Investigation of the feasibility of NRAV as a biomarker for hepatocellular carcinoma

The long non-coding RNA,Negative Regulator of Antiviral Response(NRAV)has been identified as a participant in both respiratory virus replication and immune checkpoints,however,its involvement in pan-cancer immune regulation and prognosis,particularly those of hepatocellular carcinoma(HCC),remains *** address this knowledge gap,we analyzed expression profiles obtained from The Cancer Genome Atlas(TCGA)database,comparing normal and malignant tumor *** found that NRAV expression is significantly upregulated in tumor tissues compared to adjacent nontumor ***-Meier(K-M)analysis revealed the prognostic power of NRAV,wherein overexpression was significantly linked to reduced overall survival in a diverse range of tumor ***,noteworthy associations were observed between NRAV,immune checkpoints,immune cell infiltration,genes related to autophagy,epithelial-mesenchymal transition(EMT),pyroptosis,tumor mutational burden(TMB),and microsatellite instability(MSI)across different cancer types,including ***,NRAV upregulation expression was associated with multiple pathological stages by clinical ***,our investigation revealed a substantial elevation in the expression of NRAV in both HCC tumor tissues and cells compared to normal tissues and *** inhibition of NRAV resulted in the inhibition of cell proliferation,migration,and invasion in HCC cells,while also influencing the expression of CD274(PD-L1)and CD44,along with various biomarkers associated with EMT,autophagy,and *** aforementioned results propose NRAV as a promising prognostic biomarker for HCC.

目的探索后生元那曲4580对慢性束缚应激(chronic restraint stress,CRS)小鼠睡眠障碍的改善作用,为后生元制剂的功效和应用提供理论依据。方法将225只6~8周龄、体重18~22 g SPF级C57BL/6J雄性小鼠随机分为空白对照组、CRS模型组及实验组(低剂量组、中剂量组和高剂量组),每组75只。CRS模型组和实验组连续21 d进行CRS造模,空白对照组不进行造模。实验组小鼠分别灌胃低剂量[1 g/(kg·bw·d)]、中剂量[2 g/(kg·bw·d)]、高剂量[3 g/(kg·bw·d)]后生元那曲4580,其余组灌胃生理盐水,灌胃持续30 d。在灌胃结束后均接受糖水偏好实验、直接睡眠实验、巴比妥钠睡眠潜伏期实验、戊巴比妥钠阈下剂量催眠实验及延长戊巴比妥钠睡眠时间实验。实验结束后收集小鼠胸腺、肝脏、脾脏,称重并计算脏器系数。结果经连续21 d造模后,与空白对照组相比,CRS模型组小鼠体重下降显著(t=6.444,P=0.001)且糖水偏好指数也显著降低(t=2.229,P=0.029)。对比CRS模型组,后生元那曲4580低剂量组小鼠体重增加显著(t=2.347,P=0.022);实验组小鼠糖水偏好指数均有所上升。在直接睡眠实验中,所有小鼠均未进入睡眠状态。在巴比妥钠睡眠潜伏期实验中,与CRS模型组相比,后生元那曲4580高剂量组的睡眠潜伏期显著增加(t=-2.575,P=0.018)。在戊巴比妥钠阈下剂量催眠实验中,后生元那曲4580中剂量组的入睡小鼠数量和入睡率显著增加(Fisher确切概率,P=0.025)。结论后生元那曲4580不具有直接催眠的作用,但具有改善CRS小鼠的焦虑和抑郁以及睡眠障碍的作用,以中剂量改善睡眠的效果较佳。

目的探究两歧双歧杆菌BD-1(Bifidobacterium bifidum BD-1,BD-1)以2-岩藻糖基乳糖(2'-fucosyllactose,2'-FL)作为唯一碳源时,与葡萄糖相比,其菌体和代谢产物诱导小鼠巨噬细胞系RAW264.7细胞分泌细胞因子的效应,以探索BD-1菌利用母乳低聚糖后免疫调节的菌株特性。方法(1)菌体部分干预实验:将BD-1活菌接种于不同碳源的改良MRS肉汤培养基中,改良过的MRS肉汤培养基包括以2%2'-FL作为唯一碳源(2'-FL组)和以2%葡萄糖作为唯一碳源(Glu组)的培养基。将接种后的BD-1活菌在厌氧条件下培养24 h后,分离各组菌体和培养基上清液。将菌体部分与RAW264.7细胞共同培养,并以肽聚糖(peptidoglycan,PGN)为菌体成分对照,同时设置不添加干预物的阴性对照(Ctrl组)。(2)菌体代谢产物部分干预实验:将培养基上清液与RAW264.7细胞共同培养,以含有2%2'-FL或葡萄糖但未进行BD-1菌发酵处理的培养基作为实验对照,并设置不添加干预物的培养基与RAW264.7细胞的共培养上清液作为阴性对照(Ctrl组),以PGN与RAW264.7细胞的共培养上清液作为阳性对照(PGN组)。培养24 h后,提取RAW264.7细胞总RNA,逆转录后采用实时荧光定量PCR检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素(interleukin,IL)-6、IL-10、IL-12、IL-1β的基因表达量;收集各组受试物与RAW264.7细胞的共培养上清液,采用酶联免疫吸附试验检测共培养上清液中上述细胞因子的分泌量。结果(1)菌体部分干预:与PGN组相比,菌体部分2'-FL组和Glu组的TNF-α、IL-6、IL-10分泌量及基因表达量显著上升(均Plu组相比,2'-FL组的IL-6、IL-10分泌量显著下降(均Plu上清组的TNF-α、IL-6及IL-10分泌水平以及基因表达水平较2'-FL组和Glu组(未进行BD-1菌发酵处理的组别)有显著提高(均Plu组显著增加(均Plu组能产生更多的有机酸和种类更为丰富的糖类物质。结论利用母乳低聚糖或葡萄糖作为唯一碳源培养BD-1菌体和诱导巨噬细胞分泌细胞因子的特征存在差异。当BD-1以母乳低聚糖为唯一碳源时,与葡萄糖相比,菌体成分呈现出抗炎倾向,在巨噬细胞中表现为IL-6的分泌和基因表达能力下降;而其代谢产物则具有刺激免疫激活的趋势,在巨噬细胞中导致IL-6和IL-10的分泌及基因表达能力上升。

The Fuling shale gas field in China is the largest shale gas field as well as the largest of its type discovered in any Lower Paleozoic *** this study,the geology and production of the upper and lower gas layers in the Fuling shale gas field are evaluated in terms of structure,shale quality,fault,initial production,and estimated ultimate recovery(EUR).The shale in the lower gas layer of the Jiaoshiba anticline is a high-quality reservoir,where the space is dominated by organic pores in kerogen,and the gas content is *** shale gas wells reveal relatively high initial production and ***,the shale in the upper gas layer of the Jiaoshiba anticline has reservoir space mainly composed of clay mineral pores and organic pores within bitumen,and the gas content is *** terms of structure,primary gas migration may occur in the upper gas layer,resulting in free gas accumulation in the structural high,where the development effects are generally better than those in the structural *** lower gas layer in the Pingqiao anticline,is the main interval for shale gas accumulation and development due to the high-quality *** the influence of faults,the efficiency of exploration wells emplaced on top of the anticline is much lower a compared with those in the *** residual synclines close to the Sichuan Basin,including the Baima and Baitao anticlines,are characterized by more recent uplifts,larger area,greater distance from the deep and large faults,and early fracture ***,we recommend that the shale gas exploration and development should be carried out preferentially in areas close to the center of the residual synclines,featuring relatively high-pressure coefficient and moderate burial depth.

Traditional visual interpretation is often inefficient due to its excessively workload professional knowledge and strong ***,building an automatic interpretation model on high spatial resolution remote sensing images is the key to the quick and efficient interpretation of earthquake-triggered *** at addressing this problem,a landslide interpretation model of high-resolution images based on bag of visual word(BoVW)feature was *** high-resolution images were pre-processed,and then BoVW feature and support vector machine(SVM)was adopted to establish an automatic landslide interpretation *** model was further compared with the currently widely used Histogram of Oriented Gradient(HoG)feature extraction *** order to test the effectiveness of the method,typical landslide images were selected to construct a landslide sample library,which was subsequently utilized as the foundation for conducting an experimental *** results show that the accuracy of landslide extraction using this method reaches as high as 89%,indicating that the method can be used for the automatic interpretation of landslides in disaster-prone areas,and has high practical value for regional disaster prevention and damage reduction.

目的了解农村留守和非留守儿童照护人对儿童非免疫规划(Expanded Program on Immunization,EPI)疫苗接种的意愿和影响因素。方法选取中国两个省部分农村地区1-6岁留守和非留守儿童的照护人开展问卷调查,分析调查对象对儿童非EPI疫苗接种的意愿及其影响因素。结果留守和非留守儿童照护人对儿童非EPI疫苗接种的意愿率分别为85.53%(721/843)、86.03%(751/873)。多因素Logistic回归分析显示,家庭人均年收入高(5千元-19千元vs<5千元:OR=2.56,95%CI:1.54-4.27;≥20千元vs<5千元:OR=2.85,95%CI:1.47-5.51)、同意接种非EPI疫苗有效(OR=2.62,95%CI:1.66-4.13)和有必要(OR=4.58,95%CI:2.92-7.19)、父母外出经常询问儿童疫苗接种(OR=2.21,95%CI:1.10-4.43)和给家补贴生活费(OR=2.20,95%CI:1.37-3.52)的留守儿童照护人的意愿率更高;知道自费与免费疫苗区别(OR=3.61,95%CI:1.76-7.40)、同意接种非EPI疫苗有必要(OR=7.58,95%CI:4.95-11.61)的非留守儿童照护人的意愿率更高。结论调查地区留守和非留守儿童照护人对儿童接种非EPI疫苗接种的意愿较高;需加强非EPI疫苗接种相关知识健康教育,促进儿童非EPI疫苗接种。
