A key adaptation in insects f.r dealing with variable environmental conditions is the ability to diapause. The tiger swallowtail butterf.ies, Papilio glaucus and P. canadensis are ideal species to explore the genetic causes and population genetic consequences of.diapause because divergence in this trait is believed to be a salient f.ctor in maintaining a hybrid zone between these species. Yet little is known about the f.ctors that inf.uence diapause induction in this system. Here we explored how spatial （latitudinal）, environmental （temperature） and genetic （hybridization） f.ctors af.ect diapause induction in this system. Specif.cally, a series of.growth chamber experiments using wild caught individuals f.om across the eastern United States were perf.rmed to：（1） evaluate how critical photoperiod varies with latitude, （2） isolate the stage in which induction occurs, （3） test whether changes in temperature af.ected rates of.diapause induction, and （4） explore how the incidence of.diapause is af.ected in hybrid of.spring. We f.nd that induction occurs in the larval stage, is not sensitive to a relatively broad range of.temperatures, appears to have a complex genetic basis （i.e., is not simply a dominant trait f.llowing a Mendelian inheritance pattern） and that the critical photoperiod increases by 0.4 h with each increasing degree in latitude. This work deepens our understanding of.how spatial, environmental and genetic variation inf.uences a key seasonal adaptation （diapause induction） in a well-developed ecological model system and will make possible f.ture studies that explore how climatic variation af.ects the population dynamics and genetics of.this system.

Smoking cessation prior to elective total joint arthroplasty results in sustained abstinence postoperatively

BACKGROUND Tobacco use is a well-documented modif.able risk f.ctor f.r perioperative *** To determine the tobacco abstinence rates of.patients who made cessation ef.orts prior to a total joint arthroplasty(TJA)*** A retrospective evaluation was perf.rmed on 88 self.reported tobacco users who underwent TJA between 2014-2022 and had tobacco cessation dates within 3 mo of.*** patients were contacted via phone survey to understand their tobacco use pattern,and patient reported outcomes.A total of.37 TJA patients *** Our cohort was on average 61-years-old,60%(n=22)women,with an average body mass index of.30 kg/m^(2).The average f.llow-up time was 2.9±1.9 years.A total of.73.0%(n=27)of.patients endorsed complete abstinence f.om tobacco use prior to *** cessation methods were used perioperatively including prescription therapy(13.5%),over the counter nicotine replacement(18.9%),cessation programs(5.4%).At f.nal f.llow up,43.2%(n=16)of.prior tobacco smokers reported complete *** who were able to maintain cessation postoperatively had improved Patient-Reported Outcomes Measurement Inf.rmation System(PROMIS)-10 mental health scores(49 vs 58;P=0.01),and hip dysf.nction and osteoarthritis outcome score f.r joint replacement(***)scores(63 vs 82;P=0.02).No patients in this cohort had a prosthetic joint inf.ction or required revision *** We report a tobacco cessation rate of.43.2%in patients undergoing elective TJA nearly 3 years *** undergoing TJA who were able to remain abstinent had improved PROMIS-10 mental health scores and *** *** perioperative period provides clinicians a unique opportunity to assist active tobacco smokers with cessation ef.orts and improve postoperative outcomes.

Realizing the f.ll potential of.ultrahigh-intensity lasers f.r particle and radiation generation will require multi-beam arrangements due to technology ***,we investigate how to optimize their coupling with solid ***,we show that overlapping two intense lasers in a mirror-like conf.guration onto a solid with a large preplasma can greatly improve the generation of.hot electrons at the target f.ont and ion acceleration at the target *** underlying mechanisms are analyzed through multidimensional particle-in-cell simulations,revealing that the self.induced magnetic f.elds driven by the two laser beams at the target f.ont are susceptible to reconnection,which is one possible mechanism to boost electron *** addition,the resistive magnetic f.eld generated during the transport of.the hot electrons in the target bulk tends to improve their *** simulations also indicate that such ef.ects can be f.rther enhanced by overlapping more than two laser beams.

Assessment of.Weight and Health in the Emergency Department: A Cross-Sectional Study

Introduction: The prevalence of.obesity and obesity-related diseases continues to rise. A key aspect of.prevention and treatment of.these disorders requires clear communication about weight and health between patients and healthcare providers. Objective: We sought to examine the prevalence rate of.obesity and associated comorbid illnesses in an emergency department (ED) population. A second aim was to assess patients’ perceptions of.their weight and their overall health. Methods: This is a cross-sectional study perf.rmed in an academic tertiary-care center using a representative sample of.patients (≥18 yr) who presented to the ED. Pregnant patients, patients who were medically unstable, cognitively impaired or who were unable or unwilling to provide inf.rmed consent were excluded. Anthropometric measurements were taken which include BMI (the ratio of.a patient’s weight and height expressed as kg/m2) and waist circumf.rence. In addition, the prevalence rates of.all enrolled patients who 1) f.el their health is af.ecting their weight and 2) who have had or recall discussions about their health and weight with their provider were examined by using a two-question validated survey. Results: The overall prevalence rate of.obesity in this study was 38.6%. Only 71.8% (95% CI, 63.2% - 80.6%) of.overweight patients (BMI = 25.0 - 29.9) and 28.4% (95% CI, 21.6% - 35.2%) of.obese patients (BMI ≥ 30.0) believe their present weight is damaging to their health. f.rther, only 15.5% (95% CI, 8.5% - 22.6%) and 59.4% (95% CI, 53% - 67%) of.those overweight and obese, respectively, recall being inf.rmed by their healthcare provider they are overweight. f.r morbidly obese patients (BMI ≥ 40.0), 79.6% (95% CI, 12.2 - 28.6) f.el their weight is damaging their health and 83.7% (95% CI, 9.4 - 23.2) recall having a conversation about weight with their healthcare provider. Vesicular nanocarrier f.rmulations conf.r the ability to deliver hydrophobic and hydrophilic cargos simultaneously to cells of.interest in vivo. While liposomal f.rmulations reached the clinic long ago, younger technologies such as polymeric vesicles （polymersomes） have yet to make the transition to clinical approval and use, in part due to dif.iculties in ensuring their saf. and scalable production. In this work, we demonstrate the scalable production of.poly（ethylene glycol）-block-poly（propylene sulf.de）（PEG-bl-PPS） polymersomes via f.ash nanoprecipitation, and f.rther show the saf. administration of.these nanocarriers to mice and non-human primates. In mice, PEG-bl-PPS polymersomes were f.und to be well tolerated at up to 200 mg/（***）. f.llowing the administration of.a more relevant 20 mg/（***） dosage in non-human primates, polymersomes were f.und to associate with numerous phagocytic immune cell populations, including a remarkable 68% of.plasmacytoid dendritic cells and 〉 95% of.macrophages in the spleen, while showing no toxicity or abnormalities in the liver, kidney, spleen, or blood. Despite the presence of.a dense PEG corona, neither anti-PEG antibodies nor complement activation were detected. This work provides evidence of.the translatability of.PEG-bI-PPS polymersomes into the clinic f.r therapeutic applications in humans.

Sensory cell damage is a major cause of.hearing loss： Sensorineural hearing loss （SNHL） is a common sensory def.cit characterized by tissue damage and/or cell death in the cochlea. Moderate and severe SNHL causes def.cits in communication, associated with increased isolation f.om human relationships, and negativly impacts employabf.ity. Many etiologies have been associated with SNHL： Noise overexposure, certain drugs, inner or middle ear inf.ction or immune-induced inf.ammation being common. However, the most prevalent f.rm of.SNHL is presbycusis or age-related hearing loss, of.en aggravated by other f.ctors including a history of.noise exposure, diabetes or high blood pressure （Wong and ryan, 2015）. Though systemic steroids are used in the treatment of.sudden SNHL, other drugs to prevent or cure hearing loss have not yet been developed. Cochlear sensory cells, known as hair cells （HCs） and neurons have very limited repair capacity. Moreover since these cells do not regenerate, making any loss permanent, prevention of.cell damage is critical to protecting hearing.

Estimating potential harvestable biomass f.r bioenergy f.om sustainably managed private native f.rests in Southeast Queensland, Australia

Background： Australia＇s energy f.ture is at the crossroads and the role of.renewable sources is in f.cus. Biomass f.om sustainably managed f.rests provide a signif.cant opportunity f.r electricity and heat generation and production of.liquid f.els. Australia has extensive native f.rests of.which a signif.cant proportion are on private land. However, there is limited knowledge on the potential capacity of.this resource to contribute to the expansion of.a biomass f.r bioenergy industry. In addition, there are concerns on how to reconcile biomass harvesting with environmental protection. Methods： We used regional ecosystem vegetation mapping f.r Queensland to stratif. harvestable f.rests within the 1.8 m hectares of.private native f.rests present in the Southeast Queensland bioregion in 2014. We used a dataset of.52,620 individual tree measurements f.om 541 f.rest inventory plots collected over the last 10 years. Tree biomass was estimated using current biomass allometric equations f.r Australia. Biomass potentially available f.om selective sawlog harvesting and silvicultural treatment across the bioregion was calculated and mapped. Results： Current sawlog harvesting extracts 41.4% of.the standing tree biomass and a biomass f.r bioenergy harvest would retain on average 36% of.f.lled tree biomass on site f.r the protection of.environmental and f.una habitat values. The estimated area extent of.harvestable private native f.rests in the bioregion in 2013 was 888,000 ha and estimated available biomass f.r bioenergy in living trees was 13.6 million tonnes （t）. The spotted gum （Corymbio citriodora subsp, variegata） f.rests were the most extensive, covering an area of.379,823 ha and with a biomass f.r bioenergy yield of.14.2 t-ha-1 （with approximately 11.2 ***-1 of.the biomass harvested f.om silvicultural thinning and 3 ***-1 recovered f.om sawlog harvest residual）. Validation of.a radioimmunoassay-based f.cal corticosteroid assay f.r Richardson＇s ground squirrels Urocitellus richardsonii and behavioural correlates of.stress

We validated a radioimmunoassay-based method quantif.ing f.cal glucocorticoid metabolites （f.Ms） f.om captive male and f.male Richardson＇s ground squirrels Urocitellus richardsonii. Blood samples were drawn to explore the correlation between plasma cortisol and f.M concentrations. We also injected groups of.squirrels with normal saline （CTL; control）, adre- nocorticotropic hormone （ACTH; stimulating adrenal activity）, or dexamethasone （DEX; suppressing adrenal activity）. Potential correlations between stress and behaviour were explored through quantif.cation of.f.cal pellet production and the intervention necessary to elicit def.cation, as well as the behaviour of.subjects in the context of.handling. Changes in plasma cortisol concen- tration between capture （baseline）, and f.llowing handling （stress-induced） were also quantif.ed f.r f.ee-living squirrels. While glucocorticoid concentrations recovered f.om f.ces during our captive-animal study were not well correlated with plasma eortisol concentrations, and uncorrelated with def.cation or behaviour, f.M concentrations did ref.ect the activation of.the hypothalamic- pituitary-adrenal （HPA） axis. f.M concentrations increased signif.cantly during initial captivity, but declined to baseline level as individuals acclimated to the novel environment. Injection of.subjects with ACTH increased f.Ms above baseline, conf.rming activation of.the HPA axis. Plasma cortisol concentrations increased signif.cantly with induced stress, indicating that capture and handling activated the glucocorticoid stress response even among previously handled, f.ee-living subjects. Our f.ndings validate a non-invasive tool that will af.ord new insight into the physiological processes underlying social, reproductive and antipredator behaviour of.Richardson＇s ground squirrels [Current Zoology 60 （5）： 591-601, 2014 ].
