Translocation of IGF-1R in endoplasmic reticulum enhances SERCA2 activity to trigger Ca_(ER)^(2+)perturbation in hepatocellular carcinoma

The well-known insulin-like growth factor 1(IGF1)/IGF-1 receptor(IGF-1R)signaling pathway is overexpressed in many tumors,and is thus an attractive target for cancer ***,results have often been disappointing due to crosstalk with other ***,we report that IGF-1R signaling stimulates the growth of hepatocellular carcinoma(HCC)cells through the translocation of IGF-1R into the ER to enhance sarco-endoplasmic reticulum calcium ATPase 2(SERCA2)*** response to ligand binding,IGF-1Rβis translocated into the ER byβ-arrestin2(β-arr2).Mass spectrometry analysis identified SERCA2 as a target of ER IGF-1Rβ.SERCA2 activity is heavily dependent on the increase in ER IGF-1Rβ*** IGF-1Rβphosphorylates SERCA2 on Tyr^(990)to enhance its *** of SERCA2-Tyr^(990)disrupted the interaction of ER IGF-1Rβwith SERCA2,and therefore ER IGF-1Rβfailed to promote SERCA2 *** enhancement of SERCA2 activity triggered Ca_(ER)^(2+)perturbation,leading to an increase in *** blocked the interaction between SERCA2and ER IGF-1Rβand therefore SERCA2 activity,resulting in inhibition of HCC *** conclusion,the translocation of IGF-1R into the ER triggers Ca_(ER)^(2+)perturbation by enhancing SERCA2 activity through phosphorylating Tyr^(990)in HCC.

Constraints on b.ane inflation after Planck 2015: Impacts of the latest local measurement of the Hubble constant

We investigate the observational constraints on three typical b.ane inflation models by considering the latest local measurement of the Hubble constant in the global fit. We also employ other observational data, including the Planck 2015 CMb data, the bICEP2/Keck Array b-mode data, and the baryon acoustic oscillations data, in our analysis. Previous studies have shown that the addition of the latest local H_0 measurement favors a larger spectral index, and can exert a significant influence on the model selection of inflation. In this work, we investigate its impacts on the status of b.ane inflation models. We find that, when the direct H_0 measurement is considered, the prototype model of b.ane inflation is still in good agreement with the current observational data within the 2σ level range. For the KKLMMT model, the consideration of the H_0 measurement allows the range of the parameter β to be amplified to O(10^(-2)), which slightly alleviates the fine-tuning problem. For the IR DbI model, the addition of the H_0 measurement does not provide a better fit. These results show that the consideration of the new H_0 prior can exert a considerable influence on the b.ane inflation models. At last, we show that, when β≤1.1, the equilateral non-Gaussianity in the IR DbI inflation model is compatible with the current CMb data at the 1σ level.

Micromagnetic simulations of reversal magnetization in cerium-containing magnets

Single-grain models with different cerium contents or structural parameters have been introduced to investigate the reversal magnetization behaviors in cerium-containing magnets. All the micromagnetic simulations are carried out via the object oriented micromagnetic framework(OOMMF). As for single(Nd,Ce)_2 Fe_(14)b type grain, the coercivity decreases monotonously with the increase of the cerium content. Four types of grain structure have been compared: single(Nd,Ce)_2 Fe_(14)b type, core((Nd,Ce)_2 Fe_(14)b)-shell(Nd_2 Fe_(14)b) type with 2 nm thick shell, core(Ce_2 Fe_(14)b)-shell(Nd_2 Fe_(14)b) type, and core(Nd_2 Fe_(14)b)-shell(Ce_2 Fe_(14)b) type. It is found that core((Nd,Ce)_2 Fe_(14)b)-shell(Nd_2 Fe_(14)b)type grain with 2 nm thick shell always presents the largest coercivity under the same total cerium content. Furthermore,the relationship between the coercivity and the shell thickness t in core((Nd,Ce)_2 Fe_(14)b)-shell(Nd_2 Fe_(14)b) type grain has been studied. When the total cerium content is kept at 20.51 at.%, the analyzed results show that as t varies from 1 nm to 7 nm, the coercivity gradually ascends at the beginning, then quickly descends after reaching the maximum value when t = 5 nm. From the perspective of the positions of nucleation points, the reasons why t affects the coercivity are discussed in detail.

A new linearly-combined bi-exponential model for kinetic analysis of the isometric relaxation process of bufo gastrocnemius under electric stimulation in vitro

There was a slow-relaxing tail of skeletal muscles in vitro upon the inhibition of Ca2+-pump by cyclopiazonic acid (CPA). Herein, a new linearly-combined bi-exponential model to resolve this slow-relaxing tail from the fast-relaxing phase was investigated for kinetic analysis of the isometric relaxation process of bufo gastrocnemius in vitro, in comparison to the single exponential model and the classical bi-exponential model. During repetitive stimulations at a 2-s interval by square pulses of a 2-ms duration at 12 V direct currency (DC), the isometric tension of bufo gastrocnemius was recorded at 100 Hz. The relaxation curve with tensions falling from 90% of the peak to the 15th datum before next stimulation was analyzed by three exponential models using a program in MATLAb 6.5. both the goodness of fit and the distribution of the residuals for the best fitting sup- ported the comparable validity of this new bi-exponential model for kinetic analysis of the relaxation process of the control muscles. After CPA treatment, however, this new bi-exponential model showed an obvious statistical superiority for kinetic analysis of the muscle relaxation process, and it gave the estimated rest tension consistent to that by experimentation, whereas both the classical bi-exponential model and the single exponential model gave biased rest tensions. Moreover, after the treatment of muscles by CPA, both the single exponential model and the classical bi-exponential model yielded lowered relaxation rates, nevertheless, this new bi-exponential model had relaxation rates of negligible changes except much higher rest tensions. These results suggest that this novel linearly-combined bi-exponential model is desirable for kinetic analysis of the relaxation process of muscles with altered Ca2+-pumping activity.

The oxalate co-precipitation method was used to synthesize the La3+substituted Ni-Cu-Zn(La-NCZ)nanoferrites having chemical composition Ni_(0.7)Cu_(0.1)Zn_(0.2)LaxFe_(2-x)O_(4)(x=0.015,0.025 and 0.035).DC resistivity study of nanoferrites shows both the conducting and semiconducting *** room temperature DC electrical resistivity of Ni-Cu-Zn(NCZ)nanoferrites decreases,whereas Curie temperature increases with increasing La^(3+)*** the temperature range of 30-170℃nanoferrites show p-type semiconducting behavior except x=0.015;thereafter,they show n-type *** frequency dispersive initial permeability(μi)associated with its real and imaginary(μ’andμ")parts are attributed to the domain wall movement and magnetic spin ***μi,u’andμ"of La-NCZ nanoferrites are higher than those of pure NCZ *** constant(ε’),dielectric loss(ε")and AC resistivity(ρAC)of La-NCZ nanoferrites show normal dielectric *** is found thatε’of NCZ nanoferrites decreases with the increasing content of La3+*** bandgap energy of La-NCZ nanoferrites is achieved in the range 1.36-1.70 eV confirming the semiconducting nature of materials.

The kagome metals AV_(3)Sb_(5)(A=K,Rb,Cs)under ambient pressure exhibit an unusual charge order,from which superconductivity *** this work,by applying hydrostatic pressure using a liquid pressure medium and carrying out electrical resistance measurements for RbV_(3)Sb_(5),we find that the charge order becomes suppressed under a modest pressure pc(1.4 GPabefore exhibiting another{maximum}around 22.8 GPa,signifying the presence of a second superconducting *** normal state resistance anomalies are found to be associated with the second superconducting dome,similar to KV_(3)Sb_(5).Our findings point to qualitatively similar temperature-pressure phase diagrams in KV_(3)Sb_(5) and RbV_(3)Sb_(5),{and suggest a close link}between the second superconducting dome and the high-pressure resistance anomalies.

background and Aims:Cirrhosis is the precursor lesion for most hepatocellular carcinoma(HCC)***,no biomarker effectively predicted HCC initiation before diagnosis by *** aimed to investigate the hallmarks of immune microenvironments in healthy,cirrhotic livers and HCC tumor tissues and to identify immune biomarkers of cirrhosis-HCC ***:Expression matrices of singlecell RNA sequencing studies were downloaded and integrated with Seurat package *** was performed to analyze the immune cell compositions of different sample ***:The cirrhotic liver and HCC tumors had distinct immune microenvironments,but the immune landscape of cirrhotic livers was not markedly modified compared with healthy *** subsets of b cells and three subsets of T cells were identified in the *** the T cells,naïve T cells were more prominent in the cirrhotic and healthy liver samples than in the HCC *** contrast,the neutrophil count was lower in cirrhotic *** macrophage clusters were identified,one that actively interacted with T cells and b cells and was enriched in cirrhotic blood compared with HCC blood ***:Decreased naïve T cell infiltration and increased neutrophil infiltration in the liver may indicate the development of HCC in cirrhotic *** in blood-resident immune cells may also be a sign of HCC development in cirrhotic *** dynamics of the immune cell subsets may serve as novel biomarkers to predict the transition from cirrhosis to HCC.

Compared with the traditional heteroatom doping,employing heterostructure is a new modulating approach for carbon-based ***,a facile ball milling-assisted route is proposed to synthesize porous carbon materials composed of abundant graphene/hexagonal boron nitride(G/h-bN)*** Ni powder and nanoscale h-bN sheets are used as a catalytic substrate/hard template and“nucleation seed”for the formation of the heterostructure,***-prepared G/h-bN heterostructures exhibit enhanced electrocatalytic activity toward H_(2)O_(2) generation with 86%-95%selectivity at the range of 0.45-0.75 V versus reversible hydrogen electrode(RHE)and a positive onset potential of 0.79 versus RHE(defined at a ring current density of 0.3 mA cm^(-2))in the alkaline *** a flow cell,G/h-bN heterostructured electrocatalyst has a H_(2)O_(2) production rate of up to 762 mmol g_(catalyst)^(-1) h^(-1) and Faradaic efficiency of over 75%during 12 h testing,superior to the reported carbon-based *** density functional theory simulation suggests that the b atoms at the interface of the G/h-bN heterostructure are the key active *** research provides a new route to activate carbon catalysts toward highly active and selective O_(2)-to-H_(2)O_(2) conversion.
