Screening of Protease Inhibitory Activity in Aqueous Extracts of Marine Invertebrates from Cuban Coast

Protease inhibitors have been isolated from many variable sources;however, the need to identify and characterize new molecules has increased with the discovery of new therapeutic targets and the lack of specificity of already identified compounds with inhibitory activity. The goal of this work was to search for inhibitory activity against four proteolytic enzymes already recognized as therapeutic targets: human neutrophil elastase, dipeptidyl peptidase IV, subtilisin from Bacillus licheniformis and cathepsin K in selected marine invertebrates from the Caribbean Sea. A systematic screening was carried out with selected aqueous extracts belonging to 20 species from seven different phyla: Annelida, Bryozoa, Chordata, Cnidaria, Equinodermata, Mollusca and Porifera, all collected at the coast of Havana (Cuba). All extracts showing initial inhibitory activity were characterized in terms of IC50 values and specific inhibitory activity (SIA). Model enzymes were used in the case of human neutrophil elastase (porcine pancreatic elastase) and cathepsin K (papain) for the screening and all positive results were confirmed by testing toward the therapeutic targets. Ten extracts were identified showing inhibitory activity against human neutrophil elastase, for which the most promising values were obtained for Nerita peloronta. Only one extract, Bunodosoma granulifera, showed inhibitory activity against dipeptidyl peptidase IV with rather poor values of IC50 and SIA. Seven extracts showed inhibitory activity against B. licheniformis subtilisin with very good IC50 and SIA values for Lissodendorix isodyctialis, Cenchritis muricatus, and N. peloronta. Which are the most favourable conditions for reducing soil CO_(2)emissions with no-tillage?Results from a meta-analysis

No-tillage practices have a recognised beneficial impact on soil and water conservation while reducing erosion processes and enhancing soil organic matter ***,scientists continue to debate the effectiveness of no-tillage in reducing soil carbon dioxide(CO_(2))emissions from *** the same line of inquiry pursued by the authors who reviewed the impact of conservative practices on direct CO_(2)emissions,we applied meta-analytic and machine learning techniques to unravel the effect of no-tillage under contrasting pedo-environmental conditions and agricultural *** analysed fifty-six experimental studies investigating direct soil CO_(2)emissions from no-tillage and conventional tillage practices(102 paired observations),considering pedological(soil texture,soil organic carbon content),environmental(climate type),and management(crop rotation,experiment duration)*** estimated the effect of different practices on the daily amount of soil CO_(2)emissions,and the impact of tillage in the period immediately following the *** main insights of this study are:(i)the conditions leading to the highest reduction of CO_(2)emissions due to no-tillage were long-term experiments(standardised mean differenceβ=0.64)conducted in arid environments(β=0.76)and clay soils(β=0.81),with low organic carbon content(β=0.79)where crop rotations(β=0.65)were performed;(ii)the same conditions were associated with the lowest absolute CO_(2)emissions,irrespective of soil management;(iii)the highest contribution to the variability of absolute soil CO_(2)emissions was associated with soil texture(mean decrease in accuracy of Random Forest models,MDA=4.57),rotation(MDA=3.07),experiment duration(MDA=2.93)and soil organic carbon content(MDA=2.24),rather than to tillage practices;(iv)soil CO_(2)emissions almost doubled in the first day after a tillage event,consistently across studies(p=0.001).This meta-analysis offers quantitative figures on the impact of tillage

BACKGROUND Veno-arterial extra corporeal membrane oxygenation(VA-ECMO)support is commonly complicated with left ventricle(LV)distension in patients with cardiogenic *** resolved this problem by transeptally converting VAECMO to left atrium veno-arterial(LAVA)-ECMO that functioned as a temporary paracorporeal left ventricular assist device to resolve LV *** our case LAVA-ECMO was also functioning as a bridge-to-transplant device,a technique that has been scarcely reported in the *** SUMMARY A 65 year-old man suffered from acute myocardial injury that required percutaneous *** than two weeks later,noncompliance to antiplatelet therapy led to stent thrombosis,cardiogenic shock,and cardiac *** VA-ECMO support was started,and the patient underwent a second coronary angiography with re-stenting and intra-aortic balloon pump *** VA-ECMO support was complicated by left ventricular distension which we resolved via ***,episodes of bleeding and sepsis complicated the clinical picture and the patient passed away 27 d after initiating *** This clinical case demonstrates that LAVA-ECMO is a viable strategy to unload the LV without another invasive percutaneous or surgical *** also demonstrate that LAVA-ECMO can also be weaned to a left ventricular assist device system.A benefit of this technique is that the procedure is potentially reversible,should the patient require VA-ECMO support again.A transeptal LV venting approach like LAVA-ECMO may be indicated over ImpellaTM in cases where less LV unloading is required and where a restrictive myocardium could cause LV *** ventricular over-distention is a well-known complication of peripheral VA-ECMO in cardiogenic shock and LAVA ECMO through transeptal cannulation offers a novel and safe approach for treating LV overloading,without the need of an additional percutaneous access.

Bio-mathematical models of viral dynamics to tailor antiviral therapy in chronic viral hepatitis

The simulation of the dynamics of viral infections by mathematical equations has been applied successfully to the study of viral infections during antiviral therapy. Standard models applied to viral hepatitis describe the viral load decline in the f irst 2-4 wk of antiviral therapy, but do not adequately simulate the dynamics of viral infection for the following period. The hypothesis of a constant clearance rate of the infected cells provides an unrealistic estimation of the time necessary to reach the control or the clearance of hepatitis B virus (HBV)/ hepatitis C virus (HCV) infection. To overcome the problem, we have developed a new multiphasic model in which the immune system activity is modulated by a negative feedback caused by the infected cells reduction, and alanine aminotransferase kinetics serve as a surrogate marker of infected-cell clearance. By this approach, we can compute the dynamics of infected cells during the whole treatment course, and find a good correlation between the number of infected cells at the end of therapy and the long-term virological response in patients with chronic hepatitis C. The new model successfully describes the HBV infection dynamics far beyond the third month of antiviral therapy under the assumption that the sum of infected and non-infected cells remains roughly constant during therapy, and both target and infected cells concur in the hepatocyte turnover. In clinical practice, these new models will allow the development of simulators of treatment response that will be used as an "automatic pilot" for tailoring antiviral therapy in chronic hepatitis B as well as chronic hepatitis C patients.
