Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

The Ti17(a+β)-Ti17(β)dual alloy-dual property blisk produced using Linear Friction Welding(LFW)is considered as high-performance component in advanced ***,up to now,microstructure evolution and relationship between microstructure and micro mechanical properties of LFWed Ti17(a+β)/Ti17(β)dissimilar joint have not been thoroughly *** this work,complex analyses of the phase transformation mechanisms of the joint are conducted,and phase transformations in individual zones are correlated to their microhardness and *** reveal that a dissolution occurs under high temperatures encountered during LFW,which reduces microhardness of the joint to that of Ti17(a+β)and Ti17(β).In ThermoMechanically Affected Zone of Ti17(a+β)(TMAZ-(a+β))side joint,a large number of nanocrystalline a phases form with different *** microstructure strengthens significantly by fine grains which balances partial softening effect of a dissolution,and increases nanohardness of a phase and microhardness of TMAZ-(a+β).Superlattice metastableβphase precipitates from metastableβin Weld Zone(WZ)during quick cooling following welding,because of short-range diffusion migration of solute atoms,especiallyβstabilizing elements Mo and *** precipitation of the superlattice metastableβphase results in precipitation strengthening,which in turn increases nanohardness of metastableβand microhardness in WZ.

Panax ginseng(***.)has been traditionally employed in Korea and China to alleviate fatigue and digestive *** particular,Korean red ginseng(KRG),derived from streamed and dried ***,is known for its anti-aging and anti-inflammatory ***,its effects on benign prostatic hyperplasia(BPH),a representative aging-related disease,and the underlying mechanisms remain *** study aims to elucidate the therapeutic effects of KRG on BPH,with a particular focus on mitochondrial dynamics,including fission and fusion *** effects of KRG on cell proliferation,apoptosis,and mitochondrial dynamics and morphology were evaluated in a rat model of testosterone propionate(TP)-induced BPH and TP-treated LNCaP cells,with mdivi-1 as a *** results revealed that KRG treatment reduced the levels of androgen receptors(AR)and prostate-specific antigens in the BPH *** inhibited cell proliferation by downregulating cyclin D and proliferating cell nuclear antigen(PCNA)levels,and it promoted apoptosis by increasing the ratio of B-cell lymphoma protein 2(Bcl-2)-associated X protein(Bax)to Bcl-2 ***,KRG treatment enhanced the phosphorylation of dynamin-related protein 1(DRP-1,serine 637)compared with that in the BPH group,which inhibited mitochondrial fission and led to mitochondrial *** modulation of mitochondrial dynamics was associated with decreased cell proliferation and increased *** dysregulating AR signaling and inhibiting mitochondrial fission through enhanced DRP-1(ser637)phosphorylation,KRG effectively reduced cell proliferation and induced *** findings suggest that KRG’s regulation of mitochondrial dynamics offers a promising clinical approach for the treatment of BPH.

Dy^(3+) and Lu^(3+) co-doped CaLaGa_(3)O_(7) phosphors were prepared via high-temperature solid-phase *** electronic structures of LuxCaLa_(0.98-x)Ga_(3)O_(7) were investigated by the first-principles *** influence of Lu^(3+) on yellow light emission was studied using X-ray diffraction(XRD)and photoluminescence(PL) *** XRD results indicate that compared to Dy^(3+):CaLaGa_(3)O_(7),the cell parameters of Dy^(3+):LuCaLaGa_(3)O_(7) tend to decrease,which is due to the smaller radius of Lu^(3+).When pumped by blue GaN laser diode(LD),the emission peaks of phosphors with different Lu^(3+)doping concentrations in the visible region are similar,with the strongest peak at 574 nm in the yellow light region,which is sensitive to human *** optimal doping concentration of Lu^(3+) is confirmed to be1 at%,when all emission spectra and measured fluorescence lifetimes are taken into ***,the optimal phosphor composition Dy^(3+):Lu_(x)CaLa_(0.98-x)Ga_(3)O_(7)(x=0.01) has an internal quantum efficiency(IQE) of 46.94% and an external quantum efficiency(EQE) of 15.19%.Most notably,the prepared phosphor demonstrates excellent thermal stability and a high activation energy(0.203 eV).In addition,the International Illumination Commission color coordinates of the Dy^(3+):LuxCaLa_(0.98-x)Ga_(3)O_(7) phosphors are in the yellow light *** above analysis indicates that the Dy^(3+):LuxCaLa_(0.98-x)Ga_(3)O_(7)(x=0.001)phosphor has promising application prospects in yellow light-emitting devices.

BTG2 interference ameliorates high glucose-caused oxidative stress, cell apoptosis, and lipid deposition in HK-2 cells

Objectives:To evaluate the effectiveness and safety of Qishen Yiqi Dripping Pill(QSYQ)in patients with acute coronary syndrome(ACS)after percutaneous coronary intervention(PCI).Methods:This multicentre prospective cohort study was conducted at 40 centers in *** with ACS after PCI entered either the QSYQ or Western medicine(WM)groups naturally based on whether they had received QSYQ before *** group received QSYQ(0.52 g,3 times a day for 12 months)in addition to *** primary endpoint included cardiac death,non-fatal myocardial infarction,and urgent *** secondary endpoint included rehospitalization due to ACS,heart failure,stroke,and other thrombotic *** of life was assessed by the Seattle Angina Questionnaire(SAQ).Results:A total of 936 patients completed follow-up of the primary endpoint from February 2012 to December ***,487 patients received QSYQ and *** a median follow-up of 566 days(inter quartile range,IQR,517–602),the primary endpoint occurred in 46(9.45%)and 65(14.48%)patients in QSYQ and WM groups respectively[adjusted hazard ratio(HR)0.60,95%confidence interval(CI)0.41–0.90;P=0.013].The secondary endpoint occurred in 61(12.53%)and 74(16.48%)patients in QSYQ and WM groups,respectively(adjusted HR 0.76,95%CI 0.53–1.09;P=0.136).In sensitivity analysis,the results still demonstrated that WM combined with QSYQ reduced the risk of the primary endpoint(HR 0.67,95%CI 0.46–0.98;P=0.039).Moreover,QSYQ improved the disease perception domain of the SAQ(P<0.05).Conclusions:In patients with ACS after PCI,QSYQ combined with WM reduced the incidence of the primary *** findings provide a promising option for managing ACS after PCI and suggest the potential treatment for reducing the risk of primary endpoint included cardiac death,non-fatal myocardial infarction,and urgent revascularization through intermittent administration of QSYQ.(Registration *** CTR-OOC-14005552).

Extracellular membrane proteins are crucial for mediating cell attachment,recognition,and signal transduction in the testicular microenvironment,particularly germline stem *** 18(CDH18),a type Ⅱ classical cadherin,is primarily expressed in the nervous and reproductive ***,we investigated the expression of CDH18in neonatal porcine prospermatogonia(ProSGs)and murine spermatogonial stem cells(SSCs).Disruption of CDH18 expression did not adversely affect cell morphology,proliferation,self-renewal,or differentiation in cultured porcine ProSGs,but enhanced cell adhesion and prolonged cell *** analysis indicated that the down-regulation of CDH18 in ProSGs significantly up-regulated genes and signaling pathways associated with cell *** further elucidate the function of CDH18 in germ cells,Cdh18 knockout mice were generated,which exhibited normal testicular morphology,histology,*** analysis showed increased expression of genes associated with adhesion,consistent with the observations in porcine *** interaction of CDH18withβ-catenin and JAK2 in both porcine ProSGs and murine SSCs suggested an inhibitory effect on the canonical Wnt and JAK-STAT signaling pathways during CDH18 ***,these findings highlight the crucial role of CDH18 in regulating cell adhesion in porcine ProSGs and mouse *** this regulatory mechanism provides significant insights into the testicular niche.

In this editorial,we provide commentary on the study by Gong et *** this original research article,Gong et al employed a bioinformatics approach to investigate the involvement of autophagy in active ulcerative colitis(UC).Through differential gene expression analysis,they identified 58 differentially expressed autophagy-related genes in UC patients compared to healthy ***,HSPA5,CASP1,SERPINA1,CX3CL1,and BAG3,were found to be upregulated in active UC patients,suggesting their significance as core autophagyrelated *** analysis unveiled associations with crucial signaling pathways and diseases such as middle cerebral artery occlusion and ***,immune cell infiltration analysis revealed notable differences in immune cell composition between UC patients and healthy *** findings offer valuable insights into the role of autophagy in UC pathogenesis and potential therapeutic targets.
