The CLE40 and CRN/CLV2 Signaling Pathways Antagonistically Control Root Meristem Growth in Arabidopsis

Differentiation processes in the primary root meristem are controlled by several signaling pathways that are regulated by phytohormones or by secreted peptides. Long-term maintenance of an active root meristem requires that the generation of new stem cells and the loss of these from the meristem due to differentiation are precisely coordinated. Via phenotypic and large-scale transcriptome analyses of mutants, we show that the signaling peptide CLE40 and the recep- tor proteins CLV2 and CRN act in two genetically separable pathways that antagonistically regulate cell differentiation in the proximal root meristem. CLE40 inhibits cell differentiation throughout the primary root meristem by controlling genes with roles in abscisic acid, auxin, and cytokinin signaling. CRN and CLV2 jointly control target genes that promote cell differentiation specifically in the transition zone of the proximal root meristem. While CRN and CLV2 are not acting in the CLE40 signaling pathway under normal growth conditions, both proteins are required when the levels of CLE40 or related CLE peptides increase. We show here that two antagonistically acting pathways controlling root meristem differentiation can be activated by the same peptide in a dosage-dependent manner.

Increased QT dispersion in the surface ECG (QTd = QTmax minus QTmin) is considered as an indicator of electrical inhomogeneitv and a useful predictor for severe ventricular arrhythmia and sudden cardiac death in patients with different heart diseases. Patients with ischemic and idiopathic cardiomyopathy have a very high incidence of severe ventricular arrhythmia and sudden cardiac death. We compared QT, QTc. JT and JTc dispersion in ischemic (ICMP) and idopathic (CCMP) cardiomyopathy patients with and without severe ventricular arrhythmia and normal controls.

Osseous reconstruction of large bone defects remains a challenge in oral and maxillofacial *** addition to autogenous bone grafts,which despite potential donor-site mobility still represent the gold standard in reconstructive surgery,many studies have investigated less invasive alternatives such as in vitro cultivation *** study compared different types of seeding techniques on pureβ-tricalcium phosphate scaffolds in terms of bone formation and ceramic resorption in *** scaffolds loaded with autologous cancellous bone,venous blood,bone marrow aspirate concentrate or extracorporeal in vitro cultivated bone marrow stromal cells were cultured in sheep on a perforator vessel of the musculus latissimus dorsi over a 6-month *** and histomorphometric analyses revealed that scaffolds loaded with cancellous bone were superior at promoting heterotopic bone formation and ceramic degradation,with autogenous bone and bone marrow aspirate concentrate inducing in vivo formation of vital bone *** results confirm that autologous bone constitutes the preferred source of osteoinductive and osteogenic material that can reliably induce heterotopic bone formation in vivo.
