Climate refugia can serve as a remnant habitat or stepping stones for species dispersal under climate *** largest freshwater lake by surface area,Lake superior,UsA and Canada,serves as a model system for understanding cooling-mediated local refugia,as its cool water temperatures and wave action have maintained shoreline habitats suitable for southern disjunct populations of arctic-alpine plants since ***,we seek to explain spatial patterns and environmental drivers of arctic-alpine plant refugia along Lake superior’s shores,and assess future risk to refugia under moderate(+3.5℃)and warmest(+5.7℃)climate warming ***,we examined how the interactive effects of summer surface water temperatures and wind affected onshore temperatures,resulting in areas of cooler ***,we developed an ecological niche model for the presence of disjunct arctic-alpine refugia(pooling 1253 occurrences from 58 species)along the lake’s ***,we fit species distribution models for 20 of the most common arctic-alpine disjunct species and predicted presence to identify refugia ***,we used the two climate warming scenarios to predict changes in the presence of refugia and disjunct *** type,elevation above water,inland distance,July land surface temperature from MODIs/Terra satellite and near-shore depth of water were the best predictors of disjunct ***,we predicted 2236 km of the shoreline(51%)as disjunct refugia habitat for at least one species under current conditions,but this was reduced to 20% and 7% with moderate(894 km)and warmest(313 km)climate change projections.

Dynamic wheel-rail contact forces induced by a severe form of wheel tread damage have been measured by a wheel impact load detector during full-scale field tests at different vehicle *** on laser scanning,the measured three-dimensional damage geometry is employed in simulations of dynamic vehicle-track interaction to calibrate and verify a simulation *** relation between the magnitude of the impact load and various operational parameters,such as vehicle speed,lateral position of wheel-rail contact,track stiffness and position of impact within a sleeper bay,is *** calibrated model is later employed in simulations featuring other forms of tread damage;their effects on impact load and subsequent fatigue impact on bearings,wheel webs and subsurface initiated rolling contact fatigue of the wheel tread are *** results quantify the effects of wheel tread defects and are valuable in a shift towards condition-based maintenance of running gear,and for general assessment of the severity of different types of railway wheel tread damage.

The tumor microenvironment is composed of a complex mixture of different cell types interacting under conditions of nutrient deprivation,but the metabolism therein is not fully understood due to difficulties in measuring metabolic fluxes and exchange of metabolites between different cell types in ***-scale metabolic modeling enables estimation of such exchange fluxes as well as an opportunity to gain insight into the metabolic behavior of individual cell ***,we estimated the availability of nutrients and oxygen within the tumor microenvironment using concentration measurements from blood together with a metabolite diffusion *** addition,we developed an approach to efficiently apply enzyme usage constraints in a comprehensive metabolic model of human *** combined modeling reproduced severe hypoxic conditions and the Warburg effect,and we found that limitations in enzymatic capacity contribute to cancer cells’preferential use of glutamine as a substrate to the citric acid ***,we investigated the common hypothesis that some stromal cells are exploited by cancer cells to produce metabolites useful for the cancer *** identified over 200 potential metabolites that could support collaboration between cancer cells and cancer-associated fibroblasts,but when limiting to metabolites previously identified to participate in such collaboration,no growth advantage was *** work highlights the importance of enzymatic capacity limitations for cell behaviors and exemplifies the utility of enzyme-constrained models for accurate prediction of metabolism in cells and tumor microenvironments.

Background:The direct use of medical zinc oxide in feed will be abandoned after 2022 in Europe,leaving an urgent need for substitutes to prevent post-weaning ***:This study investigated the effect of using rapeseed-seaweed blend(rapeseed meal added two brown macroalgae species Ascophylum nodosum and saccharina latissima)fermented by lactobacilli(FRs)as feed ingredients in piglet *** d 28 of life to d 85,the piglets were fed one of three different feeding regimens(n=230 each)with inclusion of 0%,2.5% and 5% *** this period,no significant difference of piglet performance was found among the three *** a subset of piglets(n=10 from each treatment),blood samples for hematology,biochemistry and immunoglobulin analysis,colon digesta for microbiome analysis,and jejunum and colon tissues for histopathological analyses were *** piglets fed with 2.5% FRs manifested alleviated intraepithelial and stromal lymphocytes infiltration in the gut,enhanced colon mucosa barrier relative to the 0% FRs *** colon microbiota composition was determined using V3 and V1-V8 region 16s rRNA gene amplicon sequencing by Illumina Nextseq and Oxford Nanopore MinION,*** two amplicon sequencing strategies showed high consistency between the detected *** sequencing strategies indicated that inclusion of FRs reshaped the colon microbiome of weaned piglets with increased shannon *** stercorea was verified by both methods to be more abundant in the piglets supplied with FRs feed,and its abundance was positively correlated with colonic mucosa thickness but negatively correlated with blood concentrations of leucocytes and ***:FRs supplementation relieved the gut lymphocyte infiltration of the weaned piglets,improved the colon mucosa barrier with altered microbiota *** the dietary inclusion of FRs from 2.5% to 5% did not lead to further improvements.

sortilin was discovered in 1997 by Petersen and his *** single transmembrane receptor was isolated as a 95 kDa membrane glycoprotein by receptor-associated protein affinity chromatography and therefore initially named *** cDNA library screening revealed sequence homology between the luminal segment of gp95/sortilin and the yeast vacuolar sorting protein 10 protein(Vps10p)domain(Petersen et al.,1997).The Vps10p domain was first identified in the saccharomyces cerevisiae protein Vps10p that directs lysosomal enzymes from the Golgi to the vacuole(Marcusson et al.,1994).
