 100%positive detection,which agreed with the results independently obtained by a provincial public health *** concentrations of sARs-CoV-2 RNA in these wastewater samples ranged from 2.4×10^(2) to 2.9×10^(6) copies per 100 mL of *** method’s capability of detecting trace and diverse concentrations of sARs-CoV-2 in complex wastewater samples is attributed to the enhanced recovery of sARs-CoV-2 RNA and efficient removal of PCR *** improved method for the recovery and detection of viral RNA in wastewater is important for wastewater surveillance,complementing clinical diagnostic tests for public health protection.

Alcohol consumption is one of the leading causes of the global burden of disease and results in high healthcare and economic *** alcohol misuse leads to alcohol-related liver disease,which is responsible for a significant proportion of alcohol-attributable deaths *** than reducing alcohol consumption,there are currently no effective treatments for alcohol-related liver *** stress refers to an imbalance in the production and elimination of reactive oxygen species and *** plays important roles in several aspects of alcohol-related liver disease ***,we review how chronic alcohol use results in oxidative stress through increased metabolism via the cytochrome P4502E1 system producing reactive oxygen species,acetaldehyde and protein and DNA *** trigger inflammatory signaling pathways within the liver leading to expression of pro-inflammatory mediators causing hepatocyte apoptosis and *** oxygen species exposure also results in mitochondrial stress within hepatocytes causing structural and functional dysregulation of mitochondria and upregulating apoptotic *** is also evidence that oxidative stress as well as the direct effect of alcohol influences epigenetic *** global histone methylation and acetylation and specific histone acetylation inhibits antioxidant responses and promotes expression of key pro-inflammatory *** review highlights aspects of the role of oxidative stress in disease pathogenesis that warrant further study including mitochondrial stress and epigenetic *** understanding of these processes may identify novel targets for therapy.

Andrographolide inhibits chikungunya virus infection by up-regulating host innate immune pathways

Objective: To investigate the therapeutic efficacy of andrographolide, a plant derived compound, against chikungunya virus(CHIKV) infection. Methods: Using flow cytometry and immunoblotting assay, in vitro viral protein expression was studied in THP-1 cells line. In Balb/c mouse neonates, viral RNA copy number was determined by real time PCR. Results:The results showed reduced CHIKV protein expression on andrographolide treatment in CHIKV-infected human peripheral blood mononuclear cells, Vero cells and THP-I cell *** vivo, andrographolide treatment to CHIKV-infected neonates reduced viral RNA copy number. Further. andrographolide also increased cytotoxic T lymphocytes both in vitro and in vivo. Andrographolide also activated host innate immune pathways, viz., protein kinase *** eukaryotic initiation factor 2 α, retinoic acid inducible gene-Ⅰ and interferon regulatory factor 3/7, thereby increasing IFN-a secretion. CHIKV-induced nuclear factor κlight chain enhancer of activated B cells and tumor necrosis factor-a was also reduced on andrographolide treatment. Conclusion: Andrographolide inhibits CHIKV by suppressing viral protein expression and up-regulating host innate immunity and hence could be an effective therapeutic agent against CHIKV infection.
